RAPD analysis of grapevine hybrids and cultivars
63-66.Views:180Utilization of the Randomly Amplified Polymorphic DNA (RAPD) technique as a molecular marker was tested to investigate the relationships between some representative grapevine cultivars and hybrids established at the Department of Genetics and Plant Breeding (CUB), to distinguish clones as well as to characterize various hybrids between species or cultivars and their parents. Vitis vinifera cultivars were easily and successfully distinguished by the RAPD technique and they were grouped according to the traditional taxonomic classification. RAPD patterns of the examined Pinot gris clones proved to be completely identical. Number of generations was reflected by the value of genetic distance of the examined hybrids. Genetic identity of parents and their offsprings was influenced by the selection applied in the process of plant breeding. Parental phenotypic and morphologic characteristics showed high degree of segregation in hybrids, but RAPD analysis revealed that their genetic similarity is considerable. The three Vitis anntrensis clones were properly discriminated from every cultivar and hybrid of Vitis vinifera, i.e. hybrids are much closer to the cultivated grapevine than to V. anzurensis due to the phenotypic selection carried out during the life-cycle of one or two generations.

In vitro multiplication and hardening of grapevine plants in aeriated media
15-18.Views:218In vitro cultures have widely been used in horticulture for rapid multiplication of new varieties and clones as well as to produce pathogen-free stock material. To improve efficient hardening and transfer in vitro grown grapevine plants were multiplied by cutting them into single-node internodes with the whole leaf. Microcuttings including the shoot tips were rooted in granulated perlite moisted with tapwater under sterile conditions. After 2-3 weeks the rooted microcuttings were supplied by nutrients and hardened by gradual opening and finally by complete removal of the lids of jars or plastic boxes used for growth. Using this method microcuttings of Vitis vinifera cvs. „Chardonnay", „Cabernet franc", „Riesling" and „Sauvignon blanc" and the rootstock varieties Vitis riparia x Vitis cinerea cv. „Barrier" and Vitis berlandieri x Vitis rupestris cv. „Richter 110" formed new roots and shoots and 100% of the tested plants survived the acclimatization procedure. Similar results were obtained when perlite was replaced with rockwool-, or pit-pot blocks. This method may highly increase the efficiency of producing pathogen-free propagating material and new transgenic lines.

Old Hungarian grapevine cultivars and their relations characterized with microsatellite markers
27-31.Views:234Thirty-one old Hungarian grapevine (Vitis vinifera L.) cultivars were investigated on 7 microsatellite loci to characterize them, to separate the cultivars from synonym names, and to confirm parent-offspring connections. Conculta (group of cultivars or bud sports) members, such as `Goher' and Tajor' representatives, were studied to find a suitable locus for the separation. Synonyms, conculta members, subcultivars and clones of Turmine, which was the most important cultivar of Tokaj, were also analyzed to separate the members of the different taxonomic levels. Pedigree of 'Kiralyleanyka' was examined to find the missing ancestor, because the parent-offspring connection between the natural hybrid and `Koverszolo' is questionable.

Biological performance of grape varieties in Eastern Hungary
31-35.Views:232Eurasian and interspecific grape varieties and candidates were inspected for biological productivity in the variety collection of the University of Debrecen at the Horticultural Experimental Station in Pallag. In this paper, data are reported on yields (kg/stock), cane production (kg/ stock) and use-up index of wood yield of each cultivar in the experimental years 2011-2012. From the range of Eurasian (Vitis vinifera L.) white wine-grape qualified cultivars and candidates ’Ezerfürtû’, ’Generosa’, ’Jubileum 75’, ’Müller Thurgau’, ’Zöld veltelíni’,’B-11’, ’CSFT-92’, ’Pintes’, from the range of Eurasian (Vitis vinifera L.) red wine-grape qualified cultivars and candidates ’Kármin’, ’Pinot noir’, ’Alicante Bouschet’ and ’Rubintos’ were highlighted. Promising data of interspecific ’Aletta’, ’Csillám’, ’Kunleány’, ’Orpheus’, ’Refrén’, ’Taurus’, ’Viktória gyöngye’, ’Alföld 100’, ’Reform’, ’RF38/32’, ’Toldi’ white, and ’Dunagyöngye’ ’Pannon frankos’ red wine-grape cultivars and candidates were emphasized. Published data refer only to the biological performance of the cultivars. The thorough evaluation of varieties needs respect to data on resistance to fungal disease and climatic extremities, and other characteristics concerning production technology and oenological parameters.

A model of full bloom starting date of some white Vitis vinifera L. varieties grown in Helvecia
21-25.Views:182Grapevine bloom happens between end of May and the middle of June in Hungary. However, climate change in the past decades and the occurring weather anomalies can modi fy this date to a diverse degree. Among the weather factors, the bloom starting dates of grapevine depend mostly on temperature and relative humidity of air. There can be significant differences between North American and East Asian grapevine varieties, and of course, the early and late ripening varieties. ln this approach we investigated the starting dates of bloom between 2000 and 2004 for grapevine varieties grown in Helvecia, as well as the effectiveness of a temperature sum model. The model is based on the widely accepted cumulated heat sum concept, and the optimization was made for the least standard deviation in days as well as on the least average absolute deviation in days and on the least maximum deviation in days. The model is connected directly to a similar model for the budburst date of the same plantings (Hlaszny & ladanyi, 2009). We set the optimum lower base temperature to I 0.45 °C and the upper base temperature to 26 °C. The absolute values of the differences between the observations and the model estimations move between one and six days with an average of 1.81 days

Historical background and constraints of a grapevine germplasm foundation in Hajdú-Bihar county, Eastern Hungary
11-17.Views:278The historical background of Debrecen linked to viticulture and wine-making stands mainly on the lack of drinkable water, the necessity of drinkable liquid during wartime and epidemics. The special character of the city evolved together with the changing lives of citizens and the increasing trade importance of the city. Period of Turkish occupation gave impetus to the formation of the 11 vine gardens of the settlement. After the devastation of rootmite and peronospora ‘Kadarica’ and ‘Nagy burgundy’ (‘Blaufrankish’), in smaller proportion - on lower sites – ‘Cabernet’ were planted. As white varieties ‘Ezerjó’, ‘Olasz Rizling’, ‘Kövidinka’, ‘white Mustos’, in smaller proportion ‘Szlankamenka’, ‘Erdei’, ‘Szilvaner’, ‘Mézesfehér’, ‘Bakar’, ‘Veltelini’ (red), ‘Fehér burgundi’ (? white burdunder), ‘Rajnai rizling’, ‘Red Tramini’, ‘Furmint’, ‘Muscat Lunel’, ‘Járdovány’ and ‘Juh-fark’ were planted. After the Trianon treaty in 1920, 2/3rd of Hungary was cut away. Érmellék wine region was also cut in two, thus Debrecen broke away from its wine region. Legal regulations after the World War II. (1959) referred back to variety application advised in 1924 for “place suitable for good wine production, not included in any wine region”, like Debrecen listing ’Ezerjó’, ’Mézesfehér’, ’Olaszrizling’, ’Bánáti rizling’, ’Furmint’, ’Hárslevelű’, ’Kövidinka’, Kecskemét virága’, ’Piros szlankamenka’,’Pozsonyi fehér’; ’Kadarka’, ’Oportó’ and ’Kékfrankos’ (Blaufrankish). The political changes of 1990 and Hungary’s admission to the Eurepoean Union almost annihilated the wine production of Debrecen. However little gardens conserved historic varieties which could date back even to many centuries. Through a local magazine a collecting work was announced pointing to gather ancient local (Vitis vinifera conv. pontica) varieties forming a genebank, established on the experimental station of the University of Debrecen. In 2014, about 112 items were collected (accessions). As a 2nd round of the work, with a more detailed and precise work, further 81 items were put into the reservatum. The latter represent single stuck collection, whereas the first ones are to be studied az mixed items. Most notable accession names (ACENAME) of the work are: ‘Fehér gohér’, ‘Veres gohér’, ‘Fekete gohér’, ‘Kék gohér’, ‘Erdei’, ‘Ezerjó’, ‘Kűbeli’, ‘Rizling’, ‘Mézes fehér’, ‘Dinka’, ‘Madling’, ‘Bakator’ and ‘Kadarka’. Simulteneously with the strenghening and morphological description of conserved stucks genetic identification of the items is being elaborated. Database comprising FAO/IPGRI multi-crop passport descriptors and OIV Primary descriptor priority list are to be published on-line in between the development of the platform.

Phytoplasma diseases of grapevine and the possible measures to control them
37-43.Views:326Phytoplasmas are a special group of phloem-living pathogens in several plant species. Grapevine yellows (GY) is a term for phytoplasma diseases occurring on Vitis vinifera and inducing the same or very similar symptoms and causing severe losses worldwide. Flavescence Dorée (16SrV) phytoplasma (FD, species name: ‘Candidatus Phytoplasma vitis’) is considered a quarantine pest in several countries due to its epidemic character and high economic loss it provokes. The leafhopper Scaphoideus titanus is the univoltine and monophagous vector of FD. Bois noir disease caused by stolbur (16SrXII-A) phytoplasma (species name: ‘Candidatus Phytoplasma solani’) is described under different disease names in different countries. Hyalesthes obsoletus (Cixiidae) is the only proved polyphagous vector of BN. However, distribution of BN disease is increasing also on those areas where H. obsoletus is not prevalent or only in a very low number. Therefore the presence of other vectors cannot be concluded. The ‘Tuf-a’ type Stolbur phytoplasma is associated with stinging nettle (Urtica dioica) and the tuf-b type one to field bindweed (Convolvulus arvensis). There are only preventive control measures against phytoplasmas: the use of pathogen-free propagating material, hot water treatment of propagating material, as well as control of vectors and weeds. S. titanus can be efficiently controlled by insecticide treatments. However, in case of H. obsoletus, insecticides are not effective due to the biological characters and feeding habits of the vector.Weed control can reduce H. obsoletus specimen and their abundance to a certain extent. Extensive research is needed on wild hosts of GY phytoplasmas especially on BN phytoplasma and its vectors to the better understanding of their epidemiology.

Bacterial diseases of grapevine
45-49.Views:420Grapevines are affected by three major bacterial diseases worldwide, such as bacterial blight (Xylophilus ampelinus), Pierce’s disease (Xylella fastidiosa) and crown gall (Agrobacterium vitis). These bacteria grow in the vascular system of their host, thus they invade and colonize the whole plant, independently on symptom development. Latently infected propagating material is a major factor in their spreading. Therefore the use of bacteria-free planting stock has a basic importance in viticulture. Today several innovative diagnostic methods, mostly based on polymerase chain reaction, are available to detect and identify bacterial pathogens of grapevines. For production of bacteria-free plants, the use hot water treatment followed by establishment of in vitro shoot tip cultures is proposed.

Grape rootstock - scion interactions on shoot growth and cane maturing
30-34.Views:214Interaction between the rootstock and scion varieties (clones) was examined. Berlandieri x Riparia T. K. 5BB, Berlandieri x Riparia T.5C being the most widely used varieties in Hungary, Fercal, Ruggeri 140 being recently used on special soil conditions ; Georgikon 28 a new established variety and Berlandieri x Riparia T. 8B GK 10 clone were the rootstocks used. The V. vinifera L. varieties were a clone of Italian Riesling and two new hybrids from the same cross population f ( Noble Italian Riesling X Ezerjó) X ( Noble Italian Riesling X Pinot Gris ) }. This study presents the results how the growth habit depends on different scion/stock combinations in the first three years.
Shoot growth characters were observed during the initial years after plantation in the vineyard until we got the first yields. Our results did not agree with those of Pospisilova (1977) that the anticipated vigour of shoot growth will appear later in the vineyard only. However we agree with Zimmerman (1970), who showed that the differences are caused by the rootstocks in the growth habit of vines during the first two years already. The differences in shoot elongation decreased similarly to the rootstocks in the first year of fruit production. It shows us that the intense root development caused vigorous shoot growth, which we think to be a rootstock effect. When the vegetative and generative development are balanced already, the effect of scions in shoot elongation became stronger than the effect of rootstock.
We also found great differences in the maturation of wood each year from planting of the vineyard to the year of fruit production.
During the first years the differences of cane maturation were greater according to the scion varieties, the means were significantly different. In the first ripening year the differences dwindled or increased due to the rootstocks. This means that the effect of the rootstock on cane maturation became stronger in the first year of fruit production.

Evaluation of different sources of resistance for breeding powdery mildew resistant grapevine varieties
35-40.Views:188The appearance of resistance to powdery mildew was investigated on seedling progenies of interspecific crossings. Hybrids of American and East-Asian wild species and one Vitis vinifera variety (Janjal kara) were used as source of resistance in the crossings. The resistance of foliage and of berries were tested in the field at the time of vintage. The degree of resistance was ranked into six classes.
In American and Asian hybrid derivatives there were few highly resistant progenies, but in the hybrids of Janjal kara variety more than 90 % of the progenies proved to be highly resistant to powdery mildew. According to the distribution of classes of resistance in the populations the resistance of American and Asian sources are mostly influenced by additive gene affects. The resistance of Janjal kara seemed to be mono or digenic.
No close correlation was found between the powdery mildew resistance of the foliage and that of the berries.

The development and reproduction of grape phylloxera (Daktulosphaira vitifoliae Fitch) strains of Hungarian vine-districts in root-bioassays
67-71.Views:142The root bioassay method allows for 4-6 week continuous observation of grape phylloxera feeding on the grape root. 10 Hungarian phylloxera strains were compared on susceptible Vitis vinifera cv. Chardonnay and the resistant rootstocks of V. berlandieri x V. riparia Teleki 5C and V. berlandieri x V. rupestris Georgikon 121 in in vitro observations. The strains originated from Villany and Eger (Figure 3) had higher reproduction on the root of V. berlandieri x V. riparia Teleki 5C (201 and 119 eggs) and were more aggressive than the others (average production 10 eggs). The continuous high level of survival, development and reproduction of the Eger strain on Teleki 5C (V. berlandieri x V. ripuria) until day 46 may be due to adaptation.

Research Note virus transmission by grape phylloxera (Daktulosphaira vittfoliae Fitch)
73-74.Views:133Grape phylloxera (Daktulosphaira vitifoliae FITCH) infests immature roots of both Vitis vinifera L. and phylloxera resistant rootstocks. A capability to transmit viticultural viruses would make grape phylloxera a phytosanitary threat even under conditions where direct damage by the insect is not likely. We tested the hypothesis that phylloxera could transmit grapevine fanleaf virus (GFLV) by planting infected and non-infected vines in common 10 liter pots and infecting roots of the infected vine with grape phylloxera. In this test infection of a previously non-infected plant in the absence of nematode population suggests that grape phylloxera is a vector of GFLV.

Virus and virus-like diseases of grapevine in Hungary
25-36.Views:283Viruses and viroids are submicroscopic infectious particles which can cause disease symptoms on grapevine. These parasites are depending completely on the energy metabolism of the plant cell. To enter the host cell plant viruses depend on injuries or on transmission via invertebrates (insects, nematodes, etc.). Viruses are classified by many characters including particle morphology, host range and information content of the genome. At present about 70 viruses including 7 viroids infecting grapevine are known. In single or mixed infections they are potentially detrimental to the quality and quantity of grape production in any growing area of the world. Some viruses can cause severe economic damage in vineyards. In Hungary many important viruses and viroids have been detected in grape. This review summarises characteristics of viruses and the results of detection and characterization of virus and virus like diseases of grapevine in Hungary. The identification of the causal agent, its transmission, geographical distribution and the development of the diagnostic methods are also discussed.

The importance of clonal selection of grapevine and the role of selected clones in production of healthy propagating stocks
15-24.Views:356Genetical alterations and phytosanitary status promote the variability and modify the appearance of vine. Old vine varieties in old vineyards are highly variable and well adapted to selection. Clonal selektion is based on a visual performance: valuable individuals (clones) are picked out according to visible symptoms or characters. The genetical stability of clones is proved by testing the vegetatively propagated progenies on the basis of morphological and molekular (SSR, AFLP, SMPL, RAPD) markes. Authors take great care of the visual phytosanitary selection as part of the clonal selection being the oreliminary step to develop pathogen-free propagation stocks. In Serbia (Vojvodina) the selection breeding has been carried on for several decades resulted in comparative clone trials with home and imported clones of Welsch Riesling, Chardonnay, Pinot gris, White Riesling. Among the clones of home selection SK.54 Welsch Riesling clone is the most valuable. Its clearing from pathogene is being carried on in an interregional IPA programme (HUSRB/0901/214/123) in Kecskemét. In Kecskemét, the centre of the Hungarian Danube vine region 5 vine clones have been registered (Cegléd szépe K.73, Irsai Olivér K.11, Kövidinka K.8, Hárslevelû K.9, Pannónia kincse K.56). Besides them 18 virus-tested clones have also been qualified.Works aiming at their complete exemption are going on in order to obtain clones free of propagation wood-borne diseases.

Primers designed for the detection of grapevine pathogens spreading with propagating material by quantitative real-time PCR
21-30.Views:291Several grapevine pathogens are disseminated by propagating material as systemic, but latent infections. Their detection and identification have a basic importance in the production and handling of propagating stocks. Thus several sensitive and reliable diagnostic protocols mostly based on molecular techniques have been developed. Of these methods quantitative real-time PCR (q-PCR) has recently got an emerging importance. Here we collected primer data for the detection and identification of grapevine pathogens which are important in the production of propagating stocks by q-PCR. Additional novel techniques that use DNA amplification, hybridization and sequencing are also briefly reviewed.

Conventional PCR primers for the detection of grapevine pathogens disseminated by propagating material
69-80.Views:304Polymerase chain reaction driven by sequence specific primers has become the most widely used diagnostic method to detect and identify plant pathogens. The sensitive and cost-effective pathogen detection is exceptionally important in the production of propagating material. In this paper we have collected primer sequence data from the literature for the detection of the most important grapevine pathogens disseminated by propagating stocks by conventional polymerase chain reaction. Basic protocols to obtain template nucleic acids have also been briefly rewieved.

Certification programme for production of virus-free propagating material of grapevine and its results in Hungary
39-43.Views:172In Hungary, detection of virus and virus-like diseases of grapevine began in 1960's at the Research Institute for Viticulture and Enology by János Lehoczky and his colleagues. At present, sixteen virus and virus-like diseases of Vitis vinifera are known to occur in Hungary.
Regular virological screening of grapevine varieties started in 1972. The present system of screening (visual selection, indexing, ELISA) has been established using methods with continuous improvement according to recommendations of international organizations.
In the first year symptomless grapevine plants are selected and marked during surveys carried out twice in the vegetation period: at about flowering and in the second half of September. At the first selection time plants are sampled for ELISA.
In the spring of the second year, overwintered canes are checked by woody indexing on 8 indicator species in the field.
In the third and fourth years the nursery is evaluated twice again. At the end, the marked grapevine plants, giving negative results on all indicators in every case, are considered virus-free.
In autumn of the fourth year, the virus-free material is planted out under screenhouse and also in a special mother block (nuclear stock) for maintenance and propagation.
Mother blocks of virus-free scion varieties have been established on 2 ha and those of rootstock varieties on 0.5 ha planted with the following number of varieties included in the national list: 71 European scion — and 12 rootstock varieties or variety candidates/clones. It is necessary to increase the area of Pre-base, Base and Certified stocks exclusively with tested virus-free (clean) material.

Recent developments in biochemical characterization of Vitis vinifera L. varieties in Hungary
57-61.Views:179Isoelectric focusing is an effective and well reproducible method to provide information for identification of various plant species and clones if breeding or other genetic modification(s) for a given species are reflected in changes of an isozyme pattern. The method has been used for characterization of plant proteins and enzymes and for identification of various species and varieties. Our aim was to continue our several-year-work carried out on a wide variety of grapevine varieties and to reveal whether analyses of esterase and peroxidase isozyme patterns are suitable to distinguish various grapevine varieties. Therefore we compared esterase and peroxidase isozyme patterns of various species. Plant samples were obtained from Szigetcsep, Kecskemet, Tokaj and Eger. The following samples were analyzed: Pinot gris, noir, blanc, Chardonnay, Riesling Theses, Chasselas from Szigetcs6p, Bianca and his parents Eger2 and Bouvier from Kecskemet, Furmint and Hárslevelű from Tokaj, Kékfrankos and Zweigelt from Eger. To identify various species according to their esterase isozyme patterns the after blooming phenological phase while according to their peroxidase isozyme pattern the dormant phenological phase was found as optimal sampling time.

Two new registered table grape varieties
51-53.Views:145Two table grape varieties — Kósa and Narancsízű — were registered of the hybrids bred in the Research Institute for Viticulture and Enology in I lungary. Both varieties ripe early and are much in demand for the attractive clusters and flavours. As they are hybrids of Vitis vinifera they require similar growing conditions. In the same time with their qualification the two varieties are included in the variety list and allowed to be propagated.

Examinations of 600-year-old seeds by means of archaeobotanical and genetical methods
79-80.Views:162About 600-year-old plant seeds were discovered in a well of a mediaeval cellar in the course of an excavation in Budapest. After the archaeobotanic purification seed of 16 species were found in large quantities. Seeds preserved in the best state were selected from each group. The existence of endosperm was analysed in these subfossils, which turned to be successful mainly in the case of grapes (Vitis vinifera) and cornels (Cornus mas). Seeds of these two species contained the most endosperm and remains of the embryo. DNA was extracted with the help of DNEasy Plant Mini Kit and analysed by RAPD-PCR method. The amplification of DNA extracted from cornel seeds resulted in detecting a 1500 by fragment, which makes the comparison of these samples with present-day cornels possible.
