  • Evaluation of lifespan values of six tree taxa in city streets

    The aim of this study was to evaluate lifespan values of trees in tree-row system in the streets of Debrecen in two years (2009 and 2017). Six selected taxa (Pyrus calleryana, Acer tataricum, Sorbus intermedia, Magnolia kobus, Acer platanoides and Crataegus x lavalleei) were estimated for the following lifespan parameters: i) trunk diameter (cm), ii) tree crown size (m), iii) trunk status (in 0-5 grades), iv) tree crown status (in 0-5 grades), and v) estimated tree viability (in 0-5 grades). Our results showed that the largest were achieved for Pyrus calleryana and the lowest for Acer tataricum. The largest tree crown diameters were achieved for Acer platanoides and the lowest Magnolia kobus. The best trunk statuses by 2017 were achieved for Pyrus calleryana and the worst Crataegus x lavalleei. The best tree crown statuses by 2017 were achieved for Pyrus calleryana and the worst for Acer tataricum. The best estimated tree viability status was achieved for two taxa (Pyrus calleryana and Acer platanoides). Overall tree values were 2.73 times higher in 2017 compared to 2009. In summary, this study demonstrated the role of appropriate choice of tree taxa for a tree-row system under city street conditions.

  • Influence of irrigation and row spacing on the growth, seed yield and active substance of milk thistle (Silybum marianum)

    Milk thistle (Silybum marianum (L.) Gaertn) has been used in medicine since ancient times, but it has been cultivated as a medicinal plant only in recent decades. The ripe fruit of milk thistle contains flavonoids, which are used to prepare anti-hepatotoxic drugs. The main purpose of this study was to substantiate the effects of irrigation and row spacing on growth, seed yield and the content of active substances (silybin and silymarin) in milk thistle. The results showed that the suitable amount of irrigation was 20 mm and the appropriate row spacing was 25 cm.

     

  • Development of in vitro propagation system for Atriplex halimus L.

    Explants excised from adult shrubs were surface sterilized and cultured on Murashige and Skoog (MS) basal medium in the presence of plant growth regulators (PGRs) at different concentrations. A high multiplication rate of 7.2-fold was achieved every four weeks on MS medium supplemented with 4.44 μM BA, 0.49 μM IBA and 0.58 μM GA3. Rooting was achieved with 73% efficiency within 2-4 weeks on agar-gelled MS basal medium free of PGRs. Rooted plantlets were gradually acclimatized to field conditions over 5-6 weeks with 65% efficiency. For in vitro selection for salt tolerance, MS medium was supplemented with increasing concentrations of NaCl ranging between 25 and 1000 mM. This study has demonstrated that in vitro shoots could tolerate up to 600 mM NaCl with optimal growth at 200 mM, while higher concentrations of NaCl affected growth negatively. Growth and shoot number decreased with increasing NaCl concentration with all plantlets died at 1000 mM NaCl.

    Shallot is a vegetable and condiment crop widely used in Ethiopia and globally. However, absence of improved and adaptable varieties has been the major cause of low productivity. Narrow genetic base of local shallot germplasm owing to vegetative reproduction of the crop, among others, has been the root cause of low productivity. Nevertheless, some plants within the germplasm were observed bolting and producing viable seeds, presenting an opportunity for genetic diversification. Consequently, a germplasm enhancement program was initiated using these naturally outcrossing genotypes where about eighty-one genotypes were generated. The present study was thus undertaken with the objective of characterizing, classifying, and selecting the eighty-one genotypes for future breeding activities. The genotypes were planted in 9x9 simple lattice design with two replications at Debre Zeit Agricultural Research Center (Ethiopia) during the dry (irrigated) season of 2021. The genotypes were evaluated for fifteen growth, yield, and quality traits. Significant variations were observed among the genotypes in terms of bulb yield, bulb height and diameter, total soluble solids, bolting percentage, and bulb skin color. Bulb yield of the genotypes ranged from 31.33 t/ha in DZSHT-79-1A to 9.63 t//ha in DZSHT-45-1A-1. DZSHT-51-2 (207.93 g) was the highest yielder per plant whereas DZSHT-065-6/90 (74.51 g) was the lowest yielder. DZSHT-14-2-1/90 had the thickest bulb (44.69 mm) significantly thicker than twenty two genotypes which had bulb diameter ranging from 28.92 mm to 20.29 mm. DZSHT-81-1/90 was a genotype with the longest bulb height (52.33 mm) while DZSHT-147-1C was a genotype with the shortest bulb (33.12 mm). DZSHT-307-1/90 had the highest TSS (16.78°Brix) significantly differing from DZSHT-002/07 which had the lowest TSS (11.17 °Brix). Dry matter of the genotypes ranged from 12.00% to 22.79%. DZSHT-004/07, DZSHT-111-2-1, DZSHT-41-2B and DZSHT-72-2 had DM% greater than 20% which coupled with greater than 14 °Brix could make them suitable for dehydrated shallots. Among the 81 genotypes characterized 4 (4.9%), 7 (8.6%), 13 (16.1%), 28 (34.6%) and 29 (35.8%) were yellow, golden, light red, red and dark red in colour, respectively.  Fifteen of the genotypes had at least 50% bolting plants whereas twenty nine of the genotypes had less than 25% bolting. The results revealed that seven principal components explained approximately 76% of the observed variation. Cluster analysis grouped the genotypes into seven clusters, with the majority falling into three clusters. The study successfully identified genotypes with diverse and important traits and availed both the genotypes and the information for future breeding programs. These genotypes could be used for the development of improved hybrid and open pollinated shallot varieties with higher yield, quality and pest resistance/tolerance attributes.

  • Single and dual effects of different cytokinins on shoot multiplication of different apple scions

    Shoot multiplication responses of three apple scions to different concentrations of BA and BAR as single source of cytokinins and in combination with two concentrations of KIN were studied. The effects of hormones depended on genotype, type and interactions of different cytokinins. Use of BAR significantly enhanced the shoot multiplication of cv. Jonagold (6.5 shoots per explant). The multiplication rate of cv. Jonagold could not be improved by using the combination of BAR and KIN. The best proliferation was achieved by 1.0 mg 1-1 BA combined with 1.0 mg 1-1 KIN of cv. Prima..(8.1) and of cv. Galaxy (10.4).The effect of 0.5 mg 1-1 BA along with 1.5 mg 1-1 KIN was similar on multiplication rate (10.9) of cv. Galaxy.

  • Genetic engineering of apple (Malus domestica Borkh.) for resistance to fungal diseases using g2ps1 gene from Gerbera hybrida (Asteraceae)

    In the present study, g2ps1 gene from Gerbera hybrida coding for 2-pyrone synthase which contribute for fungal and insect resistance was used. The aim was to work out an efficient approach of genetic transformation for apple cvs. ‘Golden Delicious’, ‘Royal Gala’ and ‘MM111’, ‘M26’ rootstocks for improving their fungal resistance using genetic engineering techniques. Adventitious shoot formation from leaf pieces of apples studied was achieved using middle leaf segments taken from the youngest leaves from in vitro-grown plants.
    Optimum conditions for ‚direct’ shoot organogenesis resulted in high regeneration efficiency of  0%, 95%, 92%, 94% in the studied apples respectively. Putative transgenic shoots could be obtained on MS media with B5 Vitamins, 5.0 mg l-1 BAP, or 2.0 mg l-1 TDZ with 0.2 mg l-1 NAA in the presence of the selection agent “PPT” at 3.0-5.0 mgl-1. Shoot multiplication of transgenic shoots was achieved on: MS + B5 vitamins + 1.0 mg l-1 BAP + 0.3 mg l-1 IBA, 0.2 mg l-1 GA3+1.0 g/l MES+ 30 g/l sucrose + 7.0 g/l Agar, with the selection agent PPT at 5.0 mg l-1 and were subcultured every 4 weeks in order to get sufficient material to confirm transformation of the putative shoots obtained. Six, seven, one and six transgenic clones of the apples studied respectively have been obtained and confirmed by selection on the media containing the selection agent “PPT” and by PCR analysis using the suitable primers in all clones obtained for the presence of the selection” bar gene (447 bp) and the gene-of- interest “g2PS1” (1244 bp), with transformation efficiency of 0.4%, 0.6%, 0.1% and 0.3% respectively. These transgenic clones were multiplied further in vitro in the presence of the selection agent ‘PPT’ and rooted in vitro. Rooted transgenic plantlets were successfully acclimatized and are being kept under-containment conditions according to the biosafety by-law in Syria to evaluate their performance for fungal resistance .

  • Effects of methyl jasmonate, salicylic acid and phenylalanine on aloe emodin and aloin in diploid and tetraploid Aloe barbadensis

    Aloe vera is one of the most famous medicinal plants. Aloin and aloe emodin are the most important active compounds in this plant. The purpose of this research was the comparison of aloin and aloe emodin production after the elicitation by methyl jasmonate, salicylic acid, and phenylalanine in diploid and tetraploid Aloe vera plants in greenhouse conditions. The plants were treated with the concentrations of 25, 50, and 100 µM. The amounts of aloin and aloe emodin were determined 24 and 48 hours after application of the treatment. HPLC analysis showed that the leaves of the control diploid plants (without applying elicitors) had more aloin (1.20 fold) and aloe emodin (1.14 fold) than the control tetraploid plants. The maximum concentration of aloin (1.15 ± 0.07 µg mg-1 dry weight) was obtained after the elicitation by 25 µM methyl jasmonate, 24 hours after treatment, in diploid plants) 6.36 fold compared to the control (0.18 µg mg-1 dry weight (. In addition, the maximum concentration of aloe emodin (0.28 µg mg-1 dry weight) was obtained after the elicitation by 25 µM salicylic acid, 24 hours after treatment, in diploid plants) 6.18 fold compared to the control (0.04 µg mg-1 dry weight)). The long-term effect of three studied elicitors (after 240 days) on plant health and survival was also studied. This investigation showed that only methyl jasmonate at a concentration of 100 µM was resulted in the death of Aloe vera plants.

  • Effects of farmyard manure (FYM) and blended NPSB fertilizer rate on growth and yield of potato (Solanum tuberosum L.) in Gewata, Kaffa, southwest Ethiopia

    Potato (Solanum tuberosum L.) is an essential crop for nutrition and food security and generating income to small-holder farmers in Ethiopia. Productivity of the crop is, however, limited by poor nutrient management practices and low soil fertility. In most parts of potato growing areas of Ethiopia, soil are deficient in macro (nitrogen, phosphorus and sulfur) and micro nutrients (boron) which are known to affect production and productivity of potato. A field trail was accompanied to determine the growing response of potatoes ’Belete varieties’ to combine the treatment of farmyard manure and blended NPSB (nitrogen, phosphorous, sulfur and boron) fertilizer at Gewata, southwest Ethiopia during 2019/2020 main cropping season. The variables are laid out four level of farmyard manure (0, 5, 10 and 15 t/ha) and four level of blended NPSB fertilizer (0, 50, 100, and 150 kg/ha). The treatments were organized in a completely randomized block design with three replications using a 4x4 factorial layout. SAS (version 9.4) was used for data were analyzed. The finding indicate that, combined application of FYM (farmyard manure) with NPSB significantly (p<0.01) affects plant height, average mass of tuber, number of marketable and unmarketable tuber per hill, and tuber yields per plot. It also significantly (p<0.05) influenced days to flowering, maturity, and tuber number per hill. The highest tuber weight (99.33 g), tuber number per hill (9.67), and yield (34.10 kg) per plot were recorded from the mixed fertilization of 15 t FYM with 150 kg/ha NPSB mineral nutrients. As a current finding, it can be stated that the combined use of FYM (15 t/ha) and NPSB blended fertilizer could improve potato growth and yield in the study area (150 kg/ha).

  • Review of the molecular background of self-incompatibility in rosaceous fruit trees

    This review gives a presentation of the gametophytic self-incompatibility system in the roscaeous fruit trees. Studies to discover the pistil (S-ribonucleases) and pollen-part components (F-box molecules) are summarized and models for the self-incompatibility reactions as well as their molecular background are discussed. We describe how mutations within the S-RNase or F-box genes can contribute to the transition from self-incompatibility to the self-compatible phenotype in many fruit tree crops. The current state of the arts is compared to the information obtained in other plant species possessing similar incompatibility system.

  • Attempting Regeneration from Cultured Cotyledons and Plant Regeneration from Cotyledonary Nodes in Common Bean (Phaseolus vulgaris L.)

    Dry seeds from two cultivars of common bean (Phaseolus vulgaris L.) were germinated on sterile cotton and sterile deionized distilled water. Cotyledonary node tissue of seedlings were cultured on Murashige and Skoog(MS)-based media supplemented with different combination of N6-benzyl-aminopurine (BAP) and indole-3-acetic acid (IAA), and benzyladenine (BA) and a-naphthaleneacetic acid (NAA). The results revealed that the regeneration percent and the average number of buds and shoots per explant were influenced by the type of explants and exogeneously added hormones. Multiple shoot induction on dry bean cotyledonary node that contain 4-5 mm from cotyledons and hypocotyl on a medium containing full concentration of MS inorganic salts supplemented with 0.5mg/1 BA and 0.1mg/1 NAA was feasible and the method can be applied in transformation experiments.

     

  • Post-effects of cytokinins and auxin levels of proliferation media on rooting ability of in vitro apple shoots (Malus domestica Borkh.) 'Red Fuji'

    Rooting ability of in vitro apple shoots of 'Red Fuji' grown on proliferation media with different hormone content were tested at three IBA levels in root induction media. Rooting percentage could be slightly increased with an increase in IBA concentration in proliferation media. The highest IBA concentration (3.0 mg 1-1) in root induction media showed strong inhibitory effect on rooting capacity of in vitro shoots. The highest rooting percentage (95%) could be achieved by shoots grown on proliferation media containing TOP or BA+KIN as cytokinins before rooting.

     

  • Impact of organic and inorganic fertilizers on the growth and yield of cabbage in Ghana

    A field experiment was conducted at Dormaa Ahenkro, Ghana, to determine the impact of inorganic and organic fertilizers on the growth and yield of cabbage. The experiment was laid in a randomized complete block design with three replications. The experiment treatment comprises of sole application of poultry manure (PM), NPK 15:15:5, the combined application of poultry manure and NPK fertilizer (PM+NPK) and the control (no fertilizer). Data was collected on the number of leaves, stem girth, plant height, head diameter, head weight and edible head weight. There was a significant difference (P<0.05) among all the treatments. The combined application of PM+NPK recorded the highest values for all the parameters measured. From the results obtained farmers should consider the combined application of PM and NPK to maximize yield on their farm.

  • Preliminary study on micro area based spatial distribution of Monilinia fructigena in an organic apple orchard

    In this study, we aimed to report a preliminary study on micro area based spatial distribution of Monilinia fructigena in an organic apple orchard. Results showed that number of symptomatic fruit ranged between 22 and 42 in 2013 and between 25 and 35 in 2014. Number of asymptomatic fruit ranged between 111 and 187 in 2013 and between 119 and 167 in 2014. Disease incidence of fruit ranged between 19.7 and 23.2% in 2013 and between 19.1 and 26.5% in 2014. Disease aggregation index ranged between 0.111 and 0.335 in 2013 and between 123 and 401 in 2014. Three of the four trees showed significant within canopy aggregation of disease for fruit brown rot symptoms in both years. However, the remaining one tree exhibited random patterns during both years. Disease aggregation indicated a disease spread by fruit-to-fruit contact and/or an aggregated
    pattern of insect damage.

  • Effect of biochar and inorganic fertilizer on the quality of beetroot (Beta vulgaris L.) in Kenya

    Despite its health benefits, the production and quality of beetroot is still low in Kenya due to the application of non-recommended rates of fertiliser and soil amendment. This research aimed at contributing to the improvement of the beetroot quality in Kenya. It was designed to determine the effects of biochar and NPK (17-17-17) on the quality of beetroot in Kenya. An RCBD factorial experiment was conducted at Egerton University farm, Kenya, for two seasons.  Biochar (0, 5, 10 t/ha) and NPK (0, 200, 300 and 400 kg/ha) were applied together before planting. Data were collected on beetroot diameter, total phenolics, total soluble solids, calcium, iron and phosphorus contents and analysed using SAS statistical software. The co-application of biochar and NPK significantly (p≤0.05) increased the beetroot diameter, iron, calcium, phosphorus, TSS and phenolics content in season two and not in season one. The sole application of biochar showed a significant increase in the iron content of beetroot in season one. However, biochar did not have a significant effect on beetroot diameter, mineral content, TSS and phenolics content of beetroot in season two. The sole application of NPK at 200, 300 and 400 kg/ha significantly (p≤0.05) increased the diameter of beetroot and iron content in both seasons one and two and also significantly improved the calcium and phosphorus content in season two. These NPK levels were not statistically different from each other, but different from the control. It is therefore recommended to apply NPK and biochar for quality beetroot.

  • Farm-scale evaluation of two widely used sanitation practices for reducing primary inoculums of apple scab in organic apple production

    Effects of two widely used sanitation practices were evaluated at farm-scale level on leaf degradation and primary infection by Venturia inaequalis in an organic apple orchard (Eperjeske) on two apple cultivars (Jonathan and Prima) from 2011 to 2013. The tested sanitation practices were eradication of fallen leaves by collection and disc cultivation. Treatments of eradication of fallen leaves by collection and disc cultivation reduced signifi cantly (P< 0.001) leaf litter density with 70–85 and 40–55%, respectively, compared to untreated plots in both years. Above treatments in the same order reduced spring scab incidence with 40–50, and 10–20%, respectively, compared to untreated plots. Incidence of leaf scab in autumn was not signifi cantly lower (P< 0.05) in the treatments in the years.

  • Effects of activated charcoal on rooting of in vitro apple (Malus domestics Borkh.) shoots

    Rooting of in vitro 'Royal Gala' shoots was studied under different conditions of root induction and root elongation phase. The rooting capacity was affected by both rooting phases. Very high rooting percentage could be reached with both liquid and solid root induction media. Raising the temperature from 22 °C to 26 °C during root induction phase increased the rooting percentage. Presence of activated charcoal in root elongation media can affect the number of roots per rooted shoots and can increase the rooting percentage, the length of roots and the rate of survival depending also on other conditions during rooting. Presence of NAA in root elongation media reduced the number and the length of roots considerably. Favourable effect of activated charcoal on rooting was mainly due to adsorption of NAA.

  • Factors affecting rhizogenesis in vitro and acclimatisation of three cherry rootstocks

    Factors affecting rhizogenesis in vitro and acclimatisation of three rootstocks of cherry, i.e. Mahaleb, Maxma-14 and Weiroot -10 were investigated.

    Rooting was easily achieved within 2-4 weeks on MS-based liquid or agar-gelled media containing auxins IBA at conc. 0.49 or 2.45 pM or NAA at conc. of 0.49 pM. On liquid media with 2.45 pM IBA, a maximum rooting efficiency of 95-100% was obtained. However, high concentrations of auxin delayed the time of root initiation for 3-5 days.

    Rooted plantlets were transplanted into pots with a mixture of 3:1 (v/v) peat:perlite and acclimatised gradually to field conditions with efficiency of 60%.

    Systemic acquired resistance (SAR) is a mechanism of induced defense that confers long-lasting protection against a broad spectrum of microorganisms. Salicylic acid (SA) is the signal molecule which is required for induce SAR and is associated with accumulation of pathogenesis-related proteins, which are thought to contribute to resistance. SA paly vital role in some related resistance gene expression in plant cell which have direct or indirect effect on pathogen growth as SA has direct toxicity for pathogen and in the same time has stimulation effect for some enzyme related to reduce the oxidative burst.

  • Participatory evaluation of tomato (Lycopersicon esculentum Mill.) varieties under irrigation conditions at Abergelle district, Eastern Amhara

    Participatory on-farm evaluation of improved tomato varieties (‘Melka salsa’, 'Roma VF' and 'Kochero’) against the local tomato variety was carried out during the 2019/2020 growing season involving six farmers of Abergelle district, Eastern Amhara. The objective of the experiment was to assess the performance of different tomato varieties for the farmers, then to collect their feedback. Based on the actual and farmers’ preference data, the analysis underscores the better performance of improved tomato varieties over local varieties by most yield-related attributes. Average marketable fruit yields of ‘Melka salsa’, ‘Roma VF’, ‘Kochero’, and the local variety were 4.62, 3.88, 3.64, and 3.10 ton ha-1, respectively. The improved varieties thus had a yield advantage of 72.38%, 37.14%, and 25.72% over the local variety in that order. Among improved tomato varieties, ‘Melka salsa' provided the highest fruit yield on top of owing the highest score of overall preference attributes rank. Scale-wide diffusion of ‘Melka salsa’ tomato variety is therefore suggested for similar and potential agro-ecologies. Biological scientists also should take farmers’ preference attributes and feedback as a backup for future breeding and adaptation studies on tomato varieties.

  • Cytokinins affect the stomatal conductance and CO2 exchange of in vitro apple leaves

    Effects of different cytokinin supplies including two types of aromatic cytokinins, such as benzyl-adenine, and 3-hydroxybenzyladenine applied at two different concentrations (2.0 and 6.0 μM) were studied on water and gas exchange parameters in in vitro apple leaves of ‘Royal Gala’ and ‘Freedom’ scions after 3 weeks of culture. Cytokinin supply affected the stomatal conductance of water vapour, transpiration rate and the sub-stomatal CO2 concentration of leaves. Effect of cytokinin depended on its applied type and concentration, moreover on the apple scion. According to the results, the rate of CO2 exchange itself is not usable for characterization of function of photosynthetic apparatus of in vitro leaves. However, measurements of stomatal conductance of water vapour and transpiration rate seemed to be good indicators for stomatal behaviour of in vitro apple leaves.

    Sour cherry (Prunus cerasus L.) is one of the most important fruit crop grown in Hungary, The flowering phenology and pollen shedding/stigma viability ratio, in an effort to elucidate the reproductive phenology of nine economically important sour cherry cultivars (Érdi bőtermő, Debreceni bőtermő, Csengődi, Kántorjánosi 3. Pándy 279. Úfehértói fürtös, Petri, Éva, and Oblacsinszka) all widespread in the Hungary, was studied at Újfehertó climatic condition. The main aims were to investigate how possible environmental cues influence timing and development of phenophases and whether different cultivars have different seasonal responses to these cues. Likely effects of climatic changes on phenological development patterns were also considered. Our results revealed important aspects of the reproductive biology of sour cherry flowers. The amplitude of the phenophase "beginning of blossoming" between cultivars did not exceed 6 days. There is maximum 3 days difference in blooming length of different direction in each cultivar. Pándy 279 showed high variability when the position of flowers changed on the tree shoots. Újfehértói fürtös was stable in all four directions of tree. The pollen shedding period and stigma viability ratio was well synchronous. Pollen shedding phenomenon occurred in the range limitation of secretary activity of stigmas in all five cultivars. Distribution of pollen shedding over the secretary activity of stigmas is almost good. In all five cultivars maximum pollen shedding occurred about the high temperature part of the day.

  • Hazardous element content and consumption risk of 9 apricot cultivars

    The heavy metals pollution is one of the problems that arise due to the increased uses of fertilizers and other chemicals to meet the higher demands of food production for human consumption. In order to assess possible health risk of apricot (Prunus armeniaca L.) consumption, levels of Arsenic, Cadmium, Mercury and Lead were determined in fresh and dried samples of "Jumbo cot", "Tom cot", "Gold strike", "Gold bar", "Bergeron", "Bergarouge", "Sweet cot", "Yellow cot" and "Zebra" apricot cultivars. Wet digestion of samples with concentrate HNO3 – H2O2 digester mixture and inductively coupled plasma–atomic emission spectroscopy was used. Highest content of As, Cd, Hg and Pb among all cultivars, were 0.5, 0.04, 1.5 and 0.5mg/kg of dried apricot samples. Fresh fruit samples also contain 0.2, 0.016, 0.6 and 0.2 mg/kg of Arsenic, Cadmium, Mercury and Lead respectively. Daily intake of metals, hazard quotient and health risk index to reveal health risk possibility of dried and fresh fruits consumption were calculate and compared.

    Two table grape varieties — Kósa and Narancsízű — were registered of the hybrids bred in the Research Institute for Viticulture and Enology in I lungary. Both varieties ripe early and are much in demand for the attractive clusters and flavours. As they are hybrids of Vitis vinifera they require similar growing conditions. In the same time with their qualification the two varieties are included in the variety list and allowed to be propagated.

    Common bacterial blight (CBB), caused by Xanthomonas campestris pv. phaseoli (Xcp). is a major disease problem of common bean (Phaseolus vulgaris L.). The inheritance of resistance in Xrl and Xr2 lines to two isolates of Xcp was studied in the F2 and F3 popu­lations from the crosses between these lines and the Masay variety (susceptible to Xcp). Segregation patterns indicated that different single recessive genes presumably in coupling phase linkage determined the resistance to the HUN and EK-1 1 strains of Xcp in both lines. The presence of some minor, modifying genes beside the monogenic genetic background of resistance was also observed. Xrl and Xr2 lines represent valuable new monogenic genetic sources in resistance breeding to CBB.

  • Eicosapentaenoic acid production by Phaeodactylum tricornutum under different culture condition

    Phaeodactylum tricornutum UTEX 640 strain of microalgae was screened under different culture conditions for their capacity to produce eicosapentaenoic acid (EPA) the most abundant polyunsaturated fatty acid (PUFA). In our experiments, the effect of sodium chlorid, nitrogen source, phosphate, initial pH, as well as the CO2 content of the medium on production of the eicosapentaenoic acid (EPA) by P. tricornutum were investigated. The EPA content of biomass was enhanced by the low pH of the medium, with increased concentrations of B17 vitamin and nitrate, and also with decreasing concentrations of sodium chlorid. The EPA is most likely associated with polar (membran) lipids and the role of EPA appears to be involved with membran permeability in microalge. The synthesis of phospholipids, enhances the EPA content of the cells, as expected. The maximum EPA yields were observed under optimum culture condition 43 — 48 mg/g of dry cell weight.
