Peach latent mosaic viroid in naturally infected sweet cherry trees in southern Italy
37-38.Views:115Peach latent mosaic viroid (PLMVd) was found in naturally infected sweet cherry trees grown in commercial orchards in southern Italy. The viroid was detected in nucleic acid extracts of symptomless leaves by molecular hybridization with a PLMVd cRNA probe. The viroid was transmitted by grafting from sweet cherry to peach seedlings and identified in peach by molecular hybridization.

First results of varios sour cherry cultivar on Oblacsinska as interstocks and Prunus mahaleb rootstocks
87-90.Views:515At the experiment orchard of the Fruit Research and Extension Institute, Újfehértó, Hungary in spring 2000 a trial was designed included: Erdi bőtermő, Debreceni bőtermő, and Csengődi sour cherry (Prunus avium L) cultivars which were budded at 1999 on Oblacsinska sour cherry (as a dwarf inter stock), which had been budded on Prunus mahaleb seedlings at 1998 with the aim to investigate the dwarfing effects of Oblacsinska sour cherry cultivars on scion cultivars. Trees of each cultivar directly on Prunus mahaleb seedling, severd as controls. Use of Oblacsinska as an interstock reduced the rootstock trunk diameter. Trees of Debreceni bőtermő, and Cseng6di sour cherry cultivars with Oblacsinska interstock were smaller than similar trees grafted directly onto Prunus mahaleb as a rootstock. Trees of Erdi bőtermő grafted on Oblacsinska interstock which buded on Prunus mahaleb were the smallest. Growth of the trees was reduced also by using of the Oblacsinszka as interstock for other examined cultivars.

Above-ground dendromass of sprouted black locust energy plantations: a case study
19-21.Views:258Black locust (Robinia pseudoacacia L.) can be considered as one of the most suitable tree species for establishing energy plantations due to its favourable growing characteristics such as vigorous growing potential in the juvenile phase, excellent coppicing ability, a relatively high resistance to pests. Based on national and international test results the mean annual increment of oven-dry weight of energy plantations regenerated by coppicing generally exceeds the first cycle plantations established by seedlings.

Scheduling of ornamental plant production
76-86.Views:165The production of ornamental plants represents an important branch of our horticulture. The growing area is relatively small (round 2800 ha), the production value, however, a rather large. Notwithstanding, its financial balance is inactive because the demand surpasses the supply copiously.
The most popular plants are as follows: carnation, gerbera, rose, bulbous flowers, chrysanthemum, other cutflowers, cutgreens and Gypsophyla — their total production value makes up to Ft 8-9 billion. The area of ornamental nurseries — about 800-900 ha — is to be found mostly on the western part of our country.
Our accession to the EU will have undoubtedly an impact on our ornamental plant production. We must take into account, that in greenhouse production the specialization extends all over the world, field production over a limited region. Our chances will not deteriorate by joining the EU. The buyer — chiefly because of ecological purposes — will prefer the domestic product to the foreign one. By the way, our products will be competitive, as far as quality or price is concerned, with those of western Europe. Last but not least, we may hope a greater saleability of home-bred, special varieties and cultivars, the so-called "hungaricums", both in the domestic as well as export markets. It is anticipated that we will have a good turnover with potted ornamental plants and flower seedlings. Our nursery products will become marketable too.
To exploit the opportunity, of course, the necessary conditions are to be created. First of all, we need development in research, with special regard to breeding, at the same time in education, in extension service, in the training of experts, on a high level. Some tasks can be solved, no doubt by improvement of the organisation within the branch. At the same time the state subsidy is indispensable in order to promote both the technical and the research activities. State subsidy is also necessary to build up more advantageous conditions of sales like in some foreign countries (e.g. the Netherlands).

Morphological examination of Hungarian apricot rootstock varieties
59-63.Views:289Apricot seedlings are one of the most widely used rootstocks in apricot growing of Hungary. The National List of Hungary contains 7 apricot seedling rootstocks varieties. The production of apricot graft on seedling rootstock takes 3 years from the seed production to the complete graft and requires more space. It is important to provide the trueness to variety name during this process. In the genus Prunoideae not only the plant but also the stone should have typical characteristics. It seems to be useful to examine the morphology of stone. The observed varieties were 'Tengeribarack C.1300', 'Tengeribarack C.1301', 'Tengeribarack C.1650', 'Tengeribarack' C.1652', 'Tengeribarack C.145', 'Tengeribarack C.1426' and 'Tengeribarack C.2546`.1t seems to be the most suitable characteristic to make a distinction between these varieties by stone, the ratio height of stone to lateral width of stone, the ratio of height of stone to ventral width of stone, the ratio of lateral width of stone to ventral width of stone and the ratio of ventral zone to ventral width. From not measured characteristic seems to be most utilizable to make distinction: the fusion of dorsal groove margins, the shape of apex, the presence of a mucro of the apex, the shape of stalk end in lateral view and the texture of lateral surfaces.

Organogenesis in eggplant (Solanum melongena L. cv. Embu) as affected by antibiotics and growth regulators
76-82.Views:258The influence of antibiotics (cefotaxime, timentin, kanamycin and hygromycin) and growth regulators (indolacetic acid and 6-benzylaminopurine) was evaluated on eggplant organogenesis. Solanum melongena hypocotyl segments (6 to 10 mm length), taken from 16 to 20-days in vitro grown seedlings, were used as explants. The basic medium was composed by MS salts, Gamborg vitamins and 2% sucrose, solidified with agar 0.8% and pH adjusted to 5.7±0.2. Morphogenesis was impaired at 50 to 100 mg L-I kanamycin and 7.5 mg L-1 hygromycin. Both Timentin and cefotaxime reduced the frequency of regenerating explants meanwhile hyperhydricity was not affected. A decrease in root regeneration was observed with increasing cefotaxime concentrations, although, timentin had no effect on root regeneration, as compared to the control treatment. Interestingly, the number of adventitious roots was more noticeable at 0.25 mg L-I IAA plus 0.5 mg L-1 BAP. However, if just IAA was added led to higher number of regenerated roots compared to other treatments.

Anatomical relations of root formation in strawberry
71-75.Views:215Anatomical relations of root formation are traced throughout the life cycle of the strawberry plant from the germinating seed up to the runners of the adult plant. Histological picture of the root changes a lot during the development of the plant. First the radicle of the germ grows to a main root, which makes branches into side roots and later adventitious roots are formed on the growing rootstock or rhizome. The anatomy of the different types of roots is also conspicuously different. First tiny branches appear relatively early after germination on the seedling's radicle, but soon the hypocotyl of the seedling thickens and develops side roots, which are already somewhat stronger. During this interval, the first true leaves are formed. The 4th or 5th of them being already tripartite, and the initiation of new roots extends into the epicotylar region of the shoot. The second years growth starts with the development of reproductive structures, inflorescences and runners starting from the axils of the new leaves. Near the tips of the runners below the small bunch of leaves, new root primordia are initiated. The tiny radicle of the germ develops a cortical region of 5-6 cell layers. Cells of the central cylinder are even smaller than the cortical parenchyma and include 3-4 xylem and 3-4 phloem elements as representatives of the conductive tissue. Roots originating from the shoot region are much more developed; their cortical zone contains 17-20 cell layers, whereas the central cylinder is about half as large. In the next year, new roots are formed at the base of the older leaves. These roots differ hardly from those of the last season in size and volume, however, they are recognised by colour and their position on the rhizome. The roots of the last year are dark, greyish-black, and grow on the lower third length of the rhizome, on the contrary, the new ones, on the upper region, are light brown. Roots starting from the shoot or rhizome are, independently from their age or sequence, mainly rather similar in size and diameter, thus being members of a homogenous root (homorhizous) system, i.e. without a main root. Plants developed and attained the reproductive phase develop in the axils of the leaves runners being plagiotropic, i.e. growing horizontally on the surface of the soil. The runners elongate intensely, become 150-200 mm, where some long internodes bear a bunch of small leaves and root primordia on short internodes and a growing tip. Runners do not stop growing, generally, further sections of 15-25 cm length are developed according to the same pattern, with small leaves on the tip. The growing tip of the runners is obliquely oriented, and small, conical root primordia are ready to start growing as soon as they touch the soil. The roots penetrate the soil, quickly, and pull, by contraction, the axis of the runner downwards, vertically, developing a new rhizome. The short internodes elongate a little and start developing adventitious roots. At the end of the growing season, the plantlets arisen on the rooted nods of runners are already similar to the original plants with homogenous root system. On the side of the adventitious roots, new branches (side-roots) are formed. The root-branches are thinner but their capillary zone is more developed being more active in uptake of water and nutrients. The usual thickening ensues later.

Self-incompatibility alleles in Esatern European and Asian almond (Prunus dulcis) genotypes: a preliminary study
23-26.Views:212Almond [Prunus dulcis (Mill.) D. A. Webb.] as one of the oldest domesticated plants is thought to have originated in central Asia. Gametophytic self-incompatibility of almond is controlled by the highly polymorphic S-locus. The S-locus encodes for an S-ribonuclease (S-RNase) protein in the pistils, which degrades RNA in self-pollen tubes and hence stops their growing. This study was carried out to detect S-RNase allelic variants in Hungarian and Eastern European almond cultivars and Turkish wild growing seedlings, and characterize their S-allele pool. Five new alleles were identifi ed, S31H, S36-S39 in Eastern European local cultivars. The village Bademli and Akdamar island are two distinct places of almond natural occurrence in Turkey. Trees growing wild around Bademli city showed greater genetic diversity than those originated on Akdamar island. Many of the previously described 45 S-RNase alleles have been also detected in these regions. Homology searches revealed that Turkish almonds carried some P. webbii alleles indicating hybridization between the two cultivars and massive introgression events. Our results supply long-awaited information on almond S-allele diversity from regions between the main cultivation centres and the centre of origin of this species; and are discussed from the aspect of methodological developments and evolution of the cultivated almond.

Effect of hydroponic and peat-free media in transplant production of Rudbeckia hirta varieties under different photoperiodic lighting and their photosynthetic parameters
110-117Views:405The purpose of this research was to determine the effects of varieties, different light conditions (short day, long day, natural short day with light pollution), and different growing media (perlite, peat-free, peat-based, aeroponics system) on Rudbeckia hirta plant production under controlled conditions (greenhouse). The morphological effects of each treatment (photoperiodic lightings and media) on different Rudbeckia varieties determined at 11 weeks-old ’Napfény’, ’Toto Gold’, ’Autumn Colors’, ’Prairie Sun’ and 16 weeks-old ’Napfény’. Plantlets received 12 hours daylight did not initiate flowers, remained stage of the leaf rosette in case of all varieties. The 14 hours light treatment in the aeroponics system and the same treatment in perlite and control (natural short day with 14 hours light pollution) plantlets had developed inflorescences or flower buds. The inflorescence axis of ‘Napfény’ was appeared at 13 weeks under long-day conditions, with 1.7 (perlite) - 2.7 (aeroponics) flower buds in 16 weeks. ’Toto Gold’, ’Autumn Colors’, ’Prairie Sun’ varieties developed inflorescences at 8 weeks, 14 hours aeroponics system resulted in the most of flower buds (’Toto Gold’: 6.5, ’Autumn Colors’: 3.25,’Prairie Sun’: 4.8 flower buds) at 11 weeks. Long daylight manipulation could be minimized crop times and achieved flowering potted plants at 11 weeks. The peat-based and peat-free media effect was observed on ‘Autumn Colors’. The number of leaves of peat-free ‘Autumn Colors’ transplants (16.8-20.3) was significantly higher than peat-based media (13.5-15.5). Other morphological parameters were not affected by the media treatments.

The use of rootstocks for European (Prunus domestica) and for Japanese (Prunus salicina) plums (review)
7-13.Views:491The worldwide tendency to increase the intensity of fruit growing technologies prefers generally for every fruit species rootstocks with week or mediocre vigour. From this viewpoint, the use of rootstocks for plums are rather unilateral in Hungary, where 95–99% of plum plantations are grafted on mirobalan seedlings (P. cerasifera v. mirobalana). The score of plum rootstocks abroad is much more diversified. The present study summarises the respective knowledge referring to the literature available.

The Effects of Some Parameters on Agrobacterium-Mediated Transformation in Muskmelon
46-49.Views:186Some parameters involved in Agrobacterium-mediated transformation in muskmelon Hales best (HBS) were studied. Cotyledon explants excised from 3.5-day-old seedlings were co-cultivated with Agrobacterium tumefaciens harbouring binary vectors which contained GUS and BAR genes. After co-cultivation on a low pH medium, explants were transferred to selective medium, with higher pH, containing Claforan and Finale. The medium was changed every two weeks till shoots were induced. All shoots rooted on MS medium supplemented with 0.3 mg/L IBA. These parameters combined as a whole led to successful transformation. The expression of the introduced gene construct was confirmed by GUS staining of shoot segments.

Propagation from root cuttings for black locust (Robinia pseudoacacia L.) improvement in Hungary: a review
39-41.Views:337Black locust (Robinia pseudoacacia L.) is a valuable stand-forming tree species introduced to Europe approximately 400 years ago from North America. Today it is widely planted throughout the world, first of all for wood production. In Hungary, where black locust has great importance in the forest management, it is mainly propagated by seeds. But since the seed-raised plants present a great genetic variation, this type of propagation can not be used for Robinia’s improved cultivars. In the Hungarian black locust clonal forestry, propagation from root cuttings can be used for reproduction of superior individuals or cultivars in large quantities. However, this method demands more care than raising seedlings from seeds and can be applied with success in well-equipped nurseries.

Exotic woody plants inclining to escape in the Buda Arboretum under strong urban effect in Budapest
93-97.Views:171The Buda Arboretum of the Szent István University is situated in the western-middle part of the city of Budapest. It contains 1640 taxa of woody plants on an area of 7.5 ha. The sheltered position and the urban micro- and meso-climate is favourable for warm-loving, even subtropical plants, of which many species not simply stay alive, but flower, bear fruits and are spreading by seedlings in suitable spots. These instances call our attention to the possibilities that urban climate offers for landscapers, but also to over 110 species, which can be garden escapes in the future. Full list of plants, which tend to escape in the Arboretum is given in the paper.

Sweetcorn production from transplants
45-48.Views:185In our trial we tried to find out how the time of propagation and transplanting influenced the growing season of sweet corn along with some major properties relevant to quality. The following technological variations were compared with the help of the variety Spirit (normal sweet, very early ripening): transplanted plants with floating row cover (with 2 planting dates); transplanted plants with no row cover: direct seeded plants with no row cover. The 21 day transplant growing period reduced the growing period by 16 to 20 days, compared to the technology used in the existing practice of production. Earliness had a negative influence on ear size, nonetheless it is worth while to attempt since the market is not so exacting with new products in the early period. Covering the seedlings in the early season was clearly beneficial, as the floating row cover provided protection for plants against lower night temperatures which are common in this period.

Genetic transformation of bean callus via Agrobacterium- mediated DNA transfer
49-53.Views:132Callus cultures were induced from hypocotyl of young bean seedlings. Callus developed and maintenaned on B5 medium supplemented with 2mg/1 2,4-D and 1 mg/1 kinetin. The results demonstrate that A. tumefacins-mediated transformation is a convenient method to obtain transient gene expression in callus of bean. The results have shown that the bean callus co-cultivated with A. tumefaciens can be transformed to get heibicide Finale (glufosinate-ammonium) resistant GUS positive tissues. Southern blot analysis of transformed calli showed integration of gusA marker gene carried by a binary vector. Transformed calli were selected on herbicide containing media. Data of molecular analysis (Southern blotting) confirmed the insertion of gusA gene in the genome of herbicide resistant calli with bar gene. There are three evidences that calli are stable transformants: (1) herbicide resistance, (2) GUS activity which is indicative since the coding region containing an intron, (3) the results of Southern hybridization technique.

Comparative investigations on protoplast culture of some Brazilian and Hungarian sweet pepper cultivars and hybrids
39-45.Views:199Cotyledon protoplasts were isolated from 16-18-day-old in vitro grown seedlings of 9 Brazilian and 3 Hungarian pepper varieties and hybrids. Large numbers (average 9.59 X 106 protoplasts g 14 fresh weight) of highly viable (average 87.0%) protoplasts were released using a pectocellulolytic enzyme mixture. Protoplasts were cultured in K8p mediuni using an alginate disc embedding method. The osmotic pressure of the medium surrounding the alginate-embedded protoplasts was reduced by replenishing the liquid medium at K8p:K8 ratios of 1:0. 2:1, 1:1 in the first. second, and third week, respectively. Initial plating efficiency (IPE) average was 38.5% and after 21 days protoplasts reached microcolonies (15-20 cells) stages. Microcolonies were transferred after 3-4 weeks to a MS-based medium supplemented with 1.0 mg I-1 zeatin, 3.0% (w/v) sucrose, 0.24% (w/v) phytagel and pH 5.8, whereupon they formed callus. Final plating efficiency (FPE) average was 0.29% at a plating density of 1.0 x 105 protoplasts Protoplast-derived calli were cultured on a range of MS-based media supplemented with either BAP, IAA, TDZ; and zeatin. No morphogenic response was observed in any genotype investigated.

Application of the Jerusalem artichoke (Helianthus tuberosus L.), as a plant origin medium additive, during the micropropogation of Ada keiliana
61-64.Views:196A procedure for in vitro propagation of Ada keiliana seedlings are suited for acclimatization, was worked out. M medium was supplemented, with Jerusalem artichoke, as plant origin complex additive. The apply of JAD (1,5g/flask) gave the best response, considering the shoot (29 mm), and the root development (24,9) mm) too. The plantlets with satisfying growth (25-30 mm, 4-5 roots) were transferred in small pine bark: Novobalt peat: coconut fibres: perlit (2:3:1:1) mix, among greenhouse circumstances.

Production of transgenic bean callus via genetic transformation by DNA-coated tungsten particles
43-47.Views:122Callus cultures were induced from hypocotyl of young bean seedlings. The B5 medium completed with 1 mg/1 KIN and 2mg/1 2,4-D proved the best. Callus developed and maintenaned on B5 medium supplemented with 1mg/1 kinetin and 2mg/I 2,4-D. The B5 medium supplemented with 1mg/1 KIN and 2mg/1 2,4-D induced much more callus than half strength MS medium supplemented with 0.5 or 0.75mg/1 BA and 0.1 mg/1 NAA. The results demonstrate that GeneboosterTM is convenient method to obtain transient gene expression in callus of bean. The results have shown that the bean callus shot by GeneboosterTM can be transformed to get (kanamycin-resistant and stress mannitoltolerant) calli. The presence of mannitol-dehydrogenase gene (mt/) was verified by PCR, showing the integration of mt/ gene carried by two plasmids. Co-transformed calli were selected after bombardment on kanamycin, mannitol and (kanamycin+mannitop-containing media. Data of molecular analysis (PCR) confirmed the insertion of mtl gene in the genome of mannitol-tolerant callus lines.

Influence of antiobiotics on NAA- induced somatic embryogenesis in eggplant (Solanum melongena L. cv. Embil)
88-95.Views:187The influence of increasing concentrations of naphthaleneacetic acid and the antibiotics cefotaxime, timentin, kanamycin, and hygromycin on eggplant (Solantun melongena L. cv. Embil) somatic embryogenesis was investigated. Cotyledon explants were excised from 16 to 20 days old in vitro grown seedlings. NAA promoted somatic embryogenesis, although its concentrations had no influence on the mean number of embryos. Callusing decreaSed significantly with increasing NAA concentrations. Morphogenesis was stopped with 50 to 100 mg L-1 kanamycin and 7.5 to 15 mg L-1 hygromycin. Although early globular embryos were observed up to 15 mg L-1, further embryo development was inhibited at 10 mg L-1. Interestingly, cefotaxime (250 and 500 mg L-1) promoted a marked effect on enhancing fresh weight of calli, accompanied by decrease in embryo regeneration, whereas timentin concentrations (150 and 300 mg L-1) did not affect embryo differentiation as compared to the control treatment.
