  • Production of transgenic carnation with antisense ACS (1-aminocyclopropane44-carboxy late synthase) gene

    Dianthus chinensis and Dianthus caryophyllus varieties were tested for shoot regeneration from leaf and petal explants and transformed with Agrobacterium tuniefaciens strains (EHA 105 and LBA 4404) harbouring an apple derived ACS cDNA in antisense orientation in order to reduce ethylene production and influence the ethylene dependant traits in carnation. After transformation regenerating shoots were selected on MS medium containing 50-75-100-125-150 mg/1 kanamycin and supplemented with 1 mg/1 BA, 0.2 mg/1 NAA. Transgene integration was proved by PCR analysis with npt II spcific primers followed by Southern hybridisation of DNA isolated from green shoots on medium containing 150 mg/1 kanamycin. Several putative transformants were subjected to RT-PCR in order to examine the npt 11 expression at mRNA level. Both the transformant and the non-transformant plants were potted into glasshouse to observe the effect of changed ethylene production on flowering time, petal senescence and vase life.

     

  • Agromorphological and nutritional quality profiles of fluted pumpkin (Telfairia occidentalis Hook F.) as influenced by cultivar, growing medium and soil amendment source

    Fluted pumpkin (Telfairia occidentalis Hook F.) is popular as food and feed around the world. Sixteen treatments were developed from factorial combinations of three factors: cultivar (ugu elu and ugu ala), growing medium (garden soil (GS) and white sand (WS)), and soil amendment source (poultry manure, NPK, supergro and no amendment). A pot experiment was conducted to investigate the agromorphological and nutritional traits of fluted pumpkin obtained from the treatments. Fresh leaves were analyzed for crude protein, crude fibre, crude lipid, total ash, phytate and nitrate concentrations. Data were subjected to analysis of variance and principal component analysis. Mean plots were used to explain the effects of the three factors and profiling was done using the GYT biplot. There were significant (p≤0.05/0.01) mean squares for measured traits, suggesting the possibility of selection among the treatments. Plants in GS consistently out-performed those in WS for shoot weight, leaf length, and number of leaves per plant possibly due to greater availability of nutrients in the GS. Inconsistent patterns observed in the proximate concentrations of pumpkin from the 16 treatments showed the role of interaction among the three factors. Principal component analysis identified some traits as contributors to differences among the treatments which can be basis of selection. Treatments 2, 4, 6, 8, 10, 11, 12, 13, 14, 15, and 16 might be useful to improve vegetative yield while 1, 3, 5, 7, and 9 could improve nutritional values of the fluted pumpkin.

  • Changing of inert content values of apricot varieties

    The consumers have been exigent increasingly about excellent quality. The speci fie flavour and fragrance of apricot evolve during ripening. We need to be acquainted with changing of inert content during ripening to the choosing of optimal harvest time. The Regional University Knowled ge Centre -Research and Development in Foodstuff Chain project had presented an opportun ity to study the inner content of apricot. Our aim was to study evaluation of health care attributes of apricot fruit. We investigated different ripeness samples of four apricot varieties ('Mandulakajszi', 'Pann 6nia', 'Gonci magyar kajszi'. 'Harcot') in our study of 2007-2009. Samples were taken the orchard of Sóskútfruct Ltd. Hungary. We had measured the water soluble antioxidant capacity, polyphenol-, f3-carotene content and the dissimilar sugar­ and acid-fractions during ripening. We had experienced significant different in inert content between varieties.

  • Self-incompatibility in pears (Pyrus communis L., Pyrus serotina Rehd. and Pyrus ussuriensis) Review

    Self-incompatibility system and allele pool of three different pear species, European pear (Pyrus communis), Japanese pear (P. serotina) and Chinese pear (P ussuriensis) are displayed. Several inconsistencies and the absence of the harmonization of three different allele series are revealed in the European pears. By collecting data from several reports eight incompatibility groups of Japanese pear cultivars could be established. A self-compatible genotype is analysed in details and shown to be a stylar-part mutant. As Japanese pear was the first fruit tree species from which S-ribonucleases were identified, the history of S-genotyping from the beginning to the latest achievements and technical developments can be also monitored from the experiments enumerated. In Chinese pears, seven S-alleles and one incompatibility group could be identified.

  • Assessment of fruit firmness of pepper using non-destructive physical measurements, in response to different growing and pruning technologies

    The European market demands vegetable products of the highest quality and this commercial quality must be maintained till the goods reach the customer. One of these important quality parameters is the fruit firmness of pepper.

    The experiments were aimed at to find out the influence of different growing methods (soil or rockwool-based) and pruning technologies (to 1, 2, 3 or 4 shoots) on the yield and fruit firmness of three pepper varieties (HO F1, Karpia Fl and Pritavit F1) which are common in Hungary. Fruit firmness was measured by the non-destructive impact method.

    On the basis of the results, in unheated forcing the pruning to 1, 2 or 3 shoots can be suggested for all three varieties, as well as the utilisation of rockwool in their growing.

    The non-destructive impact method has been found suitable for testing the fruit firmness of pepper varieties. In the experiments involving different growing mediums pepper stands were found to show significant differences, however the different pruning methods had no significant influence on fruit firmness.

  • Dynamics of the uptake of nutrient elements from the medium of in vitro cultured apple rootstocks

    Cation uptake of J-TE-F apple rootstocks propagated in vitro was studied by the analysis of culture medium. Test-plants were grown on liquid medium under different light regimes. Samples for tests were taken twice a week.

    Media without plants served as controls. The analysis of those showed, that only the uptakeable iron-content changed depending upon light treatment. The concentration of all other cations was considered unaltered.

    As a result of analysis, it could be established, that elements present in the media were taken up in different rates by plantlets: Cu, P and Zn were utilized totally, but only 50% of K and 20 to 40% of Ca and Mg were taken up under the light treatments applied.

    The dynamics of the uptake process was also observed. It was registered that they differed in the case of some cations. So Ba was utilized at the beginning of subculture, others for example B in the later phases. Some elements disappeared unevenly so K, P but the whole quantity is taken up during subculture.

  • In vitro comparative study of two Arundo donax L. ecotypes’ selenium tolerance

    Selenium tolerance of two somatic embryo-derived Arundo donax L. ecotypes (Blossom, 20SZ) were compared in in vitro culture. Sodium-selenate (1 – 100 mg L-1) as known the most phytoavailable selenium form and the less studied red elemental nanoselenium (100 mg L-1) were applied as selenium treatments. Basis on the results Blossom ecotype seemed to be more sensitive to the sodium-selenate than 20SZ. Inhibiting effect of selenate was effectuated above 10 mg L-1 in case of Blossom, which was manifested in decreased survival rate and growing parameters. Contrast to this 20SZ could tolerate the selenate ≤ 20 mg L-1 without any toxic symptoms. Lower selenate tolerance of Blossom could be explained with higher selenium accumulation. Both of two ecotypes could also uptake and accumulate the red elemental nanoselenium however in much less extent compared to selenate.

  • Challenges of the vegetable and fruit market

    The situation of the horticulture sectors have been in the limelight of the professional and economic decision makers all over Europe. This article analyses the situation of the sector from economic point of view and reveals the main reasons of its low income and high risk. It concludes that one of the biggest problems is the trading uncertainty in the vegetable and fruit sector that is caused by the asymmetric market structure of the post-regime era. Since sizes of vegetable and fruit plantations do not allow producers to supply individually the extremely concentrated food retail trade or the processing trade they must find alternative ways for trading their products. The study introduces two alternative solutions. One alternative is foundation of modern multi-level producer co-operatives with the help of EU subsidies. Secondary and tertiary co-operatives may achieve better market position and lower trading price risk with managing production, professional marketing, and improving the information flow. The other alternative is searching for new trading channels such as local provision, restructuring of local markets, and direct trade (home delivery and pick-it-yourself programmes). The shorter producer-consumer distance means better quality at lower price for customers and income in the case of smaller amount of products for producers. It is concluded that both solutions together or separately may help individual producers in their trading problems. However, whichever way they choose, producers must co-operate.

  • Frost induced changes in enzyme activities and carbohydrate content in the spurs of some pear cultivars during the dormancy

    Frost tolerance of pear cultivars was checked after artificial cold treatment in 2003-2005. Limbs collected during the endodormancy were exposed in a climatic chamber for 24 hours to —25; —28 °C, while those collected in the ecodormancy were kept at —15 and —18 °C. Frost damages of buds were registered according to a visually defined scale, then peroxidase (POD), polyphenol oxidase (PPO) enzyme activities and carbohydrate contents were checked in buds and spur-part below the buds. POD activity of untreated control in tissue below buds was higher than in the buds, which were increasing continuously during the endodormancy and decreased at the end of the ecodormancy. During endodormancy, cold treatment of —25 and —28 °C effected different changes of enzyme activity in buds of the cultivars. In the ecodormancy, enzyme activities increased after a cold treatment of —15 °C, whereas the activities decreased significantly after —18 °C. `Kaiser' — susceptible to frost — with its higher values of both enzyme activities marked out from other cultivars, which is correlated with its stress response. Changes in carbohydrate components — especially in glucose — of buds monitored well the different stress responses of tolerant and resistant pear cultivars induced by frost stress.

  • Texture changes of vegetable cultivars measured by non-destructive methods

    According to our experiments the tested group of non-destructive methods offers a useful tool not only to follow the texture changes of vegetables during storage but to characterize the firmness and vision parameters during the growing period as well. Advantages of these methods are: they are mobile, easy to set up, easy to use and quick. The suitable maturity state — and so the optimum harvest date also - can be determined by these methods. In addition, these methods can be built into sorting lines making possible to sort and classify great amounts of produces. These methods help the producer to offer homogenous products of controlled quality. They can be used for measuring different effects on vegetable's quality parameters (fertilization by different microelements, different irrigation effects on the product) as well.

  • Observing population changes of thrips (Thysanoptera) species damaging forced pepper and their natural enemies

    By the strengthening of environmental protection and food safety efforts in Hungary, integrated and especially biological pest control methods should increasingly put forward, for which a solid knowledge on the life course and efficiency of natural enemies applied against certain pests is necessary. Pepper has distinguished significance in domestic vegetable forcing, and the profitability of production is determined primarily by the efficiency of the control of thrips pests. This is why we attached great importance to study what results may be expected by introducing arthropod predators (Amblyseius cucumeris, Orius laevigatus) to control thrips species under domestic conditions on rock wool in a long vegetation period pepper culture. We also liked to find out what kind of role the cultivars play in the change of phytophagous and zoophagous populations. The A. cucumeris predatory mite introduced in late January proved to be effective in controlling thrips pests until mid-April. Despite repeated introductions, the predatory bug O. laevigatus (Heteroptera: Anthocoridae) did not proliferate. Among the three pepper cultivars (Hó, Keceli, Titán) grown at Ráckeve, thrips species proliferated in the highest number on cultivar 'Hó', while the population of predatory mites was lowest on the cultivar 'Titan', compared to the other two cultivars.

  • Nutritional aspects of producing fruits organically

    Recent interest to avoid use of agrochemicals in fruit growing to safeguard environmental and human health has stimulated interest in organic fruit production (OFP) all over the world. Organic production requires a holistic approach to agricultural ecosystem management. Because of the perennial nature of orchards, this is not a great departure from conventional orchard management, except that corrective techniques are limited primarily to naturally derived materials. Organic mineral nutrition management hinges on two principles: 1) practices that lead to the build up and maintenance of soil that is biologically active and high in organic matter; 2) supplementing the mineral nutrients provided by the soil with fertilizers from approved sources. Organic orchards should be sited on land with superior soils and preplant soil preparation to increase organic matter and correct any sub-optimal soil characteristics. For successful organic fruit growing the following statement should be considered:Weed management is critical to reduce competition for nutrients and water. Soil and leaf analysis provide the basis for correcting mineral nutrient deficiencies or imbalances in organic production. It may be necessary to use a number of strategies to supply mineral nutrients over the life of the orchard. The slower, natural methods applied require a management approach that is simultaneously patient and dynamic.

  • The impact of irrigation on the yield and tuber fraction distribution of potato cultivars

    Potato is one of the most important plants in human nutrition, it is grown in about 140 countries. As a food, it can be prepared in many ways, it is easily digestable with proteins of high biological value, favourable dietetical-physiological characteristics, high starch content and good taste. An increase in its consumption would be favourable from the dietetical point of view. In Hungary, the growing area of potato has decreased considerably for the last decades and we are lagging behind the Western-European countries regarding yield. The experiments were carried out at the Experimental Site of the University of Debrecen at Látókép between 2002 and 2004. In the large-plot experiment, the yields and the distribution of the tuber fractions were studied for 9 medium early cultivars. The experiment was set up on 50 m2 plots on calcareous chernozem soil. Among the studied cultivars, 3 were of Dutch (Desiree, Kondor, Kuroda) and 6 were of Hungarian breeding ('Góliát', 'Hópehely', 'Kankan', 'Lilla', 'Százszorszép', and 'White Lady'). The experiment was set up in 4 repetitions in a randomized design, two of the repetitions were irrigated while two were non-irrigated.

  • Anatomical structure of stem in the above- and underground portions of Rosa rugosa suckers

    Comparitive histological studies were made on the underground and the aboveground stem parts of Rosa rugosa taken from one year old suckers. The underground stem parts were characterized with thicker primary cortex, phloem and pith, weaker phloem fibers, wider cambial zone, medullary rays, xylem and phloem rays as compared with the aboveground stem parts. The most marked differences in the underground stem parts were in the wide cambial zone and in the development of some adventitious roots.

     

  • Colour stability of the flowers in some modern rose varieties in Hungary

    A variety trial has been carried out to study the colour changing — colour stability of the flowers of some modern roses, especially of Hungarian varieties. 100 floribundas and polyanthas were observed. Colours of the petal surface were examined at three fenological stages of the flowers: at the bud, at the young open flowers and at the aged open flower stages. The colour difference was described in HLS standard. Our work shows that valuable Hungarian varieties can be found in both studied classes, but more floribundas bred in Hungary had good colour stability than polyanthas. The best Hungarian floribundas were 'Pest' and 'Remenyik Sándor emléke', although 'Regen', `Szent Lász1ó em­léke' and 'Szabó Dezső emléke' were quite good. The best Western-European floribunda rose was 'Perneille Poulsen'. Their colour stability was reliable in different situations. Colour changing of the two best varieties was almost unperceivable from the bud to the young petals and from the young to the aged petals. In the polyantha class there was only one perfect variety: the Czech `Cs1 Cerveny Kriz'. The 'Fairy Damsel' was almost excellent as well. The best Hungarian polyanthas were `Domokos Pál Péter emléke' and 'Savaria'.

  • Correlation between pigment contents and FRAP values in beet root (Beta vulgaris ssp. esculents var. rubra)

    It is well known that beetroot quality is determined mainly by the red pigment content (betacyanins) and its uniformity of the root. The effect of the most important red pigment components (betanin). the total polyphenol content and antioxidants were studied in 20 beet root varieties. Antioxidants were expressed in FRAP (ferric reducing ability of plasma) values in pM/I.

    Our results indicated a close correlation (r = 0.7799 and r = 0.7435. respectively) between betanin and total polyphenol contents of the root as well as between FRAP values.

    Our measurements showed more than threefold differences in total antioxidant activity among varieties. the lowest value being 196.4 13M/1 and highest 702.57 pM/I. The corresponding betanin (16.3 and 57.8 mg/100 ml) and total polyphenol (37.5 and 85.5 mg/100 ml respectively) contents show similar differences. Based on our results it can be stated that varieties of higher betanin and poliphenol contents have higher antioxidant values as well.

    Accordingly, the two compounds must have a role in the evolution of antioxidant effects.

  • Effect of physical treatments on germination of Ginkgo biloba L.

    In our country the maindenhair tree (Ginkgo biloba L.) is raised mainly from seeds, so the aim of our experiments was to determine the most useful generative propagation method. However, some experiments have been conducted earlier connected to the germination of the species, but the comparative control of physical seed treatment was done first time by the authors.

    After the statistical evaluation of the results it can be stated that the percentage of germination has significantly increased if the seeds received physical treatment (scalding, mechanical scouring). Hereby the pericarp is getting soft or growing thinner, so the germination of the seed is easier. These treatments are extraordinarily simple, easy to carry out and their effect is very favourable, that is why their use is strongly advised.

    Comparing the seeds collected at different times was found that the ability of germination is decreasing proportionally with the time spent in the open field. On the basis of our experiments and of earlier practice in Hungarian tree nurseries, our opinion is that the stratification of seeds is not necessary.

     

  • Effect of boron fertilisation on the flavour of carrots - Applicability of organoleptic analyses to carrots

    Foliar boron fertilisation has had positive influence only on carotene content. Results were evaluated with chemical analyses and sensory tests. It has been observed that sensory tests are able to detect bitter flavour and also to measure its interaction with sweet flavour. Sensory tests for evaluation are generally used in the EU, also in the case of fruits and vegetables. With the aid of students and staff having received training and acquired practice, our University has the possibility to complete chemical analyses in such a way that satisfies modern demands, as well as facilitating the sale of the products on foreign markets.

  • The effect of different berries in human nutrition

    The aims of this study were on the one hand to compare of examined compounds according to their importance in nutrition and human-health, and on the other hand we made preexperiments to investigate the relationships among antioxidant capacity and the endogenous substances which contribute the antioxidant status of the plants. The following species were involved in the experiment: raspberry, blackberry, black currant, elderberry and sour cherry. These fruits have potent health-promoting antioxidant power. Glucose, fructose, total phenol, formaldehyde and anthocyanin contents were determined in addition to ferric reducing ability. Our results reflected considerable differences in the measured parameters of the analysed species. In blackberries and elderberries the high antioxidant capacity is coupled with low carbohydrate content. Besides the formerly proven correlations between total phenol content, anthocyanin concentration and antioxidant capacity, these parameters also correlated with the measured formaldehyde concentrations, hereby we can follow the methylation /demethylation status of the plant.

  • Storability of paprika varieties measured by non-destructive acoustic method

    During our experiments, the storability of paprika (Capsicum annuum) samples was measured by a non-destructive acoustic method. The aims of our work were the determination of the applicability and reproducibility of the acoustic stiffness method for paprika, the investigation of the optimum measuring conditions. In order to compare the main paprika varieties regarding shelf-life, our further aim was to follow the softening phenomenon or textural changes (i.e. changes in stiffness) of different paprika varieties measured by the non­destructive acoustic stiffness method. Five different varieties of paprika grown in hydroponics growing system were used for the measurements. All paprika varieties were stored at 20 °C for two weeks. Samples were tested on every 2nd or 3rd day. The acoustic method was found to be suitable to follow the softening of paprika samples. The characteristic frequency of the acoustic signal could be well detected and clearly separated from the other vibration peaks. Tapping the top of the paprika was observed to give a clearer and less noisy signal compared to the signal obtained by tapping the sample's shoulder. The acoustic results showed the same tendencies with regard to softening during storage as the impact method showed in our previous experiments.

  • Examinations of potential environmental friendly materials against tomato and pepper patogens

    In organic farming systems the focus is on prevention with regards to plant protection. To follow the rules of Good Agricultural Practice one is able to avoid serious yield losses; if it is not possible the use of allowed materials are permitted. Organic farmers have less material to protect their plants so it is necessary to find effective potential materials. Bacterial and fungal diseases of tomato and pepper can cause serious losses in yield. Different materials were tested against some plant pathogen bacterial (Clavibacter michiganensis subsp. michiganensis, Pseudomonas syringae pv. tomato, Xanthomonas campestris pv. vesicatoria and fungal (Phytoptora infestans, Rhizoctonia solani, Sclerotinia sclerotium) strains in order to find potential materials in the field of organic seed treatment. In vitro trials have shown that vinegar, cider vinegar, red wine vinegar, white wine vinegar, cinnamon and thyme oil have inhibiting effect against the causative agent of bacteria and fungi. Germination test has shown that examined vinegar types do not decrease germination ability if the concentration is low, but in higher (more than 5%) concentration it ruins the germination ability. Even in 0,5% concentration of red — and white vine vinegar have good effect on germination capacity.

  • Climate change effect on sour cherry production in Central Hungary

    Consideration of the soil and climatic conditions and their expected changes is very important in planning of new orchards. This research focuses on the expected climatic conditions and opportunities of sour cherry production in Central Hungary in the periods of 2021–2050 and 2071–2100, compared to the base period of 1961–1990. Results show that in the near future the expected changes are mostly favourable for sour cherry production, the utility values are increasing in most of the subregions. By the end of the century the utility values show further increase in the formerly colder subregions. However, in some subregions the utility values slightly decrease because of the appearance of the extreme dry and warm year types, though their values remain at the level of the base period or even higher.

  • Experimental results of the effects of Hungarian climatic conditions to German disease-resistant industrial apple varieties

    In the recent years, several disease-resistant apple varieties appeared through the modern breeding technologies. These varieties can be grown with low usage of pesticides, which mean not only environment friendly fruit growing, but the production costs are also lower. In Eastern-Hungary — it is one of the main apple growing regions — a new apple growing structure started to form by the investment of the German Wink Ltd. — several resistant apple varieties were brought from Germany. 'Resistant' refers genetic resistance that usually transferred from the genome of wild apple species. But the fruit of these apple species is not only resistant to diseases, but its quality is poorer, too. In Germany the Re-apples are grown only for the processing industry. Due to climatic circumstances in Eastern Hungary, the first experiences showed better parameters during laboratory measurement, the fruits have more beautiful view, shape and inner characters than usual industrial apples.

    In our paper we discuss the results of sensory (consumer) tests, carried out in Eastern Hungary and in the Budapest-region the data analysis of systematic storing experiments (refraction, flesh firmness, weight loss, etc.) and profile analysis of fresh and stored Re-apples. (In the profile analysis the ProfiSens software [4,5] has been used.)

  • The necessity and possibilities of irrigation in fruit growing under conditions of Hungary

    Climatic and soil conditions are highly suitable for most temperate fruit species and promise profitable yields with good quality. An accurate choice of the growing site is, however, decisive because of the wide range agro-climatic variation an soils within the country. One of the most important factors is the annual precipitation which does not exceed, in general, 700 mm. The aims of irrigation practices are, succinctly speaking, the improvement of quantity and security of yields and the guarantee of quality. The relative importance of those criteria changes according to the fruit species. In up to date apple, pear and cherry production, micro-irrigation systems are mainly considered. According to recent experiences, the micro-jet type of water distribution should be preferred to the dripping system. In cherries, the choice of the method is motivated by the need to prevent fruit cracking. Most of the peach and apricot plantations are located on the dry and moderately dry regions of the country. Because of the late freezes, the improvement of security is crucial. There the investment of irrigation systems should concentrate to the possibility of anti-freeze sprays. High water requirements of plums are met in Hungary by irrigation where the method should be decided at the plantation and adapted to the harvesting procedure which could be mechanised or (in high density plantations) picked by hand. Sour cherries are perhaps the less dependent on watering under Hungarian conditions. Yields in small fruits: currents, gooseberries, raspberries and strawberries could be increased by irrigation to 40-50 % and may improve quality too. In those cultures the system of moving flexible wing tubes are considered to be the best irrigation technique.

  • The effect of drying process on the pigment content and composition of table beet varieties

    The food industry requires natural colouring agents in increasing quantities. Beet root is highly adapted to this purpose with its red (betacyanin) and yellow (betaxanthin) pigments suitable for numerous products. The two pigments are, however, very heat-sensitive depending not only on the method of drying as well. In our experiment we tested 6 table beet varieties with vacuum drying (instrument: Eurovac KIT-150) and obtained very big differences in the suitability of varieties for drying. The less colour loss was measured in Bordó (16.84%), while Rubin and Cylindra indicated 45.15 and 47.18% loss respectively. During the drying raw material with higher colour content produced higher rate of pigment loss (r = 0.880). In our experiment we found adverse effects on colour material and dry matter content in the fresh beet root. The variety with higher solids (mainly sugar) resulted in higher pigment loss (r = -0.847) during the vacuum drying process.
