  • Selenium enriched vegetables as biofortification alternative for alleviating micronutrient malnutrition

    There is a very difficult equation for malnutrition and over-consumption. That means malnutrition even of vitamins and/or minerals (Ca, Cu, Fe, I, Mg, Se and Zn, vitamin A) affects more than two billion people worldwide, largely due to low concentrations or poor bioavailability of the nutrients in the diet. In some developed countries in contrast, over-consumption, particularly of over-refined cerealbased foods, has contributed to the development of an epidemic of metabolic diseases. So, producing nutritious and safe foods sufficiently and sustainably is important target at the same time challenge of modern agriculture. In the past, great efforts have focused only on increasing crop yields, but enhancing the concentrations of mineral micronutrients has become an urgent task. The main daily food source is the staple crops specially in developing countries of the world, i.e., wheat, rice, cassava, beans, sweet potato or maize. These kind of plants are often deficient in some of mineral elements. Thus, the increasing of bioavailable concentration of micronutrients in edible crop tissues (via biofortification) has become a promising strategy in modern agriculture, providing more nutritious foods, to more people, with the use of fewer lands. Biofortification of these trace elements can be achieved application with agronomic process such as soil or foliar fertilization or crop breeding even conventional technic and/or genetic engineering. This review highlight progress to date and identify challenges faced in delivering biofortified vegetable crops as well as the agronomic approaches and tools to improve crop yield and micronutrient content of food crops.

  • Impact of boron foliar fertilization on annual fluctuation of B in sweet cherry leaves and fruit quality

    The goal of the study was to examine response of sweet cherries (Prunus avium L.) to boron (B) fertilization. The experiment was conducted during 2005-2007 in West Hungary on mature cv. `Germersdorfi 3' grafted on Prunus mahaleb rootstock.

    Sweet cherry trees planted on a calcareous chernozem soil. Trees were foliar-fertilized with B. Foliar B sprays were performed: (1) in the spring, at the stage of white bud, beginning of flowering (B1), and (2) repeated 5 weeks after full bloom (B2). In each of spring spray treatments, B was applied at a rate of 0.15 kg ha-I. Trees untreated with B served as a control.

    The results showed that B fertilization had effect on B concentration in leaf tissues, mostly after ripening. B was present significantly higher amount in leaf in treated samples after ripening.

    Mean fruit weight was slightly increased by B fertilization. Fruit sensitivity to cracking was not influenced by B fertilization. Nevertheless, from our data it can be conclude that the sensitivity of fruit to cracking is improved when the fruit is riper, the fruit density and fruit weight are higher. The soluble solids varied between 15.0 and 15.9% according to the treatments. Our results for the monosaccharides investigated varied between 5.1 and 7.2 as glucose and fructose as well. Galactose and sucrose was detected very small amount in the unprocessed cherries. Applied B treatments increased sugar contents but decreased organic acid contents in sweet cherry fruits.

    It is concluded that under conditions of this experiment, B fertilization can be recommended in sweet cherry culture to improve fruit quality and their appearance.

  • Salt tolerance of sweet pepper seedlings

    Laboratory germination tests have been made with three white fruit pepper varieties and with one spice pepper in filter paper rolls wetted with KCl solutions of different concentration. Parallel tests have been conducted with the other species (lettuce, tomato, kohlrabi) to compare the salt tolerance of paprika with that of other vegetable crops.

    In greenhouse, the action of KCl has been investigated with transplants raised in soil mixture, in rockwool and with seedlings transplanted from rockwool into soil mixture. Like the trials in the laboratory, the experiments in soil mixture have been made with other plant species, too.

  • Fire blight (Erwinia amylovora) susceptibility of old Hungarian apple cultivars

    The aim of the Hungarian apple breeding program started in the Department of Fruit Science was to find resistant apple cultivars against major diseases (scab, powdery mildew, fire blight). The outbreak of fire blight (Erwinia amylovora) in 1996 motivated us to search new resistant sources principally from old traditional apple cultivars. First of all, cultivars have been gathered since 1997 from Carpathia (Visk) and evaluated between 2001 and 2003. In this recent study evaluation of resistance of old Hungarian genotypes to fire blight collected from the English National Fruit Collection (Brogdale, Faversham) is presented.

    13 old Hungarian apple cultivars in 2002, and 38 genotypes in 2005 have been evaluated. We used `Idared' and 'Jonathan M41' as susceptible controls and 'Liberty' and 'Remo' as resistant ones. Shoots of two-year-old potted plants were inoculated with a mixture of virulent E. amylovora isolates (Ea2, Ea60, Ea67) at a concentration of 5 x 108 cells/ml. Resistance of apple cultivars was evaluated weekly, four times after inoculation by disease severity of symptoms. Numbers of bacterial colonies in 1 cm length shoot were determined in the fourth week after infection.

    8 cultivars in 2002 and 9 cultivars in 2005 displayed notable resistance to fire blight based on one-year data. Based on the coincident data of both years, out of the cultivars collected also from Carpathia ‘Pónyik', and `Sikulai' were found to be resistant and gene sources additional old Hungarian valuable apple genotypes could be selected: `Szabadkai szercsika' and `Tordai piros !davit'. The cultivar `Szemes alma' originated from Visk has proved to be recurrently resistant.

    The number of bacterial cells in shoots of the investigated cultivars correlated with the severity of symptoms. With this method, which was introduced by us earlier, we can screen cultivars displaying weak visible symptoms, which cannot be proposed as a source of resistance because of their latent infection.

  • In vitro shoot multiplication of apple: comparative response of three rootstocks to cytokinines and auxin

    Shoot multiplication responses of rootstocks cvs. M.26, MM.106 and JTE-H to different concentration of BA, BAR and IBA in eight various combinations were tested on MS-medium. The effect of hormones depended on genotype, type of cytokinin and interaction of cytokinin and auxin. Shoot multiplication was significantly enhanced with the use of BAR as cytokinin. High multiplication rate could be achieved in cvs. M.26, MM.106 and JTE-H: 7.7, 6.9 and 9.9 shoots per explant, respectively.

  • Effect of variety and cultivation technology on phenols and antioxidant activity of sweet and sour cherry

    The goal of the present work was to compare different sweet and sour cherry cultivars and cultivation methods (bio/integrated) with respect to polyphenol content and antioxidant activity. The concentration of total polyphenols ranged between 880–1050 mg kg-1 of fresh fruit, whereas antioxidant activity expressed as TEAC was found to be between 5.4 and 10.3 mmol kg-1 for the sweet cherry cultivars examined. In case of sour cherry the level of polyphenols ranged between 1283 and 3490 mg/kg fresh edible part of the fruit. Antioxidant activity was recorded between 15–32 mmol kg-1 for the different sour cherry cultivars included in this work. After one-month storage at low temperature, the total phenols and antioxidant activity decreased by 2–40% in the sour cherry cultivars studied. The anthocyanin content in cherry cultivars was less (131–312 mg kg-1) than the135–1893 mg kg-1 found in sour cherries. Anthocyanin level was higher in samples produced under organic farming conditions than in those produced with integrated cultivation.

    The knowledge of the finest composition of the Tokaj Wine Specialities is an indispensable task. The presence of the biogenic amines as an eventual combination of hygiene, technology and activity of Botrytis cinerea is a very important point of view in the future. The objective of this study was to measure the biogenic amine-content of Tokaj Wine Specialities.

    According to our findings, the histamine-concentration remained lower than the prescribed end values, but ill the case of serotonine, there were significant measured values. During the fermentation period, in the first cycle there was a decrease in the concentration of biogenic amines, but with the advancement of fermentation, an increase was found.

  • Physiological and biochemical evolution of peach leaf buds during dormancy course under two contrasted temperature patterns

    Budbreak anomalies in temperate fruit trees grown under mild conditions have often been described. However, only few authors approached the physiological evolution of leaf buds all along the dormancy period according to the temperature pattern. The aim of this study was to characterize the evolution of peach leaf bud dormancy through some physiological and biochemical parameters under temperate winter conditions and under total cold deprivation after the endodormancy onset. Two treatments were applied in peach trees cv. Redhaven: (i) Regular Chilling Amounts — RCA and (ii) Total Chilling Deprivation — TCD. Buds were sampled periodically from different parts of the stem (terminal, medium and basal ones). We recorded the evolution of: carbohydrate concentrations (glucose, fructose, sucrose, sorbitol and starch), respiration rate, water contents and energy metabolism (ATP and ADP ratio). The dynamics of these parameters were compared and correlated with dormancy evolution ("one node cuttings" test) and budbreak patterns in plank:. The endodormancy intensity of terminal buds was significantly lower than those of median and basal buds in early October. Under RCA treatment, this gradient faded and the bud endodormancy release was completed at the same time in all positions along the stem. Thereafter, the "cuttings" test indicated that terminal buds grew slightly faster than median and basal buds, and, consistently, budbreak in planta started with the terminals buds, followed by the medians and then by the basal ones. The carbohydrate contents showed a transitory change only when the buds began to grow after the endodormancy was released under RCA. Respiration, water content and ATP/ADP changed dynamics only under RCA and only after the end of the endodormancy (their respective changes were very parallel). The dynamics of none of the tested parameters could be related with the endodormancy dynamics, but respiration, water content and ATP/ADP could be consistent markers of the actual bud growth before bud break (in this respect, ATP/ADP could not show differences between the terminal and axillary buds while respiration and water content could).

  • High-velocity microprojectile mediated DNA delivery into Phaseolus vulgaris callus cells

    We report the method for the establishment of rapidly growing callus cultures of Phaseolus vulgaris and the conditions required for efficient transformation using high velocity microprojectiles and high level of transient gene expression. Using hypocotyl explant and vertical culture on B5 medium with lmg/1 kinetin and 2 mg/1 2,4-D, we can recommend to get a rapidly growing callus from bean which is a good starting material to introduce foreign DNA into bean cells. The GeneBooster particle delivery system was used for the bombardment of bean callus and the Hgm resistance gene (Hgmr) was used as a selectable marker gene. 25mg/I hygromycin (Hgm) concentration was sufficient to kill the control callus. We used the standard physical factors, the appropriate pressure of N2 gas for the bombardment of the callus tissue, the shooting distance and the size of tungsten particles used as microprojectiles. Selective and nonselective tests were made by transferring the healthy green and white calluses, subcultured for 4 months on selective and nonselective medium. Several Hgm resistant calli had been obtained. Selective pressure was maintained over a period of 10 months.

  • Ascospore dispersal of Venturia inaequalis and subsequent development of scab symptoms in a Hungarian organic apple orchard

    In this study, we aimed to study ascospore dispersal of Venturia inaequalis and subsequent disease development in an organic apple orchard (Eperjeske) in 2012 and 2013 on apple cultiva ’Mutsu). Burkard spore trap in March and April were used to monitior ascospore concentration and number of scab symptoms were assesed 20 May in both years. Three peaks were detected in ascospore dispersal in the period of examination which was clearly related to the Mills infection periods. On the basis of the incubation period’s length in April (15–18 days), the appearance of first symptoms had direct connection with the peak of the ascospore discharge. The largest number of symptoms were observed on those parts of the orchards where where the inoculum sources were accumulated.

  • The effect of different berries in human nutrition

    The aims of this study were on the one hand to compare of examined compounds according to their importance in nutrition and human-health, and on the other hand we made preexperiments to investigate the relationships among antioxidant capacity and the endogenous substances which contribute the antioxidant status of the plants. The following species were involved in the experiment: raspberry, blackberry, black currant, elderberry and sour cherry. These fruits have potent health-promoting antioxidant power. Glucose, fructose, total phenol, formaldehyde and anthocyanin contents were determined in addition to ferric reducing ability. Our results reflected considerable differences in the measured parameters of the analysed species. In blackberries and elderberries the high antioxidant capacity is coupled with low carbohydrate content. Besides the formerly proven correlations between total phenol content, anthocyanin concentration and antioxidant capacity, these parameters also correlated with the measured formaldehyde concentrations, hereby we can follow the methylation /demethylation status of the plant.

  • Spore dispersal, diurnal pattern and viability of Monilinia spp. conidia and the relationship with weather components in an organic apple orchard

    In a two-year Hungarian study, spore dispersal diurnal periodicity and viability of Monilinia spp. and their relation to weather components were determined in an organic apple orchard. Conidia of Monilinia spp. were first trapped in late May in both years. Low number of conidia were trapped until end-June. Thereafter, number of conidia continuously increased until harvest. Conidia in a 24-h period showed diurnal periodicity pattern, with th highest concentration in the afternoon hours. Spore viability with FDA staining showed that viability of
    conidia ranged from 45 to 70% with showing lower viability in the dry than in the wet days in both years. Temperature and relative humidity correlated positively with mean hourly conidia numbers in both years. Mean hourly rainfall was negatively but poorly correlated with conidiacatches in both years. Results were compared and discussed with previous observations.

    Selenium (Se) is an essential trace element for animals, microorganisms and some other Eukaryotes. It has become increasingly evident that Se plays a significant role in reducing the incidence of lung, colorectal and prostate cancer in humans. Although it is well known that some species among higher plants are able to accumulate selenium in their tissues, but others are not able to do so, and there is evidence that selenium is needed for the growth of algae, meanwhile the question of essentiality of Se in vascular plants is unresolved. We aimed to study the in vitro growing and to characterise some physiological properties in radish (Raphanus sativus L.) seedlings treated with 0 to 200 mg/1 sodium-selenate. The results showed that lower (2 mg/1) concentration sodium-selenate increased the biomass as well as the total antioxidant capacity of seedlings. The seedling's selenium content showed linear correlation with the sodium-selenate content of the medium.

  • Dynamics of the uptake of nutrient elements from the medium of in vitro cultured apple rootstocks

    Cation uptake of J-TE-F apple rootstocks propagated in vitro was studied by the analysis of culture medium. Test-plants were grown on liquid medium under different light regimes. Samples for tests were taken twice a week.

    Media without plants served as controls. The analysis of those showed, that only the uptakeable iron-content changed depending upon light treatment. The concentration of all other cations was considered unaltered.

    As a result of analysis, it could be established, that elements present in the media were taken up in different rates by plantlets: Cu, P and Zn were utilized totally, but only 50% of K and 20 to 40% of Ca and Mg were taken up under the light treatments applied.

    The dynamics of the uptake process was also observed. It was registered that they differed in the case of some cations. So Ba was utilized at the beginning of subculture, others for example B in the later phases. Some elements disappeared unevenly so K, P but the whole quantity is taken up during subculture.

  • First evaluation of fruit quality of some new and old sour cherry cultivars in Hungarian climatic condition

    Indeed, in the last decade a large number of new sour cherry cultivars with new notable flavor characteristics have been released. There is no enough information related to qualitative characteristics which consistently segregated based on their predominant sensory characteristics. The present study carried out at tow consecutive years (2008 and 2009) and examined some qualitative characters which are the main importance in the specialty cherries trade, The main objective was to determine fruits quality of Érdi bőtermő, Oblacsinszka, Debreceni bőtermő, Csengődi, Pándy 279, Éva, Kántorjánosi, Újfehértói fürtös and Petri sour cherry cultivars. Changes observed in major characteristics such as fruit detachment force, width, length, weight, peduncle length, flesh firmness, brix, citric acid, Suger/acid ratio and pit weigh.While observed no change in flesh/pit ratio and Coordinate of colour (X*, y*, x*), hue angle and chroma value, in different years. In all measured factors, there was significant difference between selected cultivars of sour cherry fruit. Debreceni bőtermő showed higher values of fruit detachment force in 2008, while had the lover of fruit detachment force in 2009. Fruit firmness of “Érdi bőtermő” were monitored at lover value in both 2-year.

  • Comparative analysis of Hungarian Matricaria recutita (L.) Rausch. populations

    Matricaria recutita L. is a traditional medicinal plant in Hungary and its drug is known as „Hungaricum", world-wide. Plant samples and seeds were collected from 12 different habitats of three significant geographical regions of Hungary in 2001. Morphological, production biological and chemical properties of samples were examined. In relation to the morphological characteristics, a negative correlation (r= -0.75) could be observed between the average height of the plants (height of flowering shoots) and the pH value of the soil. According to the composition of the essential oil, the populations accumulating typically chamasulene (10-20%), a-bisabolol (30-50%) or bisabolol-oxid (30-50%) could be completely distinguished. Concerning the flavonoid composition the quantity of apigenin-7-glucoside was outstanding in the populations originating from the Great Hungarian Plain, it has reached the concentration of 1.8-2.8 mg/g, while the samples collected in Transdanubia could be characterised by much lower level of apigenin-7-glucoside (around 1.5 mg/g).

  • Effects of nitrogen fertilizer application rate and time to first harvest on leaf area index; leaf nitrate content and yield of irrigated leaf rape (Brassica napus L. var. Giant)

    In Zimbabwe, smallholder cultivation of leaf rape is of considerable importance due to the increasing demand for the leaf vegetable from the rapidly increasing urban populations.  A two-season field trial was conducted in the Agricultural Practice Experimental Plots of the Midlands State University in central Zimbabwe. The objective of the study was to establish selected biometric responses of leaf rape to N fertilizer rates and time to first harvesting. Increasing the rate of N fertilizer application considerably improves the yield of fresh rape leaf; LAI and concentration of leaf nitrate. Delaying rape leaf harvest after N fertilizer side dressing can be used as a strategy to boost rape leaf yield for vegetable markets with high single delivery demand. When harvesting is deliberately delayed after N fertilizer application rape LAI are significantly increased. Delaying the first leaf harvests by 7; 14 and 21 days after N fertilizer side dressing amendments increases the content of nitrate in fresh rape leaves. Fresh rape yield can be more accurately predicted by values of rape LAI. Nitrate concentrations in fresh rape leaf is a poor predictor of rape yield under field conditions.

  • Role of wholesale markets in fresh fruit and vegetable distribution in Hungary

    The primer function of fruit and vegetable wholesale markets — which was the physical concentration of supply and demand previously — is changing gradually as consumer habits change and logistics functions (e.g. product manipulation, redistribution without the physical presence of the product) become more important with the technical development. Mainly individual growers sell their product to retailers at the Hungarian regional producers' markets and the Budapest Wholesale market on NagykOrosi Street, which is the largest wholesale market in Hungary regarding area, produce arrival and turnover. Parallel to the establishment of Producers' Organisations (P0) their market presence should be urged in selling their produce and also in ownership — and decision-making procedure — of the markets. The produce auction system and the on-line sales experiences of New Jersey State (USA) can contribute to the investigation of development possibilities and realization of development of Hungarian wholesale markets.

     

  • Allelopathic effect of the Cladonia verticillaris lichen extracts and fumarprotocetraric acid on the early growth of germinating seeds and seedlings in Allium cepa L.

    The allelopathic activity of the different extracts of the lichen Cladonia verticillaris and fumarprotocetraric acid on the early growth of A cepa (IPA 6) seedlings depends on their chemical composition and concentration, respectively. It was observed that the length of the radicle was significantly stimulated by fumarprotocetraric acid at high concentrations and by the total extract of C. verticillaris thalli, which contained a high level of fumarprotocetraric, acid confirmed by HPLC-technique. In addition, it was found, that the phosphate buffer extract, which contained high level of methyl (3-orcinol carboxilate measured by HPLC, reduced the length of the hypocotyls significantly. Under our experimental conditions there was no influence of the different types of extract and fumarprotocetraric acid on the ratio of seed germination, in relation to the control. From the study of HPLC it was found that fumarprotocetraric acid and methyl 13-orcinol carboxilate were present in all extracts at different concentrations, depending on the method of extraction.

    In our work we have studied red wines of some vine-districts. In the centre of interest was resveratrol. We declared, there was not significant difference between varietys in polyphenol comparison. There was significant variety in anthocyanin and colour-intensity.

  • Nutrient demand of stone fruits

    Effects of nitrogen, phosphorus and potassium fertilization were investigated on the change of nutrient content, vegetative and generative production of apricot, peach and sour cherry trees, as well as on frost hardiness in long term experiments. Nitrogen and phosphorus fertilization increased only the concentration of these elements in cherry leaves without effect on growth and yield. Consequent potassium effect was proved on these stone fruit species. Effect on yield appeared following the first higher crop load.

    Potassium supply has positive effect on frost hardiness of apricot and sour cherry flowers and peach flower buds.

    In peach, the lime content of soil decreased the yield but it could be compensated by potassium dressing to some extent. Favourable nutrient boundary values were determined for soil and foliage.

  • Floral biological investigations of apple cultivars in relation to fire blight

    Floral activity was studied in two apple cultivars: an Erwinia-tolerant (Treedorn') and a sensitive one (`Sampion'). Since more types of protogyny occur in apples, the period of stigma activity is different. Papillae of the exposed stigma in flowers of 'Freedom' function longer (usually more than a week) than in the delayed homogamous `Sampion'. Despite of this, cv. 'Freedom' is tolerant to Erwinia amylovora (Burr.) Winslow et al., suggesting no relationship between the floral biological type (including the exposure and longevity of stigma) and the infection by E. amylovora. According to SEM micrographs, nectary stomata in `Freedom' are already open in the flower bud, where nectar secretion starts and continues until the senescence of the stigma. However, the long period of nectar secretion does not create optimal conditions for bacterial growth, since nectar production is scant in the flowers of 'Freedom'. The surface of the nectary, its nectar-retaining capacity, and the amount and concentration of nectar may influence the susceptibility of apple cultivars. It is manifested well by the smooth nectary surface with nectary stomata rising slightly above the epidermis in flowers of cv. 'Freedom', contrasting the wrinkled, striate nectary surface with slightly sunken stomata in the flowers of 'Sampion'.

  • Development of in vitro propagation system for Atriplex halimus L.

    Explants excised from adult shrubs were surface sterilized and cultured on Murashige and Skoog (MS) basal medium in the presence of plant growth regulators (PGRs) at different concentrations. A high multiplication rate of 7.2-fold was achieved every four weeks on MS medium supplemented with 4.44 μM BA, 0.49 μM IBA and 0.58 μM GA3. Rooting was achieved with 73% efficiency within 2-4 weeks on agar-gelled MS basal medium free of PGRs. Rooted plantlets were gradually acclimatized to field conditions over 5-6 weeks with 65% efficiency. For in vitro selection for salt tolerance, MS medium was supplemented with increasing concentrations of NaCl ranging between 25 and 1000 mM. This study has demonstrated that in vitro shoots could tolerate up to 600 mM NaCl with optimal growth at 200 mM, while higher concentrations of NaCl affected growth negatively. Growth and shoot number decreased with increasing NaCl concentration with all plantlets died at 1000 mM NaCl.

  • Attempting Regeneration from Cultured Cotyledons and Plant Regeneration from Cotyledonary Nodes in Common Bean (Phaseolus vulgaris L.)

    Dry seeds from two cultivars of common bean (Phaseolus vulgaris L.) were germinated on sterile cotton and sterile deionized distilled water. Cotyledonary node tissue of seedlings were cultured on Murashige and Skoog(MS)-based media supplemented with different combination of N6-benzyl-aminopurine (BAP) and indole-3-acetic acid (IAA), and benzyladenine (BA) and a-naphthaleneacetic acid (NAA). The results revealed that the regeneration percent and the average number of buds and shoots per explant were influenced by the type of explants and exogeneously added hormones. Multiple shoot induction on dry bean cotyledonary node that contain 4-5 mm from cotyledons and hypocotyl on a medium containing full concentration of MS inorganic salts supplemented with 0.5mg/1 BA and 0.1mg/1 NAA was feasible and the method can be applied in transformation experiments.

     

  • Analysis of amino nitrogen content affecting fermentation and wine quality

    A suitable amount of nitrogen source is needed for the optimal process of alcoholic fermentation. The professional literature mentions the complexity of nitrogen compounds exploitable for yeast-plants as an assimilable or immediately assumable nitrogen source. Former experiments proved that in case of low nitrogen concentration , yeast-plants produced a bigger quantity of hydrogen sulphide. According to the findings above, it is not indifferent when and how we complement the nitrogen content of the must. The aim of our present work was to elaborate such a quick, easily applicable method that can be used for routinish measurements in the viticultural practice.
