  • Determination of the cold tolerance off sour cherry cultivars with frost treatments in climatic chamber

    Nowadays, sour cherry buds can be seriously damaged by spring and winter frosts. Unlike other fruit species threatened by high frost damage, sour cherry cultivars have not been assessed for frost tolerance. The aim of .our survey was to establish the relative cold tolerance of the Hungarian cultivars after treatment in a climatic chamber, and to optimize the methodology formerly elaborated for the frost treatment of apricot. Fourteen cultivars of Hungarian sour cherry (Prunus cerasus) were used in the experiments, which spanned the winters of 2005/2006 and 2006/2007. Our data were used to rank cultivars in two groups according to their levels of cold resistance. We also recommend critical temperatures and treatment times for the testing of sour cherry cultivar resistance to cold in climatic chambers.

  • Transmethylation and the general defense reaction of plants

    Plant breeding for resistance, namely building specific resistance genes into cultivated plants to ensure resistance against certain pathogen species, is a several-decade-long practice. While looking for purposes of failures appearing during the cultivation of varieties created in this way, a plant feature that ensures non-specific reactions against effects which evoke biotic stress attracted our attention. We named this plant defense form the general defense reaction. The general defense reaction is a fundamental attribute of the plant kingdom, fulfils the role of plant immune system and manifests itself in cell enlargement and cell division. Plants with a high level general defense reaction endure abiotic stresses as well.

    In studying the biochemical background of the interaction of the general defense reaction and transmethylation, we found that transmethylation has important role in warding off both biotic and abiotic stresses. According to our observations, plants possessing high level general defense system are suitable for thorough examination of the process and plant physiological role of transmethylation. Biochemical studies also strengthened our observation, which has been taken on the basis of phenotype, that the general defense system can not be ignored during future plant breeding.

    The goal of plant breeders is to improve the resistance of crops against virus, bacterium and fungus pathogens was easiest to achieve by selection for phenotypes displaying the hypersensitive reaction. The resistant plant of that type keeps its health by preventing or delaying the systemization of the pathogen by destruction of cells and tissues of variable size or amputation of the contaminated organs. The faster the reaction of the host plant is the more efficient and economical is the defense, since the extent of tissue destruction decreases proportionally with the speed of reaction.

    During a breeding program for resistance carried out on several plant species, mainly vegetables over thirty years, also an alternative defense reaction has been experienced, which fundamentally differs from the hypersensitive reaction. In that reaction the cells and tissues of the host plant being exposed to the pathogen do not die, on the contrary they hinder systemization of the pathogen by tissue thickening. An additional significant difference is that on the contrary to hypersensitive reaction this reaction is less host- or pathogen-specific and works excellently even at high temperature (over 40 °C).

  • Effect of Erwinia amylovora infection on peroxidase enzyme activity in resistant apple cultivars

    Two apple cultivars that display enhanced resistance to fire blight (causal agent: Erwinia amylovora) were selected. The aim of the present study was to characterize the peroxidase (POD) enzyme activity of `Szemes alma' (a historical cultivar) and MR-03, (a Hungarian multiresistant hybrid of 'Prima') and compare them to susceptible 'Jonathan M 40' and resistant 'Remo' controls. Peroxides enzyme activity during E. amylovora infections was investigated in artificially infected apple shoots. Increases in enzyme activities were observed in a `Jonathan M40' and in 'Remo', MR-03, `Szemes alma' cultivars. There was a consistent relationship between total enzyme activity and fire blight disease severity. High activity of the peroxidase was positively correlated with the degree of resistance to fire blight. A general hypothesis that POD activity is related to fire blight susceptibility/resistance is supported by our results.

  • Co-transformation of bean callus using high-velocity microprojectiles- mediated DNA transfer

    We have found that 50 mg/I kanamycin and 0.8 Mo1/1 mannitol concentration was sufficient to kill the control callus of bean (Phaseolus vulgaris L.) and differentiate transgenic from the non-transgenic cells. The GeneBooster particle delivery system was used for the bombardment of bean callus. The kanamycin resistance gene was used as a selectable marker. The test was made by transferring the healthy white callus, subcultured for three months on selective and non-selective medium. After selection on kanamycin containing media, several kanamycin resistant calli had been obtained, survived and grew. After selection on mannitol containing media no drought resistant calli had been obtained. Resistance of the selected calli were verified by their ability to grow repeatedly on selective medium containing 150 mg/I kanamycin. Selective pressure was maintained over a period of 8 months.

    Eggplants transformed with Sw-5 gene, regenerated by organogenesis and somatic embryogenesis, were resistant to the Tomato chlorotic spot virus, while wild plants did present systemic infection. TO plants were selfed and the segregation analysis of T1 and T2 generation indicated the existence of one or more insertion sites. Southern blot analysis confirmed one or two independent insertions in T2 plants. Different lesions associated with the insertion number were observed in TI and T2 plants. T2 plants with two copies displayed faster hypersensitive reactions and characteristic necrotic lesions that contrasted with slower responses and necrotic ring lesions in plants with one copy. These results suggest that the Sw-5 confers resistance to tospovirus in transgenic eggplants and that the resistant phenotype depends on the number of transgene copies.

  • Comparison of Xanthomonas arboricola pv. juglandis isolates from walnut trees grown in Romania and Hungary

    Among diseases that affect walnut, bacterial blight is considered the most important one in all walnut growing areas both from Hungary and Romania. For the determination of susceptibility/resistance of walnut cultivars in our posterior work planned, 61 bacterial isolates were collected from walnuts showing symptoms of blight, with the purpose of isolating Xanthomonas arboricola pv. juglandis (Xaj). The characteristic Xaj colonies, frequently present as saprophytes in the infected plant tissues, were separated from other bacteria according to their morphology, yellow colour, hydrolysis of starch, and oxidation of glucose. All isolates were tested for pathogenicity by hypersensitive reaction on tobacco leaves (Nicotiana tabacum L.), bean pods (Phaseolus vulgaris L) and unripe nuts (Juglans regia L.). Determination of taxonomy of the selected isolates denotes a possible subdivision (races, biotypes) of Xaj occurring in different geographical areas, API 20NE and API 50CH kits were used. Hungarian and Romanian strains showed a high degree of similarity of carbohydrate utilization but slightly differed from the type strain. All were Cu-sensitive.

  • Experiments on the Resistance or Pepper Cultivars to Macrophomina phaseolina

    The ashy stern blight of pepper (Capsicum annuum) is often caused by Macrophomina phaseolina. Serious wilt disease occurred between 1994 and 1996 of pepper plants in Hungary. In 1996-98 screening experiments were made on many pepper cultivars. Culture (Knopp) solution experiments, pot experiments, greenhouse and field trials were carried out. We determined the incidence of disease by visual examinations, testing on PDA culture, and light microscope. There were significant differences in susceptibility of cultivars and breeding materials.

     

  • Fire blight (Erwinia amylovora) resistance in apple varieties associated with molecular markers

    The invasive bacterial disease fire blight, caused by Erwinia amylovora has the potential to destroy fruit tree orchards all over Europe. Effective plant protection methods are lacking in many countries, highlighting the increasing importance placed on identification of germplasm with heritable disease resistance. Recent l y. a promising QTL (quantitative trait locus) was identified on linkage group 7 in the apple cultivar 'Fiesta· which is derived from ·cox's Orange Pippin' . I n the present study, 144 Swedish and foreign apple cultivars were analysed with the SCAR markers AE I 0-375 and GE-8019. which flank-. this QTL. Twenty-nine of the analysed cultivars had both markers. 78 had either AE I 0-375 or GE-8019, and 37 cultivars did not carry an) of the two markers. Seventeen cultivars. 7 with both markers and I 0 not having either of the two markers, were then inocoluted with the bacterium in a 4uaran1i ne greenhouse test. Cultivars carrying both DNA markers were significantly less susceptible than cultivars lacking the markers, P<0.001. Cultivars that were most resistant had both markers and had 'Cox· in their pedigree. Unrelated cultivars with the markers may still lack the QTL.

    In addition to successes achieved in certain varieties in resistance breeding based on a defense reaction of host plants involving hypersensitive tissue destruction, resistant varieties putting a very strong selection pressure on pathogens have selected more and more aggressive types of pathogens. The never-ending race between plant and pathogen resulting from this can only be controlled by a defense system characterised by a different strategy. In each of the plant species that we bred a defence system was found, which contrary to hypersensitive reaction strives to keep the tissues at all costs and is not pathogen specific. This is implied in the term general defense system.

     

  • Six promising selections from the Hungarian apple breeding program for multiple resistance

    An apple breeding program has been carried out at the Department of Fruit Science for more than a decade. Several apple selections have been released from the progenies of crosses in 1992 and 1993. Six candidates were submitted for national recognition out of the hybrids examined for more than a decade. The six selections are resistant against all the three most important apple diseases (apple scab, powdery mildew and fire blight). Scab resistance is controlled by the Vf gene originating from the species Malus floribunda 821 and transmitted by cultivar Prima. Heterozygote Vfvf genotype of the six cultivar candidates was proved by molecular genetic examinations of Dept. Genetics and Plant Breeding. Characteristics of these selections from 'Prima' progenies are shown on the base of our own observations.

  • Apple powdery mildew caused by Podosphaera leucotricha: some important features of biology and epidemiology

    In this review, some important features of biology and epidemiology are summarised for apple powdery mildew (Podosphaera leucotricha). In the first part of the review, the geographical distribution or the pathogen are discussed, then the morphology and taxonomy of the causal organism are described. Disease symptoms or apple powdery mildew are also shown and then host susceptibility/resistance is discussed in relation to durability of resistance. In the second part of this review, the general disease cycle of powdery mildew on apple are demonstrated and some basic features of powdery mildew epidemiology (such response of the pathogen to temperature, relative humidity, and rain as well as spore production, spore dispersal, diurnal patterns and temporal dynamics of the pathogen/disease) are also given on apple host.

  • The effect of Regalis® (prohexadione calcium) on the reduction of fire blight (Erwinia amylovora) severity in apple trees

    Due to the lack of effective and non-phytotoxic materials for control of the blossom and shoot blight phase of fire blight in pome fruit trees, two novel control strategies have emerged: shoot growth retardation by bioregulators and applying resistance inducer compounds. Prohexadione calcium (ProCa) is the active ingredient of the bioregulator Regalis® registered in several European countries. The reduction of shoot growth elongation is the most obvious effect of ProCa. Furthermore, it causes significant changes in the spectrum of flavonoids and their phenolic precursors, leading to the considerable reduction of susceptibility to fire blight.

    In Poland, potted one-year-old apple trees of cvs. Gala Must grafted on M.26 and Sampion on M.9 (in 2001) as well as Gala Must on P.60 (in 2002) were treated with Regalis® at a range of concentration of 250, 150 or 150 + 100 ppm, respectively. The inoculation of shoots was made with the strain No.691 of E. amylovora (107 cfu/ml), on the 7th and 21st'day after treatments with Regalis. In Hungary, during the years of 2002 and 2003 one-year-old container grown apple trees of the cvs. Idared/M.9 and Freedom/M.9 were treated with the prohexadione-Ca, the active ingredient of Regalis® 100, 150 or 200 ppm, two weeks before inoculation with the Ea 1 strain of E. amylovora (107 cfu/m1).

    In Poland, the suppression of fire blight in shoots reached up to 80%, depending on concentration and application time of Regalis®. In Hungary, the effect of prohexadione-Ca treatments, determined by the length of necrotic lesion developed, proved to be better than that of streptomycin used for comparison.

  • The examination of the broadleaf evergreen ornamental shrub species in Keszthely

    The examination of production possibilities of woody ornamental shrubs (originating from warmer regions than our country) commenced with the financial support of the INTERREG IIIA Slovenia – Hungary – Croatia programme at the Department of Horticulture of the University of Pannonia’s Georgikon Faculty in spring 2007. Our examinations focused on the appearance of species in our country as well as on their frost resistance abilities. The species participating in the experiment are as follows: Abelia grandiflora, Aucuba japonica ’Rozzanie’, Calycanthus floridus, Cotoneaster franchettii, Elaeagnus pungens ’MaculataAurea’, Ilex cornuta, Ligustrum sinense, Ligustrum texanum, Nandina domestica, Osmanthus heterophyllus, Perovskia atriplicifolia, Phyllyrea angustifolia, Photinia fraseri ’Red Robin’, Prunus lusitanica, Sarcococca hoockeriana, Spartium junceum, Teucrium fruticans, Viburnum cinnamonifolium, Viburnum tinus.

  • Some biological properties of new sweet cherry cultivars in Bulgaria and their susceptibility to Blumeriella jaapii

    Investigations were made on 12 sweet cherry cultivars (‘13-S-22-8’, 'Sunburst', `Kozerska'., 'NY 13791', 'Royalton', 'NY 13688', 'Hartland', `Sumerset', 'Pollax', 'Patriotka Krima', 'Castor', and `Lapins') in an experimental orchard of cherry cultivar collection in the Institute of Agriculture at Kyustendil, Bulgaria during the period of 1997-2003. The trees were planted in 1996. All cultivars were grafted on Prunus mahaleb. Four biological properties of the cultivars were assessed such as blooming time, resistance to late spring frost, fruit ripening and fruit mass. Among cultivars, a good resistance to late spring frost was observed on cultivar 'Sunburst'. The fruit mass varied between 4.2 g (cv. 'Patriotka Krima') and 8.5 g (cv. `Sunburst'). The susceptibility of the cherry cultivars to cherry leaf spot caused by Blumeriella jaapii was assessed in mid-September in each year. The estimation of the rate of attack was made according to the grade of Townsend and Neuberger. All cultivars showed symptoms of cherry leaf spot but the degree of susceptibility was different. Cultivar `Patriotka Krima' was the least susceptible, while cultivar `Somerset' was the most susceptible to Blumeriella jaapii.

    The effect of salicylic acid on reducing brown rot caused by Monilinia laxa infection in postharvest apricot fruit Prunus armeniaca L. Freshly harvest Jumpo Cot fruit were treated with water as control treatment and other group treated with salicylic acid 2 Mm as induced resistance treatment, Fruits were infected by M. laxa (1 × 10-3spores ml-1) and incubated at 25 oC for 2, 4 and 6 days. Treatment fruits with salicylic acid resulted in direct effect on mycelial growth as in the salicylic acid treatment the growth reached to 45% after 6 days of incubation while in the control treatment it reached to 100%.

  • Anatomical study of the leaves and petioles of scab resistant and susceptible apple cultivars

    Anatomic studies have been performed on the leaf blade, petiole and annual shoot on six apple cultivars by means of scanning electron as well as light microscope. Four of the cultivars examined are resistant to scab (Florina, Freedom, MR-10, MR-11), whereas two of them are susceptible (Jonathan and Idared). Preliminary results suggest that differences in the width of cross sections of leaf blades, in hairyness, in the shape and size of epidermal cells, moreover, in the cross sections of petioles and shoots are considerable. Some of the anatomical properties seem to be correlated with scab resistance or susceptility of the respective cultivars. Therefore, further studies extending to other cultivars may corroborate our claims to find causal relations between anatomical traits of the leaves and disease, especially scab resistance of apple cultivars.

  • Susceptibility of European pear genotypes in a gene bank to pear psylla damage and possible exploitation of resistant varieties in organic farming

    We evaluated 285 pear genotypes (commercial cultivars, ancient local varieties, unnamed local strains, seedlings, wild seedlings) in the largest gene bank of pear in Hungary from the point of view of psylla resistance to explore their possible exploitation in organic farming. We have found some 10 new resistant types (Bókoló körte, Bôtermô Kálmán, Füge alakú körte, Nagyasszony körte, Nyári Kálmán, Rozs nyári körte, Viki körte, Pb-242, Pb-299, 0-632) and 7 highly tolerant ones (Cure-6, Kései Kálmán, Kieffer, Kieffer Éd, Steiner, Téli Kálmán, II. B-3- 6/4, 96-16/5) (Table 1). These made up 3.5 + 2.8 per cent of the investigated genotypes, while 93.7 per cent of them were susceptible to pear psylla damage. Taking earlier and present results into account we can list more than 30 European pear cultivars being resistant or highly tolerant to pear psylla infestation and damage. In fact, the list of resistant and highly tolerant cultivars may serve as a basis selecting pear cultivars fitting to the specific requirements of the organic farming. By the end we can conclude that there is some real hope to exploit some resistant or highly tolerant ancient or local cultivars in organic farming but further investigations are needed to estimate their yield capacity and fruit quality.

    Powdery mildew, Phylloxera and downy mildew epidemics devastated large areas of vineyards in Hungary in the 1800s. To establish new plantages resistant so called "direct producers" were imported from America (direct means "without grafting"). Direct producers have secure yield but their quality is inferior. After World War II, an organized breeding programme was started to replace them and parallel to this the law prohibited their production. As a resistance gene source, the E.2 clone (S. V. 12375 E.2) of Seyve-Villard 12375, Franco-American hybrid selected in Eger was used by Hungarian breeders. Several valuable hybrid families have originated from crosses of this variety hybrid.

    In the Research Institute for Viticulture and Enology at Kecskemét, two valuable table grape hybrids were registered under the names of R.65=Esther and R.78=Fanny coming from 2 hybrid families of the parent S. V. 12375 E.2. Both varieties may be cultivated "environmentally".

  • Reactions of some cucumber (Cucumis sativus L.) lines and hybrids to zucchini yellow mosaic virus (ZYMV) and selection of tolerant breeding lines

    In the past years zucchini yellow mosaic virus (ZYMV) has been appeared as a new pathogen of cucurbitaceous plants in Hungary. It caused severe disease outbreaks on the fields of pickling cucumber hybrids which are highly tolerant to cucumber mosaic virus (CMV). Preliminary inoculation tests have showed that all of the important pickling cucumber hybrids produced in Hungary were susceptible to ZYMV-P. However, a selection of Chinese Long and the breeding line I- KS 10C proved to be highly tolerant. The Fl plants of the crosses between the tolerant lines and the susceptible genotype H I were susceptible to ZYMV-P. F2 populations segregated for susceptible and tolerant individuals at a ratio of 3:1. The results strongly suggest that the resistance to ZYMV in our sources is controlled by a single recessive gene. Inoculations of the ZYMV and CMV susceptible cultivar Budai csemege and the CMV tolerant hybrid Perez Fl with the complex of ZYMV+CMV resulted extremely severe symptoms (strong mosaic and necrotic spotting of the leaves) on both cultivars. On the Chinese Long line, which is tolerant to ZYMV and CMV, respectively, the complex of the two viruses caused mild symptoms. The results show a synergistic pathological effect of ZYMV and CMV on differrent cucumber genotypes. More detailed studies on the interactions among the plant genotypes, viruses and virus strains are needed to develop cucumber hybrids that are highly resistant to the ZYMV+CMV complex.

  • Brown rot blossom blight of pome and stone fruits: symptom, disease cycle, host resistance, and biological control

    In this paper, important features of symptoms, biology and biological disease management are summarised for brown rot blossom blight fungi of pome and stone fruit crops (Monilinia laxa, Monilinia fructicola and Monilinia mali). Firstly, European brown rot caused by Monilinia laxa is discussed highlighting the blossom epidemiology features, then host susceptibility of the most important stone fruit species including several Hungarian and international cultivars. At the end of this chapter, recent biological control possibilities against Monilinia laxa are also included. Secondly, American brown rot caused by Monilinia fructicola is discussed. Symptoms, biological features of blossom blight and host susceptibility of flowers to Monilinia fructicola are demonstrated. Finally, the symptoms and the biology of the least frequent species, Monilinia mali are shown.

  • Experimental results of the effects of Hungarian climatic conditions to German disease-resistant industrial apple varieties

    In the recent years, several disease-resistant apple varieties appeared through the modern breeding technologies. These varieties can be grown with low usage of pesticides, which mean not only environment friendly fruit growing, but the production costs are also lower. In Eastern-Hungary — it is one of the main apple growing regions — a new apple growing structure started to form by the investment of the German Wink Ltd. — several resistant apple varieties were brought from Germany. 'Resistant' refers genetic resistance that usually transferred from the genome of wild apple species. But the fruit of these apple species is not only resistant to diseases, but its quality is poorer, too. In Germany the Re-apples are grown only for the processing industry. Due to climatic circumstances in Eastern Hungary, the first experiences showed better parameters during laboratory measurement, the fruits have more beautiful view, shape and inner characters than usual industrial apples.

    In our paper we discuss the results of sensory (consumer) tests, carried out in Eastern Hungary and in the Budapest-region the data analysis of systematic storing experiments (refraction, flesh firmness, weight loss, etc.) and profile analysis of fresh and stored Re-apples. (In the profile analysis the ProfiSens software [4,5] has been used.)

  • High-velocity microprojectile mediated DNA delivery into Phaseolus vulgaris callus cells

    We report the method for the establishment of rapidly growing callus cultures of Phaseolus vulgaris and the conditions required for efficient transformation using high velocity microprojectiles and high level of transient gene expression. Using hypocotyl explant and vertical culture on B5 medium with lmg/1 kinetin and 2 mg/1 2,4-D, we can recommend to get a rapidly growing callus from bean which is a good starting material to introduce foreign DNA into bean cells. The GeneBooster particle delivery system was used for the bombardment of bean callus and the Hgm resistance gene (Hgmr) was used as a selectable marker gene. 25mg/I hygromycin (Hgm) concentration was sufficient to kill the control callus. We used the standard physical factors, the appropriate pressure of N2 gas for the bombardment of the callus tissue, the shooting distance and the size of tungsten particles used as microprojectiles. Selective and nonselective tests were made by transferring the healthy green and white calluses, subcultured for 4 months on selective and nonselective medium. Several Hgm resistant calli had been obtained. Selective pressure was maintained over a period of 10 months.

  • Ecological drought resistance and adaptability of apple varieties

    For adequate yields in apple plantations, during the long growing period of the fruit primordia, one of the decisive factors is water supply. Indicators of stress are valuable signs for the diagnosis of drought and necessity of watering, i.e. planning the irrigation of plantations. The aim of the present study was to find reliable signs of water stress on apple trees and at the same time conclude on the drought tolerance of different varieties. The plantation of apple varieties grafted on various stocks and cultivated according to different systems (irrigated, non irrigated, integrate and biological) has been examined continuously by leaf analysis. Along the period of growing fruits, measurements were made in the field, then the leaf samples were analysed in the laboratory for composition of pigments, carbohydrates and antioxidants, as well as the histology of the tissues checked. Without irrigation, the mean leaf mass and the relative chlorophyll content (SPAD) of the variety Idared on M4 stocks increases beyond the values of 50 SPAD, whereas on M26 stock and integrated system, it declines. In the collection of varieties, grown according to the „integrated” system, during the development of fruits, ‘Gála’ and ‘Remo’ varieties have been affiliated to the “less susceptible” group regarding drought tolerance because of the leaf morphology, structure and content of SPAD and antioxidants. However, ‘Idared’ and ‘Jonagold’ belong to the “susceptible” group. To the same group are ranged the ‘Akane, Red Rome vanWell, Pink Lady’ varieties. Those varieties reacted to a short period of drought by increased production (content) of carbohydrates. Regarding changes of carbohydrate content ‘Greensleeves’ and ‘Ozark Gold’ varieties belong to the moderately water dependent (requiring) group. In the field, SPAD-tests facilitated the measurement of nutrient-uptake and incorporation, which is proved by the tight correlation between the data of SPAD and the increment of leaf weight (r=0.76–r=0.88), however, this depends on the variety too. SPAD is an indicator of water supply and is related with the density of stomata, cannot used for the selection of water-exigent varieties but for (drought) tolerant ones.     Bacterial soft rot is a polyetiological disease attacking calla lily [Zantedeschia spp (L.) Spreng.]. This disease has reduced the commercial value of this crop. This work aims to review scientific information to give an insight into calla lily soft rot causal agents, symptoms, factors favouring the disease, virulence mechanisms and management strategies. Special emphasis is put on the current progress with regards to understanding calla lily mechanisms of resistance to soft rot and their potential for the development of tolerant/resistant cultivars with commercial traits.
