The evaluation of grape vine decline pathogens in the experimental field of the Georgikon Faculty of Agriculture in Cserszegtomaj
Vine decline causes important economic loss in viticulture, especially in longer view. Several causal pathogen were described lately, however little is known about the etiology or epidemiology of these pathogens on grapevine rootstock. It is well known that these diseases affect grafted and rooted grapevines and are not specific to any scion-roots...tock combinations. Our aim was to determine what pathogens are presents in the experimental field, especially causal agents of the rootstock decline. Different grapevine rootstocks and scion varieties were tested in our trial. Isolations were made from the wood tissue and pathogenity tests were done with isolated Cylindrocarpon destructans. The possibility of infection during the propagation process was studied as well. Most commonly Cylindrocarpon sp. and Phomopsis sp. species were identified from the examined varieties. Cylindrocarpon destructans was able to spread to apical (shoot) and basal (root) direction from the point of infection with uneven speed. Callus development is not inhibited by the fungi causing the leaf symptom of the vine decline. Shoot development is reduced if unhealthy parts are grafted to each other.
In a two-year study, we aimed to determine the susceptibility of four plum cultivars to two fungal pathogens of plum (Stigmina carpophila and Polystigma rubrum) in two training systems with tree spacings of 4 x 1.5m and 6 x 3m. Results showed that shothole symptoms were not detected on cvs ’Bluefre’ and ’Stanley’ in August, 2016. Disease i...ncidence was above 50% in the case of ’Čačanska lepotica’ in both training systems in 2016. There were no significant difference between the two training systems. Shot hole incidence was lower in the 6 x 3m spacings compared to the 4 x 1.5m spacings on cv ’President’ in 2016. Cultivar ’Čačanska lepotica’ showed the highest incidence of Stigmina carpophila in the 4 x 1.5m spacing in 2017. Disease incidence of Stigmina carpophila was significantly lower in the 6 x 3m spacing compared to the 4 x 1.5m spacing. Shothole incidences on cv ’President’ were similar to the values in 2016 ranging from 40% to 60%. Leaf disease incidence was higher in the 4 x 1.5m spacing compared to the 6 x 3m plot. Low disease incidence (below 10%) was observed on cv ’Stanley’ in 2017 and only in the 4 x 1.5m spacing. There were no visible symptoms of blackhorn dotty in 2016 due to inadequate weather conditions for the Polystigma rubrum fungus. However, all the four cultivars were infected by Polystigma rubrum in 2017. The most susceptible cultivar was cv ’Čačanska lepotica’ with the highest disease incidence in the 4 x 1.5m spacing. Disease incidence of this cultivar was lower in the 6 x 3m spacing which was significantly less than in the 4 x 1.5m spacing. The least susceptible cultivar was ’Bluefre’ and symptoms were observed only in the spacing of 4 x 1.5m. The disease incidence of cv ’President’ was similarly low to cv ’Stanley’ in both spacings.
Most of the cultivars grown before the disaster of phylloxera in Tokaj can be found only in grapevine collections. The aim of our study was to determinate the cultivation values of six old grapevine cultivars of the wine-region, namely 'Balafánt', 'Budai gohér', 'Purcsin', 'Török gohér, 'Piros (red) furmint' and 'Változó (altering) furmint'.... Based on the results of the investigations from 2004 to 2006 it can be concluded that among from the examined cultivars of Tokaj `Budai gohér', Turcsin' and 'Változó furmint' match the best with the technological and consumer requirements of recent days. However the yield of 'Budai gohér' in certain vintages is not acceptable and red wine cultivars are not permitted at this time in the wine-region, but in qualitative responses these presented steadily good data. On the other hand the lowest sugar content was measured and development of noble rot was not registered at all in case of Piros furmint' and `Balafánt'. Their spread in Tokaj is not probable.
Thirty-one old Hungarian grapevine (Vitis vinifera L.) cultivars were investigated on 7 microsatellite loci to characterize them, to separate the cultivars from synonym names, and to confirm parent-offspring connections. Conculta (group of cultivars or bud sports) members, such as `Goher' and Tajor' representatives, were studied to find a suitable... locus for the separation. Synonyms, conculta members, subcultivars and clones of Turmine, which was the most important cultivar of Tokaj, were also analyzed to separate the members of the different taxonomic levels. Pedigree of 'Kiralyleanyka' was examined to find the missing ancestor, because the parent-offspring connection between the natural hybrid and `Koverszolo' is questionable.
Selection of the chance seedlings of `Mézes körte' (Pyrus communis L.) from the gene bank of Keszthely
We have concluded the selection tests of the `Mézes körte' seedlings planted in the spring of 2006, with special emphasis on the cotyledonary, foliage leaf and the height of plant. Out of the 75 seeds planted in rows, there were 40-45 pieces growing out, so during the first cotyledonary test we had to calculate with almost 40% decay. On 12th Apr...il 2006, we recorded some of the important characteristics of the seedlings in their cotyledonary stage which characteristics were important from the point of view of selection (cotyledonary form, cotyledonary length, cotyledonary thickness, cotyledonary colour, cotyledonary petiole length, cotyledonary petiole thickness, cotyledonary petiole colour). The above morphological characteristics are shown in Table No. 1-6. We have also tested the seedling in foliage leaf state, paying special attention on the development stage of the plants (colour of foliage leaf , height of plant). We have completed statistical calculations of the two above mentioned characteristics. The result of that is summarised in Table No. 8-9. The variation coefficient show smaller value in the case of the foliage leaf number (15-32%), while the wider range of spread of the data referring to the height of the plant is shown by the 33-61% CV values. On charts No 4-9. we present the relationship between the height of the plant and the number of foliage leaf, as well as the differences between the two graphs. Based on the above charts and graphs it can be defined that the 40% destruction of the developing seedlings during the period till the next measurement reached 70-80% level. In spite of this however some seedlings showed strong and balanced growth (A44, B42, C25, D16, E5, E39, F38), the further testing and selection of those is to be completed in the future.
Primers designed for the detection of grapevine pathogens spreading with propagating material by quantitative real-time PCR
Several grapevine pathogens are disseminated by propagating material as systemic, but latent infections. Their detection and identification have a basic importance in the production and handling of propagating stocks. Thus several sensitive and reliable diagnostic protocols mostly based on molecular techniques have been developed. Of these methods... quantitative real-time PCR (q-PCR) has recently got an emerging importance. Here we collected primer data for the detection and identification of grapevine pathogens which are important in the production of propagating stocks by q-PCR. Additional novel techniques that use DNA amplification, hybridization and sequencing are also briefly reviewed.
In continuous studies regarding the conservation of the advantageous condition of the soil structure, the highland plantations that are surrounded by living waters have major importance where the erosion of the soil, nutrients and chemicals endangers the quality of the water and the living world of the waters. At the same time the extreme weather ...and dry summers of the past years have made it important to look for soil cultivation solutions and technologies that are capable of decreasing the evaporation of the soil, for enhanced conservation of soil moisture and to improve the soil structure that is required for the growth of healthy roots. In our experiment we have studied soil mulching with organic plant residue, by sods and the effect of the mechanical cultivation on the soil and on the grape through two growing seasons. From the studied treatments, the mulching with organic plant wastage showed the most efficient results from the point of view of the nutrient distribution in the grape and yield. It also had a positive effect on the soil structure and humidity.
The use of pathogen-free planting stock for new vineyard establishment is a key component in the maintenance and expansion of vine and quality table grape production. The success of the necessary changes in the structure of the grape industry is forced by the globalization process, the climate change, the rediscovery of autochton varieties as well... as breeding of new tolerant and resistant varieties. The renewal of vineyards largely depend on the availability of planting stocks. Serbia and Hungary found a common interest in establishing pathogen-free stock materials from newly breed resistant varieties and clonal selections of varieties which are traditional in the Serbian-Hungarian border area. During a cross-border cooperation program a complex system for the production of pathogen-free grapevine propagating material was established. Using heat therapy, in vitro shoot tip culture and traditional and molecular diagnostic techniques new pathogen-free stock materials were established from 26 varieties. They have been or will be tested for the presence of most important grapevine viruses, phytoplasmas, as well as bacterial and fungal pathogens. The complex system applying green grafting for indexing on grapevine indicators can shorten the duration of the procedure from 4 years to two-three years.
The present paper evaluates the result of irrigation experiments carried out on snap beans sown in spring and summer and grown with and without irrigation. The experiments were run over the course of 12 years. In the average of 12 years, the yield was 2.8t ha-I for spring sown and 1.9 t ha-I in summer-sown plants without irrigation. The lowest lev...el of profitable production, the 5.5t ha-I was reached twice in the case of spring sowing and only once in the case of summer sowing. Profitable yield production can be ensured only with regular irrigation and thus the yield may be increased by 4-5 times. In four of the twelve years we determined the canopy surface temperature of snap bean stands with and without irrigation. A Raynger II infrared remote thermometer determined the canopy surface temperature every day at 13.00 hours. The canopy temperature can well characterize the water supply of plant stands. This parameter may be used for describing the degree of drought and the water turnover of plant stands with different water supply. The positive values of foliage-air temperature differences (SDD) numerically express the degree of drought and the water supply of the crops. The results indicated that a 1 °C higher SDD value may cause 90-130 kg/ha yield loss.
According to previous studies some anatomical features seem to be connected with resistance or susceptibility to scab caused by Venturia ineaqulis (Cke./Wint.) in case of a given cultivar. Study of leaf anatomy of three scab resistant (‘Prima’, ‘Florina’, MR–12) and two susceptible (‘Watson Jonathan’, ‘Golden Delicious Reinders’)... apple cultivars have been made. Preserved preparations made of leaves has been studied by light microscope. Studied parameters were: thickness of leaf blade, thickness of palisade and spongy parenchyma, thickness of epidermal cells, thickness of the cuticle. By measuring leaf thickness and epidermal cell thickness visible differences appeared in certain cultivars, while most conspicuous difference has been shown in thickness of the cuticle.
Correlation between pigment contents and FRAP values in beet root (Beta vulgaris ssp. esculents var. rubra)
It is well known that beetroot quality is determined mainly by the red pigment content (betacyanins) and its uniformity of the root. The effect of the most important red pigment components (betanin). the total polyphenol content and antioxidants were studied in 20 beet root varieties. Antioxidants were expressed in FRAP (ferric reducing ability of... plasma) values in pM/I. Our results indicated a close correlation (r = 0.7799 and r = 0.7435. respectively) between betanin and total polyphenol contents of the root as well as between FRAP values. Our measurements showed more than threefold differences in total antioxidant activity among varieties. the lowest value being 196.4 13M/1 and highest 702.57 pM/I. The corresponding betanin (16.3 and 57.8 mg/100 ml) and total polyphenol (37.5 and 85.5 mg/100 ml respectively) contents show similar differences. Based on our results it can be stated that varieties of higher betanin and poliphenol contents have higher antioxidant values as well. Accordingly, the two compounds must have a role in the evolution of antioxidant effects.
Results of our experiments prove that pear is more or less sensitive to the reduced bee pollination period. However, the reaction (or the sensitivity) of cultivars may be different to the reduced bee pollination. Most cultivars produce much less yield under reduced bee pollination or no yield with the exclusion of bees but in the case of some cult...ivars total exclusion of bees does not prevent the yield formation and what is more sometimes reduced bee pollination can be resulted in somewhat higher yield than open pollination. Typical reaction, however, is a significant yield reduction with reduced bee pollination. Pear seems to be somewhat less sensitive to the partial reduction of bee pollination period than apple or quince. The first half of the flowering period seems to be more important in yield formation because usually higher yield was resulted when pear cultivars received open pollination in the first than in the second half of the blooming period. Based on our experimental results no definite relationship between parthenocarpic capacity of cultivars and the yield under reduced bee pollination can be established. So reduced bee pollination does not seem to contribute the parthenocarpic fruit formation in pear.
Production of transgenic carnation with antisense ACS (1-aminocyclopropane44-carboxy late synthase) gene
Dianthus chinensis and Dianthus caryophyllus varieties were tested for shoot regeneration from leaf and petal explants and transformed with Agrobacterium tuniefaciens strains (EHA 105 and LBA 4404) harbouring an apple derived ACS cDNA in antisense orientation in order to reduce ethylene production and influence the ethylene dependant traits in car...nation. After transformation regenerating shoots were selected on MS medium containing 50-75-100-125-150 mg/1 kanamycin and supplemented with 1 mg/1 BA, 0.2 mg/1 NAA. Transgene integration was proved by PCR analysis with npt II spcific primers followed by Southern hybridisation of DNA isolated from green shoots on medium containing 150 mg/1 kanamycin. Several putative transformants were subjected to RT-PCR in order to examine the npt 11 expression at mRNA level. Both the transformant and the non-transformant plants were potted into glasshouse to observe the effect of changed ethylene production on flowering time, petal senescence and vase life.
The aim of this study was to demonstrate the 10% threshold level for cherry leaf spot incidence on 23 commercial sweet cherry cultivars in two training systems. Twenty three cherry cultivars were evaluated in the two training systems with a spacing of 4 x 1 m and with a spacing of 2 x 5 m. Results showed that leaves of many cultivars were heavily ...infected, e.g. cultivars (cvs) ’Biggareau Burlat’, ’Sunburst’, while others showed low disease incidence e.g. cvs ’Celeste’ and ’Blaze Star’. According to the 10 % threshold level, cv ’Celeste’ proved to be the most resistant cultivar to leaf infection, while cv ’Münchebergi korai’ exceeded extremely the 10% threshold level. Leaf spot incidence was affected by training system on most cultivars. Thirteen cultivars had less than 10% infection in the 4 x 1 m spacing (i.e. they did not reach the 10% disease threshold level). At the same time, only 3 cultivars showed less than 10% infection in the 5 x 2 m spacing.
