Model experiments for establishment of in vitro culture by micrografting in apple

Micrografting was used in our experiments for establishment of in vitro culture from one rootstock (`JTE-F') and three scion cultivars (`Remo', 'Rewena' and `Reanda') of apple. Shoot tips of these cultivars were harvested from field and grafted onto in vitro rootstock cultivars. Their survival and development were studied. 42-...93% of shoot tips survived and developed further depending on cultivar. Impermanent browning of sticking agar-agar could be observed in 21-25% of the micrografts depending on cultivars but discolouration of agar-agar ceased within one week and did not cause any death of shoot tips. We used micrografting successfully for establishment of in vitro culture from cultivars, from which earlier with conventional methods the culture establishment was not possible because of hard tissue browning. However, further studies are necessary to ensure the survival and development of shoots after removing them from micrografts.

In vitro comparative study of two Arundo donax L. ecotypes’ selenium tolerance

Selenium tolerance of two somatic embryo-derived Arundo donax L. ecotypes (Blossom, 20SZ) were compared in in vitro culture. Sodium-selenate (1 – 100 mg L-1) as known the most phytoavailable selenium form and the less studied red elemental nanoselenium (100 mg L-1) were applied as selenium treatments. Basis on the results Blossom ecotype seem...ed to be more sensitive to the sodium-selenate than 20SZ. Inhibiting effect of selenate was effectuated above 10 mg L-1 in case of Blossom, which was manifested in decreased survival rate and growing parameters. Contrast to this 20SZ could tolerate the selenate ≤ 20 mg L-1 without any toxic symptoms. Lower selenate tolerance of Blossom could be explained with higher selenium accumulation. Both of two ecotypes could also uptake and accumulate the red elemental nanoselenium however in much less extent compared to selenate.

In vitro propagation of 'Echo' cultivars of Eustoma grandiflorum (Raf.) Shinn.

Eustoma grandiflorum (Raf.) Shinn. 'Echo' Fl cultivars ('Echo White', 'Echo Rose', 'Echo Blue', 'Echo Blue Picotee') were used and multiplication of shoots was evaluated on Murashige and Skoog (1962) basal medium with 11 g/1 agar-agar and 20 g/1 sucrose. To test the effect of BA different concentrations were added: 0.10, 0.25 mg/1 and ...a culture medium without BA. Differentiation of roots was examined on Jámbor-Benczúr and Marta (1990) basal medium with the same concentration of agar-agar and sucrose. To examine the effect on rooting, various concentrations of NAA were used: 0.5, 1.0, 2.0, 3.0 mg/l. The pH was adjusted to 5.6 in every case using KOH. We studied the after-effect of different concentrations of BA during the acclimatisation. During the multiplication, the cultivar 'Echo White' formed the most shoots and the smallest leaves on the medium with 0.10 mg/1 BA. Fortunately, in the case of this cultivar, the number of shoots was reduced and the length of leaves was increased succesfully on the medium without BA. The other three cultivars developed the longest leaves on the medium containing 0.10 mg/1 BA. Sometimes not only shoot regeneration but spontaneous rooting was observed during the multiplication. Examining the rooting, the highest percent of roots was found on the medium with 1.0 mg/1 NAA, and the cultivar 'Echo Rose' formed the most roots on this medium. Higher concentration (2.0 and 3.0 mg/1) of NAA already reduced the number of roots in all of the cultivars. During the acclimatisation, the percentage of survival was 76.3% and the tallest plants with the longest leaves were found on the multiplication medium with 0.25 mg/1 BA. 'Echo Blue Picotee' gave the best results with the tallest pieces and longest leaves on this medium.

The development and reproduction of grape phylloxera (Daktulosphaira vitifoliae Fitch) strains of Hungarian vine-districts in root-bioassays

The root bioassay method allows for 4-6 week continuous observation of grape phylloxera feeding on the grape root. 10 Hungarian phylloxera strains were compared on susceptible Vitis vinifera cv. Chardonnay and the resistant rootstocks of V. berlandieri x V. riparia Teleki 5C and V. berlandieri x V. rupestris ...Georgikon 121 in in vitro observations. The strains originated from Villany and Eger (Figure 3) had higher reproduction on the root of V. berlandieri x V. riparia Teleki 5C (201 and 119 eggs) and were more aggressive than the others (average production 10 eggs). The continuous high level of survival, development and reproduction of the Eger strain on Teleki 5C (V. berlandieri x V. ripuria) until day 46 may be due to adaptation.

Damages caused by winter frosts, their temporal variation and frequencies in the main fruit growing region of Transdanubia and of the East Tisza regions of Hungary

The aim of the study was the study of winter frost damages, especially their changes expressed in temporal frequencies on the main fruit growing regions of the country. In our earlier paper, we introduced the calculation with the term LT50 as the quantitative expression of temperature threshold, when the lethality halves the survival of plant o...rgans, buds or cells causing 50% death rate. The damage is highly dependent on the temperature and on its duration (length of time), but not at least on the frost tolerance of the fruit trees. The incidence and severity of damage is analysed according to the apricot and peach varieties of their different susceptibility or tolerance too. Four fruit growing regions, two of the in Transdanubia and two belonging to the regions east of the Tisza river have been selected to trace the incidence and severity of frost damages. For that purpose, we analysed the history of the past 60 year period, 1951–2010, utilising the database of the network of 16 meteorological stations of the countrywide service. Being aware of the values of LT50 during the rest period and afterward, the compulsory dormancy caused by low temperature, the number of days, the probability of frost damage could be predicted. The role of the orographical profi le, the height above sea level and the exposition of plantations are also decisive. Within the same plantation, 20–30 m difference of level may cause large diversity in temperature and frost damage. Air circulation and regular incidence of winds within the Carpathian basin modify the occurrence and severity of damages. Lowlands near the southern and northern country borders are particularly exposed to winter frosts. Most damages are reported in February, as temperatures below –20 °C especially if the fi rst part of the winter was mild, or in January was a warm period. With the end of the physiological rest period of the trees, the frost-susceptibility increases signifi cantly, and a cold period of –15 °C may cause heavy damage. This study proves that tolerance of varieties infl uence the damages substantially. By planting frost tolerant varieties, winter frost damages could be diminished by 40–50% at the same growing sites. Present results may also offer a tool to estimate the risk of frost damages and express the security of yields at a given site based on the data accumulated in the database over many years.

Study of Erwinia amylovora colonization and migration on blossoms of susceptible and tolerant apple cultivars

The stigmata of detached flowers of susceptible and tolerant apple cultivars were inoculated with about 104 gfp labeled Erwinia amylovora . There were no apparent differences in the colonization, multiplication and survival of the bacteria on the stigmatic surface of the culivars. Bacteria were washed down to the hy...panthium surface 24 hours after inoculation. The visual symptoms of the infection were the discoloration and shrinkage of the floral parts. The gradual browning associated with the infection appeared first on the surface of the hypanthium followed by the discoloration of the style. The color of the filaments turned into brown only 120 hours after the inoculation. Bacterial cells were not detected in the tissues of the styles and filaments. The traits of the hypanthium surface are of prominent importance in the progression of the infection. The wrinkled surface, the convex shape of the outer epidermal cell walls with thin cuticle and the sunken stomata helped to preserve a water film for a longer period providing medium for the motility of the bacteria in the susceptible cultivar. Bacteria were restricted to small water droplets on the flat and waxy surface of the hypanthium of the tolerant cultivar and only a few were able to enter the tissues.

Large bacterium aggregations were detected in the intercellular spaces of the parenchyma of the susceptible cultivar 48 hours after the inoculation. In the next period the Erwinia amylovora cells gradually invaded the intercellulars of the hypanthium wall, the wall of the ovary and the pedicel. Low level of bacterium aggregation was found in the intercellulars of the tolerant cultivars. It is suggested that the progression of the infection was inhibited also by physiological factors.

Water relations of sour cherries (minireview)

Recently, the sour cherries as food resources become more important for health preservation and so the modernization of growing technology in sour cherry production will be timely. The global warming and inadequate distribution of precipitation result a decrease in the alternancy of sour cherry production, as well. Sour cherries rather adapted survival of drought than sweet cherry trees therefore a few studies performed to explore the water requirement of sour cherry varieties. The rootstocks, the type of soils in plantation and the water balance influence the water management of sour cherries. In orchards, in particular first year plantation, use of various row covering contribute to preservation of the natural water pool of soil and affect on the tree vigor, yield and fruit quality. Wide-spread application of integrated fruit growing technology and climate changes the researches are pointed to develop efficient irrigation technology based on transpiration yield model. The crop model based on use of meteorological data was developed for cherry orchards in order to predict transpiration of trees, dry matter production and fruit yield. The linear relationship between dry matter accumulation and transpiration was verified for sour cherry trees. Other models essay to asses the effects of climate changes on crop production. Importance of economical production and fruit quality such as ingredients of raw materials and food increases in intensive sour cherry orchards used by irrigation techniques. Because of climate changes it should more pay attention to research concerning on the stress physiological response of sour cherry varieties and post-harvest fruit quality.

Microsporogenesis of peach (Prunus persica L. Batsch) varieties

Bud dormancy during winter is a critical factor in peach production in Hungary. The yield is determined basically by the survival rate of flower buds during winter frosts and by their ability to develop normal floral organs. It is important to investigate the genetic basis of slow floral development during dormancy for the purpose of breeding p...each varieties with better winter hardiness. The aim of the present research was to examine microsporogenesis in 14 peach varieties during three successive winters in a Hungarian germplasm collection and to study the effectiveness of this method in variety evaluation. There were significant differences in the dynamics of microsporogenesis both between the varieties and between the years. Of the varieties, ‘Mayfire', bred in California, possessed the quickest pollen development rate. The microsporogenesis of `Piroska', a Hungarian local variety, was the slowest. Rapid floral bud development was observed in `Aranycsillag', `Springcrest' and 'Venus'. A medium developmental rate was characteristic of `Babygold 6', Fairlane', `Michelini' and `Red June', while development was slow in 'Champion', 'Early Redhaven', `Redhaven', `Harko' and `Mariska'. Based on these results, the study of microsporogenesis represents a reliable method for the phenological description of peach varieties during dormancy. The application of this method makes it possible to identify varieties and landraces with slow flower bud development, suggesting better winter hardiness.

Micropropagation of an orchid Dendrobium strongylanthum Rchb.f.

A simple and reliable procedure for in vitro propagation of an orchid Dendrobium strongylanthum Rchb.f. was studied. Protochorm was induced from seed explants on 1/2 MS medium supplemented with 0.2 mg/L NAA. A mass of protochorm could be multiplied on proliferated medium of 1/2 Ms containing 0.5 mg/L V-6-BA. And bud differenti...ation of green global body was cultured in the same media, 2-2.5 cm shoots were formed after 30 day of culture. Addition of mashed banana and 0.5 mg/L NAA to 1/2 MS medium promoted root formation and vigorous growth. The plantlets were acclimatized and transplanted to compound materials of humus:sawdust (1:1) in greenhouse, the survival rate was more than 98%.

Effects of activated charcoal on rooting of in vitro apple (Malus domestics Borkh.) shoots

Rooting of in vitro 'Royal Gala' shoots was studied under different conditions of root induction and root elongation phase. The rooting capacity was affected by both rooting phases. Very high rooting percentage could be reached with both liquid and solid root induction media. Raising the temperature from 22 °C to 26 °C during root in...duction phase increased the rooting percentage. Presence of activated charcoal in root elongation media can affect the number of roots per rooted shoots and can increase the rooting percentage, the length of roots and the rate of survival depending also on other conditions during rooting. Presence of NAA in root elongation media reduced the number and the length of roots considerably. Favourable effect of activated charcoal on rooting was mainly due to adsorption of NAA.

Comparison of flower bud development in almond, apricot and peach genotypes

The phenological processes of flower bud development of stone fruits during dormancy are not thoroughly known. The yield of these species, especially of almond, apricot and peach is determined basically by dormancy of flower buds, the survival rate of buds during winter frosts and by their ability to develop normal floral organs in the next After the initiation of floral primordia, flower bud development is taking place in continuous space until blooming, though at different speed characteristic to the species. To study flower bud development during dormancy we applied two alternative methods in different genotypes of almond, apricot and peach: (1) examination of pollen development (microsporogenesis), and (2) the measurement of pistil length. The samples were collected from the central part of Hungary during the dormancy period of 2004/2005. The three fruit species differed significantly in the speed of flower bud development, it was the quickest in almond, followed by apricot and peach. In addition to the species, there were significant differences in the process of microsporogenesis and pistil development between genotypes within species and also between the different types of shoots on which the buds were located. On short shoots buds developed at a higher speed, than on long shoots. Based on our observations, on the short shoots the period of endodormancy was shorter with 5-30 days, according to genotypes, compared to the long shoots. This difference, however, decreased to 2-3 days by the time of blooming.

New in vitro micrografting method for apple by sticking

The requirements for in vitro micrografting in apple are described. In vitro multiplicated shoots of cv. Royal Gala were the sources of rootstocks and scions after different pre-treatment, respectively. Oxidative browning of cut surfaces could be inhibited by the use of antioxidant mixture during grafting process. Sci...on base cut in v-shape was stuck by 1% agar-agar solution into the vertical slit of rootstock. There was no any displacement and the rate of fused and further developed grafts was 95 percent. Agar-agar between the rootstock and scion made the transport of different materials possible and hold the graft units together until the fusion took place. Fusion was proved also by histological studies. Some of in vitro micrografts were planted and acclimatisated and the survival was 100 percent.
