The effect of different carbohydrates on the multiplication of Hosta cultivars
115-117.Views:159The effect of seven concentrations of two carbohydrate sources were compared to determine the best source and the most suitable source and concentration for micropropagation of some Hosta cultivars: H. 'Gold Haze', H. 'Gold Drop' and H. 'Dew Drop'. 0, 5, 10, 20, 30, 40 and 50 g/1 sucrose or glucose were added to a MS basic medium supplemented with 3 mg/1 kinetin and 0.1 mg/1 IAA. For 'Gold Haze' 40 g/1 sucrose proved to be the best source and concentration, the proliferation ratio was 15 shoots per explant. Thirty g/1 sucrose concentration was the optimum for 'Gold Drop', the proliferation rate was 14.6 shoots per explant. In 'Dew Drop,' the best results were obtained with 30 g/1 sucrose but 40 g/l sucrose gave good results too. Both cultivars rooted well on these media. On glucose containing media, very low propagation rates were found in all concentrations and all examined cultivars.

Changing of carbohydrates by inoculation of Pseudomonas savastanoi pv. phaseolicola oil bean lines with different resistance
82-85.Views:181The Pseudomonas savastanoi pv. phaseolicola (PS) is one of the most significant stressors of bean (Phaseolus vulgaris L.). Chemical and agrotechnical treatments have minor importance, so breeding has great part in the protection against this pathogen. Most of the cultivars are susceptible to PS. The genetic background of resistance in the plant is a complex system. Leaf resistance is a monogenic system, but there are some modifier genes. The pathogen species can be divided into different races.
To understand the functioning of this resistance gene, experiments were carried out using bean varieties with different genotypes and near isogenic lines of bean. Eight lines were tested. Our main objective was to test bean lines with PS with high virulence.
The experiment was made in greenhouse and on field. The virulent bacterium strain has been isolated in Hungary.
The changes of carbohydrates were tested after infection. In homeostasis the level of carbohydrates (especially glucose and fructose) were higher in susceptible lines. In case of artificial and natural infection the decrease of glucose were more significant in susceptible lines than in resistant lines. In the leaf samples from systemic chlorosis the level of this carbohydrate increased.
These changes are connected with the level of resistance, but more experiments are needed to verify this assumption.

Does foliar nutrition influence the pear fruit quality?
25-28.Views:147The influence of the foliar nutrition on the pear fruit quality (Pyrus communis L.) cv. 'Williams' was studied in years 1997 and 1998. We determined the contents of individual sugars (glucose, fructose, sucrose and sorbitol) and organic acids (malic, citric, fumaric and shikimic) by HPLC (High Performance Liquid Chromatography). The sizes of the fruits (diameter, length, weight) were measured as well as the amounts of soluble solids and titrable acids. The experiment comprised two treatments: the foliar nutrition and the control. In the treatment of foliar nutrition the trees were sprayed five times (from May 22 to July 7) with a foliar fertilizer, which contained 15% of P205, 20% of K20, 0.1% of Mn, 0.1% of B and 0.1% of Mo. The foliar nutrition influenced higher quantities of sugars (glucose, sorbitol, soluble solids) and organic acids (malic, citric) but had no effect on the contents of shikimic acid, pH juice and titrable acids. At the treatment of foliar nutrition a trend of decrease in the contents of fructose, sucrose, fumaric acid, boron, and zinc was noticed.

Influence of strawberry and raspberry consumption on the antioxidant status of human body
109-113.Views:169Several parameters influencing the antioxidant capacity were measured in our experiments to compare strawberry and raspberry cultivars. The analysed cultivars were: 'Honeoye', 'Elsanta', 'Onebor (strawberries) and 'Glen Ample', Fertődi zamatos', 'Mailing Expoit' (raspberries). Besides the determination of dry matter content the content of glucose, fructose, total phenol and anthocyanine, the reducing power, H-donating activity and total scavenger capacity were measured as well as the compound of mineral nutrients. Among the species and the cultivars, differences were obtained. These results call the attention to the features of cultivars and to count on the antioxidant-capacity describing values in case of consuming these fruits. In early spring and summer the consumption of strawberry and raspberry has a beneficial contribution to the antioxidant status of our body. In this respect these are more valuable than the year-long consumable apple. Accordingly, our measurements showed higher amount of sucrose, phenolic compounds and anthocyanine in raspberries, but strawberries contained more glucose and fructose and had stronger H-donating activity. It is worth to keep in mind that e.g. which cultivar contains more of certain compounds because of the strictly determined allowable dose in some diseases (e.g. diabetes).

Composition and storage of pear cultivars from Nagykanizsa
63-68.Views:258The composition of five pear varieties (‘Abate Fètel’, ‘Bosc’, ‘Williams’, ‘Conference’, ‘Packham’s Triumph’) grown in Nagykanizsa was investigated in three consecutive years (2008, 2009, 2010). A storage experiment was performed in 2008. Four winter pear cultivars were kept in an ULO store for four months and their parameters measured after two and four months. The parameters tested were: size, weight, water soluble solids, titratable acidity, glucose, fructose, sucrose, water soluble pectin, total polyphenols, free radical scavenging capacity, copper and zinc content. The fruits of ‘Conference’ and ‘Bosc’ varieties were found to contain the highest sucrose and total sugar content, while ‘Abate Fétel’ had the lowest sucrose and highest glucose levels among cultivars tested. ‘Williams’ pear was the most acidic. Brix, total sugar, sucrose and water soluble pectin were decreased during storage. Titratable acidity slightly decreased in fruits of Conference pear. Polyphenols and free radical scavenging capacity did not show a significant change during storage.

In vitro rooting and anatomical study of leaves and roots of in vitro and ex vitro plants of Prunus x davidopersica 'Piroska'
42-46.Views:189The process of in vitro rooting and the anatomical characters of in vitro and ex vitro leaves and roots of Prunus x davidopersica 'Piroska' were studied. Best rooting percentage (50%) and highest root number (5.0) was achieved in spring on a medium containing 0.1 mg/I NAA + 30 g/1 glucose. At the end of rooting the parenchyma of the in vitro leaves was more loose and spongy, than during the proliferation period. In the first newly developed leaf of an acclimatised plant, the parenchyma was much more developed, contained less row of cells and less air space too, compared to the leaves developed in the field. The in vitro developed root had a broad cortex and narrow vascular cylinder with less developed xylem elements, but at the end of the acclimatisation the vascular system became dominant in the root.

Evaluation of fruit quality parameters of Rosa taxa from the Carpathian basin
81-87.Views:172From the wide range of genetic sources available in Hungary, we have chosen as objective the evaluation of those rose taxa, which — on the base of their quality and morphologic features — are suitable for selecting fruit production varieties or parent varieties for further breeding. We examined physical and inner parameters of fruits of Rosa taxa, and evaluated the correlations by mathematical statistic methods. Namely, if a correlation can be found between physical and inner parameters, fruit production value can be determined by less examinations in the future. According to our results, there is a large variability in some physical parameters (weight, diameter of fruits; weight, number of achenes) as well as in inner content values (vitamin C, glucose, fructose, K, Ca, Fe, Zn and Cu content) of rosehip, regarding production year, habitat and even the individual of examination. Twice as much ascorbic acid can be found in achene-free fruits, and nearly five times as much in their dry product as in fresh whole fruits. It was established repeatedly, that vitamin C concentrates in fruit flesh, and ascorbic acid content can be preserved better in achene-free flesh. There is no essential difference in citric acid and carbo-hydrate content of whole and achene-free rosehips. However, macro- and microelement content of whole fruits is 30-40% higher than that of achene-free fruit flesh. The connection between fresh weight and diameter, as well as achene number and seed weight of fruits can be described by the function y=axb. A significant relationship can be found in case of K-Fe, Ca-Mg, Ca-B, Ca-Mn and Zn-Cu, between fresh weight and B content of fruits. According to our examinations, fresh weight of rose species with a higher citric acid content is usually higher, while their Fe content is smaller. Glucose content of fruits of rose species is directly proportional to their Ca and Zn contents. Zn content of rose species with higher fructose content is usually smaller.

Effect of foliar fertilization on leaf mineral composition, sugar and organic acid contents of sweet cherry
45-48.Views:366Influence of a three-year-long foliar fertilization on mineral composition of leaf, content of sugars (glucose, fructose, galactose and sucrose) and organic acids (citric, malic and fumaric acid) of sweet cherry (Prunus avium L.) fruits was studied. The experiment was conducted during 2005-2007 in West Hungary on mature cv. `Germersdorfi 3' grafted on Prunus mahaleb rootstock, planted in 1999. Trees spaced 7 x 5 m, and growing in a calcareous chernozem soil. Trees were foliar-fertilized with potassium (K) as KNO3 and calcium (Ca) as Ca(NO3)2. Potassium spraying was carried out 3 (K1) and 5 (K2) while calcium was applied at 3 (Ca 1), 5 (Ca2) and 6 (Ca3) weeks after full bloom. Beside fruit analysis, complete soil and leaf analysis were done to study the rate of nutrient uptake and its effects on fruit quality. Contents of nutrients of soil and leaf were determined by atomic absorption and spectrophotometric method, while sugars and organic acids in fruit were determined by HPLC. The applied treatments (except K1) had been increasing leaf K significantly compared to the control till ripening. Most of treatments had no significant effect on Ca content of leaf till ripening. From applied treatments only the boron treatments had significant increasing effect on contents of all examined sugars, compared the control. Furthermore, the effect of calcium spraying on the contents of organic acids was significant.

Carbohydrate utilization of Erwinia amylovora in vitro
31-34.Views:188Nectar is a multi-component aqueous solution that promotes bacterial multiplication. The concentration of nectar in plant flowers is not stable since it is under the influence of environmental conditions, especially free moisture and relative humidity. Experiments were conducted with "artificial nectar" and directed along two lines: (1) determination of the optimal concentrations of carbohydrates for the growth of E. amylovora development (2) consumption of different carbohydrates besides basic sugars.
Solutions of "artificial nectar" were prepared in different compositions by changing the dominance of basic sugars (fructose — glucose —sucrose) in proportions of 2:1:1, 1:2:1, 1:1:2 and between concentrations of 10-0.6% (diluted with Basal minimum broth) in order to determine optimal conditions for the development of E. amylovora.
At a basic sugar concentration of 10% bacterial multiplication started and continued until I log degree (from 106 to 107 cfu/ml). At concentrations of 5% and 2,5 % cells developed with nearly the same kinetics (from 106 to 8x107 cfu/ml and from 106 to 9x107 cfu/ml, respectively). Multiplication was more pronounced and nearly the same at concentrations of 1.2 % and 0.6 % (from106 to 2x108 cfu/ml). At a basic sugar concentration 30% total sugars bacterial multiplication did not occur, while at 20 % it was negligible, not measurable photometrically.
At minimal concentrations of F, G, S (between 1-0.1 %) bacterial cells were still able to multiply, producing organic acids from sugars.
Our study showed that E. amylovora requires only a small amount of sugars (0.1%) for multiplication (acid production) while high concentrations inhibit multiplication. There was a negative correlation between sugar content and cell density. The optimal range of sugar concentration was at about 1%.
Effect of "less frequent carbohydrates" to E. amylovora multiplication was also determined using the API 50 CH strip. We could provide information on utilization of 39 carbohydrates by the bacterium at different categories as follows: Not utilized-, Slowly and weakly utilized-, Slowly and completely utilized-, Quickly and completely utilized carbohydrates. We suppose that carbohydrates that belong to the latter two groups could play an important role as nectar components in promoting E. amylovora multiplication in the blossoms of pome fruit trees.

Frost induced changes in enzyme activities and carbohydrate content in the spurs of some pear cultivars during the dormancy
41-44.Views:226Frost tolerance of pear cultivars was checked after artificial cold treatment in 2003-2005. Limbs collected during the endodormancy were exposed in a climatic chamber for 24 hours to —25; —28 °C, while those collected in the ecodormancy were kept at —15 and —18 °C. Frost damages of buds were registered according to a visually defined scale, then peroxidase (POD), polyphenol oxidase (PPO) enzyme activities and carbohydrate contents were checked in buds and spur-part below the buds. POD activity of untreated control in tissue below buds was higher than in the buds, which were increasing continuously during the endodormancy and decreased at the end of the ecodormancy. During endodormancy, cold treatment of —25 and —28 °C effected different changes of enzyme activity in buds of the cultivars. In the ecodormancy, enzyme activities increased after a cold treatment of —15 °C, whereas the activities decreased significantly after —18 °C. `Kaiser' — susceptible to frost — with its higher values of both enzyme activities marked out from other cultivars, which is correlated with its stress response. Changes in carbohydrate components — especially in glucose — of buds monitored well the different stress responses of tolerant and resistant pear cultivars induced by frost stress.

Floral biological studies on pear cultivars in relation to fire blight susceptibility
25-30.Views:185Floral biological characteristics that may influence cultivar susceptibility to fire blight were studied in 10 pear cultivars in two Hungarian orchards from 1999 to 2003. The receptaculo-ovarial, automorphic nectary is usually bigger in tolerant cultivars than in susceptible ones. Nectary stomata are meso- or xeromorphic. Susceptible cultivars tend to have more xeromorphic stomata, where guard cells are located 1-3 cell rows below the epidermis. The size of nectar chambers is usually smaller in susceptible cultivars. Floral nectar, consisting mainly of glucose and fructose, is more abundant and less concentrated if the cultivar is susceptible to fire blight. The amount of chlorogenic acid was higher in the flowers of tolerant cultivars than in susceptible ones.

The effect of different berries in human nutrition
147-151.Views:227The aims of this study were on the one hand to compare of examined compounds according to their importance in nutrition and human-health, and on the other hand we made preexperiments to investigate the relationships among antioxidant capacity and the endogenous substances which contribute the antioxidant status of the plants. The following species were involved in the experiment: raspberry, blackberry, black currant, elderberry and sour cherry. These fruits have potent health-promoting antioxidant power. Glucose, fructose, total phenol, formaldehyde and anthocyanin contents were determined in addition to ferric reducing ability. Our results reflected considerable differences in the measured parameters of the analysed species. In blackberries and elderberries the high antioxidant capacity is coupled with low carbohydrate content. Besides the formerly proven correlations between total phenol content, anthocyanin concentration and antioxidant capacity, these parameters also correlated with the measured formaldehyde concentrations, hereby we can follow the methylation /demethylation status of the plant.

Physiological and biochemical evolution of peach leaf buds during dormancy course under two contrasted temperature patterns
15-19.Views:120Budbreak anomalies in temperate fruit trees grown under mild conditions have often been described. However, only few authors approached the physiological evolution of leaf buds all along the dormancy period according to the temperature pattern. The aim of this study was to characterize the evolution of peach leaf bud dormancy through some physiological and biochemical parameters under temperate winter conditions and under total cold deprivation after the endodormancy onset. Two treatments were applied in peach trees cv. Redhaven: (i) Regular Chilling Amounts — RCA and (ii) Total Chilling Deprivation — TCD. Buds were sampled periodically from different parts of the stem (terminal, medium and basal ones). We recorded the evolution of: carbohydrate concentrations (glucose, fructose, sucrose, sorbitol and starch), respiration rate, water contents and energy metabolism (ATP and ADP ratio). The dynamics of these parameters were compared and correlated with dormancy evolution ("one node cuttings" test) and budbreak patterns in plank:. The endodormancy intensity of terminal buds was significantly lower than those of median and basal buds in early October. Under RCA treatment, this gradient faded and the bud endodormancy release was completed at the same time in all positions along the stem. Thereafter, the "cuttings" test indicated that terminal buds grew slightly faster than median and basal buds, and, consistently, budbreak in planta started with the terminals buds, followed by the medians and then by the basal ones. The carbohydrate contents showed a transitory change only when the buds began to grow after the endodormancy was released under RCA. Respiration, water content and ATP/ADP changed dynamics only under RCA and only after the end of the endodormancy (their respective changes were very parallel). The dynamics of none of the tested parameters could be related with the endodormancy dynamics, but respiration, water content and ATP/ADP could be consistent markers of the actual bud growth before bud break (in this respect, ATP/ADP could not show differences between the terminal and axillary buds while respiration and water content could).

Comparison of Xanthomonas arboricola pv. juglandis isolates from walnut trees grown in Romania and Hungary
65-69.Views:286Among diseases that affect walnut, bacterial blight is considered the most important one in all walnut growing areas both from Hungary and Romania. For the determination of susceptibility/resistance of walnut cultivars in our posterior work planned, 61 bacterial isolates were collected from walnuts showing symptoms of blight, with the purpose of isolating Xanthomonas arboricola pv. juglandis (Xaj). The characteristic Xaj colonies, frequently present as saprophytes in the infected plant tissues, were separated from other bacteria according to their morphology, yellow colour, hydrolysis of starch, and oxidation of glucose. All isolates were tested for pathogenicity by hypersensitive reaction on tobacco leaves (Nicotiana tabacum L.), bean pods (Phaseolus vulgaris L) and unripe nuts (Juglans regia L.). Determination of taxonomy of the selected isolates denotes a possible subdivision (races, biotypes) of Xaj occurring in different geographical areas, API 20NE and API 50CH kits were used. Hungarian and Romanian strains showed a high degree of similarity of carbohydrate utilization but slightly differed from the type strain. All were Cu-sensitive.

Influence of foliar nutrition on apple production
15-18.Views:141On the basis of different experiments the technology of intensive foliar nutrition was studied in apple orchards in the period from 1996 until 2000. Yearly apple nutrition programs based on soil analyses. The foliar nutrition program was prepared in accordance with the soil, fruit analyses and climatic conditions. If needed, certain improvements were made according to the analyses of leaves and precipitation in the current year. The richness of the soil and ratios between individual nutrients gradually improved. The foliar nutrition influenced the yield quantity, quality and flower bud differentiation. The yield was doubled and the mean of five years reached 70 tons per hectare. In spite of high yields there were no problems with alternative bearing. The analyses of soluble solids, firmness, fructose, glucose, sucrose, malic acid, citric acid in the fruits and mineral soil composition indicated that the produced fruits were of high quality despite greater yields.

Impact of boron foliar fertilization on annual fluctuation of B in sweet cherry leaves and fruit quality
27-30.Views:322The goal of the study was to examine response of sweet cherries (Prunus avium L.) to boron (B) fertilization. The experiment was conducted during 2005-2007 in West Hungary on mature cv. `Germersdorfi 3' grafted on Prunus mahaleb rootstock.
Sweet cherry trees planted on a calcareous chernozem soil. Trees were foliar-fertilized with B. Foliar B sprays were performed: (1) in the spring, at the stage of white bud, beginning of flowering (B1), and (2) repeated 5 weeks after full bloom (B2). In each of spring spray treatments, B was applied at a rate of 0.15 kg ha-I. Trees untreated with B served as a control.
The results showed that B fertilization had effect on B concentration in leaf tissues, mostly after ripening. B was present significantly higher amount in leaf in treated samples after ripening.
Mean fruit weight was slightly increased by B fertilization. Fruit sensitivity to cracking was not influenced by B fertilization. Nevertheless, from our data it can be conclude that the sensitivity of fruit to cracking is improved when the fruit is riper, the fruit density and fruit weight are higher. The soluble solids varied between 15.0 and 15.9% according to the treatments. Our results for the monosaccharides investigated varied between 5.1 and 7.2 as glucose and fructose as well. Galactose and sucrose was detected very small amount in the unprocessed cherries. Applied B treatments increased sugar contents but decreased organic acid contents in sweet cherry fruits.
It is concluded that under conditions of this experiment, B fertilization can be recommended in sweet cherry culture to improve fruit quality and their appearance.

Sugar composition of floral nectar in sour cherry cultivars
109-113.Views:210The composition of floral nectar in sour cherry cultivars studied in 1997 at Újfehértó was in agreement with our previous data, the three most frequent sugar components being glucose, fructose and sucrose. Nectar secreted at night is generally more diluted than nectar produced during the day. None of the nectar samples produced at night reached the threshold value (100 mg/ml) of bee visitation. In the majority of cultivars the difference in concentration between night and day nectar is not too high, but in two cultivars, 'Korai pipacs' and ‘Újfehértói fürtös’ the difference is quite significant. Most sucrose was found in the nectar of cvs. 'Érdi jubileum' and ‘Újfehértói fürtös’, but a high amount of sucrose was measured also in the flowers of 'Korai pipacs' and 'Meteor USA'. Nectar concentration varies from opening of the flower to petal fall to a smaller or greater degree, depending on the given cultivar. From the 9 sour cherry cultivars studied ‘Újfehértói fürtös’ and Tandy 48' had the most attractive nectar for bees. There was no correlation between nectar composition and free fertilisation. On the basis of nectar composition the majority of the sour cherry cultivars studied can be classified into the sucrose-rich category; only one cultivar, 'Érdi jubileum' had a sucrose-dominant secretory product. The composition of nectar in the studied sour cherry cultivars is preferred by bees.
