Micropropagation of an orchid Dendrobium strongylanthum Rchb.f.61-64.Views:309
A simple and reliable procedure for in vitro propagation of an orchid Dendrobium strongylanthum Rchb.f. was studied. Protochorm was induced from seed explants on 1/2 MS medium supplemented with 0.2 mg/L NAA. A mass of protochorm could be multiplied on proliferated medium of 1/2 Ms containing 0.5 mg/L V-6-BA. And bud differentiation of green global body was cultured in the same media, 2-2.5 cm shoots were formed after 30 day of culture. Addition of mashed banana and 0.5 mg/L NAA to 1/2 MS medium promoted root formation and vigorous growth. The plantlets were acclimatized and transplanted to compound materials of humus:sawdust (1:1) in greenhouse, the survival rate was more than 98%.
The role of rejuvenated and adult forms of English oak (Quercus robur) in in vitro cultures81-83.Views:137
In vitro plant material of clones (Q. robur) NL 100 A (adult) and NL 100 R (rejuvenated) received from Germany (A. Meier-Dinkel, 1995) were used in these experiments. WPM medium was used for the multiplication phase. Plantlets were subcultured monthly. Differences in quality and colour of the adult and rejuvenated cultures induced us to follow and compare the changes of mineral- and chlorophyll content and dry weight during the propagation phase. Mineral and chlorophyll content as well as dry weight were measured weekly on three samples during the subculture period.
In the case of propagation rates we stated, they were similar around the year, but both clones had a high peak in April. Examining the cation-content, we detected that, the plantlets had a highest quantity of several elements during the 2nd and 3rd week of subculture. The iron content was the highest in the 1st week and after that it decreased continuously. It is supposed, that the content of iron is not enough in the media. The chlorophyll content of the rejuvenated clone was higher than that of the adult one.
In the rooting experiments it was stated that, after one-week cold treatment the rooting ability was the best.
Morphological examination of Hungarian apricot rootstock varieties59-63.Views:169
Apricot seedlings are one of the most widely used rootstocks in apricot growing of Hungary. The National List of Hungary contains 7 apricot seedling rootstocks varieties. The production of apricot graft on seedling rootstock takes 3 years from the seed production to the complete graft and requires more space. It is important to provide the trueness to variety name during this process. In the genus Prunoideae not only the plant but also the stone should have typical characteristics. It seems to be useful to examine the morphology of stone. The observed varieties were 'Tengeribarack C.1300', 'Tengeribarack C.1301', 'Tengeribarack C.1650', 'Tengeribarack' C.1652', 'Tengeribarack C.145', 'Tengeribarack C.1426' and 'Tengeribarack C.2546`.1t seems to be the most suitable characteristic to make a distinction between these varieties by stone, the ratio height of stone to lateral width of stone, the ratio of height of stone to ventral width of stone, the ratio of lateral width of stone to ventral width of stone and the ratio of ventral zone to ventral width. From not measured characteristic seems to be most utilizable to make distinction: the fusion of dorsal groove margins, the shape of apex, the presence of a mucro of the apex, the shape of stalk end in lateral view and the texture of lateral surfaces.
The effect of modified bacterial virulence to host-pathogen relationship (Phaseolus vulgaris L. Pseudomonas savastanoi pv. phaseolicola)53-56.Views:166
The Pseudomonas savastanoi pv. phaseolicola is one of the most expressive biogen stressors of the bean (Phaseolus vulgaris L.) in Hungary. The chemical and agrotechnological defence is inefficient, so breeding is the only workable way. The conventional cultivars are susceptible to PS while most of the new industrial varieties have genetic resistance to the pathogen. The genetic background of resistance is, however, a complex system in the bean. Leaf resistance is a monogenic system, but this gene is not expressed in juvenile stage of the host. The pathogen species can be divided into different races. After inoculation with virulent strains, typical symptoms appeared on the leaves. To understand the details of host-pathogen relationships, there were carried out experiments using bacterial strains with altered virulence. Six transposon mutants of the PS were tested. Our main objective was to test these modified bacterial strains on bean cultivars of known genetic background. First we analysed the symptoms, and then the correlation between the symptoms and the multiplication of mutant bacteria. Three cultivars (Cherokee, Inka and Főnix) were tested.
The infection by the virulent PS isolate produced typical symptoms on the three cultivars tested. Mutant bacteria (except strain 756) did not cause any significant symptoms on the hosts. The mutant 756 induced visible symptoms on the cultivars Cherokee and Inka. On Cherokee there were small watersoaked lesions, and HR (hypersensitivity reaction) was detected on Inka, but this was restricted to some cells only (mikro HR). The rate of multiplication of the wild type strain was much higher than the multiplication of the mutants. Bacteria were detected in the cotyledons and primordial leaf, but there is not any substantial number of bacteria in leaves, except for strains 757, 1212 and 1213. The rate of multiplication of strain 756 was intermediate. These, and other experiments can help to understand the genetic background of resistance and the host-pathogen relationship in the Pseudomonas-bean pathosystem.
Determination of auxine content of soft wood cutted `Marianna GF8/1' (Prunus cerasifera x P. munsoniana) by High Performance Liquid Chromatography during rooting period67-70.Views:123
The content of different auxins of soft-wood cutted plum rootstock 'Marianna GF8/1' (Prunus cerasifera x P. munsoniana) was determined during the rooting period. The level of auxin-concentration (exogenous and endogenous) of basic and intemodal part of cuttings was determined by WATERS HPLC equipment every 7 days during rooting period. The lengths soft wood cuttings were app. 30 cm long. The basal part of shoots were treated with 2000 pg/g concentrated indole-butiryc acid in talcum powder. After treatment the cuttings were placed in propagation green-house under intermittent mist. The plant hormones were extracted by methanol the solution was cleaned by paper-filter, and further cleaned by centrifuge. The effluent was examined by reversed phase High Performance Liquid Chromatography, with WATERS 2487 dual detector at 220 am on Symmetry C18 4,6x 150 column. Recovery and reproducibility assessment indicates good accuracy and acceptable relative standard deviation (RSD) 5%. Linear responses (r20.997) for calibration curve was obtained with IAA, IPA and IBA standard in range, with a limit of quantification of 0.15 g•m1-1. The concentration of IAA, IPA and IBA in the basal part of cuttings were measured, during the rooting period. We proved the external IBA was taken up by the plants. In the plants were found the IBA, and the IAA concentration of IBA treated cuttings was higher, than the untreated one.
Anatomical study of the bud union in „Chip" and „T" budded 'Jonagold' apple trees on MM 106 rootstock27-29.Views:338
The traditional methods for vegetative propagation of apple and its varieties are the T-budding, and the winter grafting, but this latter way is a difficult and expensive procedure.
In our experiment carried out in the Fruit Tree Nursery Soroksár, the healing process of chip- and T-budded apple trees 'Jonagold' on MM 106 rootstock was studied.
The budding (T- and Chip-) was made in the first week of August, samples for microscope examination were taken monthly after this time until leaf fall.
The investigated part of plants was made soft with 48 % HF (hydrogenfluoride), then cross and longitudinal section were made and examined by microscope.
Based on analysis of microscope pictures in case of Chip-budding, it was established, that development had started quickly after budding on the rootstock and scion too. But the callus originated almost entirely from the rootstock tissue as new parenchyma cells fills the gap between the two components of graft (scion and stock), becoming interlocked and allowing for some passage of water and nutrients between the stock and the scion. This quantity of callus in case of T budding was under the scion buds larger, than the Chip-budded unions, where the thickness of callus mass is uniformly thick round the chip. The large mass of callus pushes the scion bud outwards from the shoot axis, which later results in a larger shoot-curvature above the bud union.
Following this process on the Chip-budding it can be observed also, that a continuity of the cambium is established between bud and rootstock. Then the newly formed cambium started typical cambial activity, forming new xylem and phloem.
Later the callus begins to lignify, and it is completed within about 3 months after budding.
Changing of carbohydrates by inoculation of Pseudomonas savastanoi pv. phaseolicola oil bean lines with different resistance82-85.Views:134
The Pseudomonas savastanoi pv. phaseolicola (PS) is one of the most significant stressors of bean (Phaseolus vulgaris L.). Chemical and agrotechnical treatments have minor importance, so breeding has great part in the protection against this pathogen. Most of the cultivars are susceptible to PS. The genetic background of resistance in the plant is a complex system. Leaf resistance is a monogenic system, but there are some modifier genes. The pathogen species can be divided into different races.
To understand the functioning of this resistance gene, experiments were carried out using bean varieties with different genotypes and near isogenic lines of bean. Eight lines were tested. Our main objective was to test bean lines with PS with high virulence.
The experiment was made in greenhouse and on field. The virulent bacterium strain has been isolated in Hungary.
The changes of carbohydrates were tested after infection. In homeostasis the level of carbohydrates (especially glucose and fructose) were higher in susceptible lines. In case of artificial and natural infection the decrease of glucose were more significant in susceptible lines than in resistant lines. In the leaf samples from systemic chlorosis the level of this carbohydrate increased.
These changes are connected with the level of resistance, but more experiments are needed to verify this assumption.
Changing of inert content values of apricot varieties39-41.Views:162
The consumers have been exigent increasingly about excellent quality. The speci fie flavour and fragrance of apricot evolve during ripening. We need to be acquainted with changing of inert content during ripening to the choosing of optimal harvest time. The Regional University Knowled ge Centre -Research and Development in Foodstuff Chain project had presented an opportun ity to study the inner content of apricot. Our aim was to study evaluation of health care attributes of apricot fruit. We investigated different ripeness samples of four apricot varieties ('Mandulakajszi', 'Pann 6nia', 'Gonci magyar kajszi'. 'Harcot') in our study of 2007-2009. Samples were taken the orchard of Sóskútfruct Ltd. Hungary. We had measured the water soluble antioxidant capacity, polyphenol-, f3-carotene content and the dissimilar sugar and acid-fractions during ripening. We had experienced significant different in inert content between varieties.
Growth Rings in the Stem of Thuja orientalis L.22-27.Views:119
During the winter dormancy period of 1998-99, the differentiation process of xylem and the formation of annual and pseudo-annual growth rings was studied in sections of the central stem of Thuja orientalis of different age (1-14 year old), starting from the top (the tip of leader shoot) towards the bottom. In the apical 1-2 cm portion of the one year-old leader shoot, only the protoxylem was formed by the end of vegetation. The protoxylem elements appeared first in 6 bundles than gradually merged into 2 semiquadrangular (triangular) bands (each containing 3 protoxylem bundels) around the pith. In this stem portion, the pith was cross-formed first and became gradually flattened at the lower stem parts, following the generally flattened shape of the stem and the respective facial and marginal position of leaves. A continuous xylem ring (with formation of metaxylem elements) apeared 3-5 cm below the shoot tip. In fact, it was the stem part where a "real" annual ring was formed by the end of vegetation. The first pseudo-annual rings were observed 16 cm below the top. The "regular" annual rings were completely continuous all around the stem, consisted of strongly flattened in radial direction thick-walled latewood tracheids and had a distinct border (demarcation line) at the end of the year. The "pseudo-annual" rings formed incomplete dark rings or semicircular bands within the earlywood. They were composed of tracheids with thick cell walls but somewhat wider radial diameter than those of the "real" annual rings, and the border between their outside margin and the next earlywood was less distinct.
In the xylem of two year old stem portion, the innermost central annual ring appeared not at the transition zone between the current and the former years of growth, but about 2 cm lower. Above that, only the thick-walled bundles of the former years protoxylem were found. Down the stem, the older sections showed similar features: the next annual ring appeared always somewhat lower than the borderzone of the given and the former years growth. The "pseudoannual" rings (or more correctly the growth rings) continued regularly to appear in the lower (older) sections of the stem as well. They were found untill the age of up to 14 years (the bottom of the studied plants). Their number was 3-4 per year first, than (from the 5th annual ring counted from the centre) decreased to 3,2,1, and in the youngest outer part of the xylem there was no pseudo-annual ring at all. The development of pseudo-annual rings was usually more marked on the thicker (more branched) than on the thinner side of stem. Stems older than 14 years were not studied.

In this paper we introduce our results of three years (2007-2009) investigatio ns carried out in the framework of "Research and development in foodstaff chain" - project of Regional University Knowledge Center by the members of Corvi nus U niversity of Budapest - Department of Pomology and. The main objectives of the project were the followings: submi ssion of sour cherry candidate suitable for industrial process for state approval; determination of physica l parameter and inner content value changes of sour cherry varieties duri ng ripening; evaluation of health care attributes of sour cherry fruit; ripen ing process description by the colour and the force required to pick fruits of sour cherry. We stated that the optimal begi nning and period of the harvest can be determined wi th the fruit removal force. In this period the fruit growth stops. ju iciness rat io does not change. refractions are approximately equal.acidic content turns to decreasing trend.However the proportion of anthocyan in and polypheno l can still increase. Fruits harvested in this period fulfil a wide range of industrial process opportunities. Further information to the acclimatization of "in vitro" plants54-58.Views:142
The experiment was carried out with in vitro propagated 'MM 106' apple-rootstock plantlets. The transpiration of the plantlets was examined, and the changes followed by SEM analysis.
Data about the transpiration intensity of the acclimatized plants, of its value under different conditions of relative humidity and influenced by the existence of roots, as well as by the degree of acclimatization are presented.
Leaves were also examined and it was found, that stomata of in vitro developed leaves closed slowly, and the number of stomata of newly developed leaves decreased.
It is also shown, that in vitro propagated roots, generally, lose their hairs during acclimatization, but these roots are all the same important, as new roots of full value develop out of them.
The effect of timing and IBA treatments on the rooting of plum rootstock hardwood cuttings7-10.Views:209
In propagation of plum by hardwood cuttings, the success of rooting is affected by several factors. Many authors deal with the timing of cutting collections, others investigate the optimal extent of hormonal stimulation. However, there is no data, as yet, about the coherence between these two factors. The aim of this experiment was to find the most advantageous condition s, with regards to both timing and IBA doses, for rentable propagation of the examined varieties. Five varieties were studied: 'INRA Marianna GF 8- 1', 'Myrobalan B', 'MY-KL-A', 'INRA Saint Julien GF 655/2' and 'Feher besztercei'. Cuttings were collected on seven occasions, during late autumn and winter, and were treated with IBA solutions of different concentrations. The optimal dose of IBA was found to be dependent both on the variety and the actual date of cutting collection . Results are reported, along with suggestions for optimal doses and collection periods.
Dynamics of the uptake of nutrient elements from the medium of in vitro cultured apple rootstocks50-53.Views:141
Cation uptake of J-TE-F apple rootstocks propagated in vitro was studied by the analysis of culture medium. Test-plants were grown on liquid medium under different light regimes. Samples for tests were taken twice a week.
Media without plants served as controls. The analysis of those showed, that only the uptakeable iron-content changed depending upon light treatment. The concentration of all other cations was considered unaltered.
As a result of analysis, it could be established, that elements present in the media were taken up in different rates by plantlets: Cu, P and Zn were utilized totally, but only 50% of K and 20 to 40% of Ca and Mg were taken up under the light treatments applied.
The dynamics of the uptake process was also observed. It was registered that they differed in the case of some cations. So Ba was utilized at the beginning of subculture, others for example B in the later phases. Some elements disappeared unevenly so K, P but the whole quantity is taken up during subculture.
