  • Effect of selenium supplement on proteome of chicken liver

    The aims of the present study were to optimize a two dimensional polyacrylamide gel electrophoresis method to chicken liver proteome and to determine the changes of protein expression caused by selenium. Twelve broiler chicken were used in this experiment. The selenium intake was 0,2 mg/kg in the control group (6 chicken) and 4,25 mg/kg in the experimental group (6 chicken). Using the optimized proteomic approach, we have succesfully separated 747 proteins in the experimental group and 741 proteins in the control group. We found six proteins, wich expressed only in the samples of experimental group. Further investigations need to determine these six proteins with mass spectrometry (MS) and look for the correlation between the physiological effects of selenium and the expression of these proteins.

  • Analysis of longevity in Holstein Friesian cattle using proteomic approaches

    The aim of the present study was to determine marker proteins those are associated with functional longevity of dairy cattle. Holstein-Friesian cows were grouped based on their performance as follows: group 1) individuals with good longevity traits; group 2) short production life because of poor reproduction traits; group 3) short production life with low milk yield. Twelve individuals were sampled in each group, blood and milk samples were collected from cows. Blood samples were analysed with two dimensional polyacrylamide gel electrophoresis (2D PAGE), MALDI TOF/TOF and nanoLCMS/MS. The milk samples were analysed with MALDI TOF/TOF and nanoLC-MS/MS. Using the optimized gel based proteomic approach,
    we have succesfully separated 143 proteins in the group1, 139 proteins in the group2 and 136 proteins in the group3, but we could not find significant differences between groups in the expression pattern. Using MALDI TOF/TOF and nanoLC-IonTrap MS, we have found eleven protein sequences those were expressed only in the samples of good longevity group.

  • The relationship between gluten proteins and baking quality

    Wheat, one of the most important cereals, is grown on the largest area in Hungary. During hydration of storage proteins of wheat – gliadin and glutenin – the gluten complex is evolved. The gliadin is responsible for the extensibility of gluten complex as well as the glutenin for the strength of gluten. The structure, composition and rheological properties of gluten proteins influence significantly the baking quality. The gliadin/glutenin ratio and the quality and structure of glutenin fraction play the most important role in evolving gluten complex. Changes in the steps of breadmaking technology also have effect on the quality of product. Several tests proved that the higher glutenin content increases the strength of dough while the higher gliadin content increases the extensibility of dough and decreases maximum resistance to extension. The monomer gliadins play a great part in plasticity of glutenin polymer. The quality of glutenin fraction significantly influences the evolving gluten complex, because of the spiral structure of glutenin which deforms under stress conditions, then the β-spiral structure resumes their original conformation by releasing from stress.
    The final quality of product evolves as a result of complex characteristics of wheat proteins, so detailed knowledge on the roles of different protein compounds is the base of the quality oriented product development.

  • The composition of gluten proteins and their effect on the rheological properties of gluten

    Wheat is the major cereal component of bread in the world and is grown worldwide. Of the cereals only the bread wheats – and less the triticale – includes storage proteins that play an important role in the performance of gluten. Proteins of gluten complex may be present in two classes:
    − low molecular weight (gliadin-) components, and
    − high molecular weight (glutenin-) components.
    Gliadins shown appreciable heterogenity and can be separated into 40-50 components with gel electrophoresis. The composition of gliadins is employable for the identification the wheat varieties and to investigate the varieties. In the decreasing electrophoretic mobility sequence may be distinguish α-, β-, γ- and ω-gliadins. A glutenin subunits may be include in two classes:
    − high molecular weight glutenin subunits (HMW-GS),
    − low molecular weight glutenin subunits (LMW-GS).
    Wheat varieties can be identified by glutenin and their quality selection is also possible. The gliadin’s polypeptides encoding genes are located on the short arm of chromosomes 1A, 1B and 1D, 6A, 6B and 6D. Genetic coding for HMW subunits is located on the long arms of chromosomes 1A, 1B and 1D, the LMW-GS are also located on chromosomes 1A, 1B and 1D (Glu-3 loci) near the gliadin-coding loci.
    Storage proteins affect the rheological properties of gluten by two factors:
    1. The quality and quantity of the protein components of the gluten complex,
    2. The interactions between the protein fractions.

  • Investigation of the milk composition of three Tsigai sheep varieties, and the detection of cazein fractions of sheep milk by Reversed-Phase HPLC

    The measurements started in 2005, the author used the milk of 3 types of Tsigai Sheep that are suitable for milking, they examined the milk composition, the possible differences between the milk types, the coagulation characteristics, cheese processing, yielding characteristics and the transfer of solid content into cheese. From the large number of measurements, the results of milk composition examinations are described.
    During the research the author examined the milk coagulation property of three types of Tsigai Sheep QTS 25 is a substance investigating instrument, and its primary mission is the analysis of the substance profile. The advantage of this instrument is the representation of the survey graphs from which the part of the substance characteristics could be directly read off. By the help of the applied mathematical and statistical operations we tried to establish connection between the congealment-hardness and the specialty of the different types of Tsigai Sheep.
    In 2006 the author began the identification and qualification of the major sheep milk proteins by RP- HPLC method. She used a scientific article written by Bordin et al. The method is able to separate and quantify the seven major proteins. However Bordin’s procedure was developed using bovine milk, the author adapted it to sheep milk.

  • Effect of the claw disorders on production performance in dairy cattle

    The aim of the study was the influence of claw disorders on production traits in dairy cattle. Observed were claw traits as claw angle, claw length, heel index, claw height, claw diagonal and claw width. Right hind claw after functional trimming was evaluated. Occurrence of claw disorders like interdigital dermatitis and heel erosion (IDHE), digital dermatitis (DD) and sole ulcer (SV) as well as the progress of disease were observed. Basic summary and variation statistics was performed by the SAS software. Two herds were included in the study. Holstein dairy cows (n=101) produced 11 875 kg of milk, 468 kg (3.94%) of fat and 396 kg (3.34%) of proteins. Holstein cows were affected with IDHE in 22 cases, with DD in 6 cases and with sole ulcer in 10 cases. Slovak Simmental dairy cows (n=101) produced 5834 kg of milk, 258 kg (4.44%) of fat and 209 kg (3.59%) of proteins. Slovak Simmental cows were affected with IDHE in 8 cases, with DD in 5 cases and with sole ulcer in 19 cases. Observed was that claw disorders caused deviations of milk production.

  • Microbiological and Chemical Characterization of Different Composts

    Composting of agricultural waste is considered particularly important from the point-of-view of environmental protection. Degradation of organic substance results in a significant reduction of waste volume.
    The end product of the composting process, mature compost, can be used as soil coverage against excess loss of wastes, for mulching, for organic manure etc. The problem of composting has come into limelight in environmental studies and in agriculture.
    The quality of the mature compost is determined by physical, chemical and biological parameters of the composting process which, in turn, depend on initial composition of the raw materials, the technology, e.g. regular mixing and moistening and on environmental factors. Quality is the key question in compost use.
    We studied the composting process in compost windrows of different raw material composition. We measured temperature, humidity content, pH, organic substance content, nitrogen and carbon content.
    We counted the number of bacteria, microscopic fungy, ammonifying and cellulose decomposing microorganisms. We directed the composting process with turning weekly (to provide oxygen) and watering (to provide humidity content 40-60%).
    We set up windrows of 1 m3 volume from dry plant substances (cornstalk, pea straw, tomato stalk and crop, weeds) and cow manure not older than 1 week. The cow manure was used at ratios of 0%, 35%, 50%, 65% and 100%, respectively.
    We measured changes in compost temperature relationship with outside temperature until they were almoust the same. Humidity was 40-60% in most cases.
    At the beginning of the process, pH was slightly acidic-neutral; it later becomes neutral-slightly alkaline (pH: 6.93-8.02) as ammonia is liberated from proteins.
    At the end of the process, pH decreased again, due to humification.
    Organic substance content decreased as microorganisms mineralized them. Organic carbon content decreased gradually due to microorganisms used it as an energy.
    Total nitrogen content increased until middle of july and decreased gradually until than.
    The carbon/nitrogen rate were higher in the beginning, it decreased until july-august and increased by smaller degree until end of the process.
    The number of bacteria was higher in the first three weeks and between june-september. The number of cellulose degrading bacteria was the highest in the first three month, the number of ammonifying bacteria was the highest from the end of may until sepember.
    The number of microscopic fungy was significant in the second part of process, after july.

  • Comparison of alveohraph and extensigraph properties of winter wheat samples

    The rheological properties of dough are determined by the amount and proportion of gliadin and glutenin proteins. Extensibility and resistance to the extension of doughs which can be measured using the extensigraph or alveograph allow good assesment of the baking behavior under realistic conditions. In our study, we compared the extensigraph and alveograph parameters of 87 flour samples. The results showed that the alveograph parameters were different from the extensigraph parameters. The classification of wheat varieties was different, and the order of varieties, as well. The alveograph properties were affected by other factors than the extensigraph properties, except the W value. The two rheological tests did not give similar results, therefor neither test are replaceable.

  • Fractionation of chicken egg proteome by isoelectric point in liquid phase

    The application of proteomics is relevant to physiology, reproduction, immunology, muscle and lactational biology in animal science, altough its use is still limited. One of the greatest challenges of proteome analysis is the reproducible fractionation of the complex protein mixtures. The fractionation methods can increase the probability of biomarker protein discovery. The fractionation by liquid-phase isoelectric focusing is one of the prefractionation methods. As a result, protein fractions can be easily collected, pooled and refractionated. There is a lack in the knowledge of gel-based proteomic methods of egg as only a limited number of protocols can be found in the literature, thus sample purification and fractionation require a time consuming optimisation procedure. The aim of this study was to fractionate egg yolk and white proteins by isoelectric point in liquid phase.

  • Alternative protein sources in the nutrition of farm animals

    Protein requirement and its demand of farm animals became one of the critical problems in nutrition on a global scale. Protein requirement has been an explicit demand for a long period with soybean meal and animal protein, but recently there are some limitations in relation to their use and the availability of the high quality fishmeal decreases constantly. For this reason there is increased demand for finding new protein sources which could be the alternatives of soybean meal and fishmeal. Alternative protein sources can be divided into seven categories, according to their origin. In different countries, their use depends on the availability in large quantity and at reasonable price.

    There is a long tradition of using legume seeds, as alternatives of soybean. Most of them contain some anti-nutritive compounds, but it can be reduced with systematic selection. Oilseed meals are also generally use in poultry and pig nutrition, but those crude protein content varied, depending on the oil extraction technology. Green fodder and leaf protein was also proposed as alternative protein sources, but their use is limited, in particular because of the market price. The amount of bioethanol and starch industry by-products increases gradually in recent years, therefore those became alternatives of soybean meal, or in much less extend, fishmeal. However, amino acid composition of such by-products are far from optimal for poultry and pig; therefore, in the case of their use amino acid supplementation is necessary. Several novel protein sources are proposed in the last decade, such as algae or insect proteins. Recently, their availability and use is limited, but in the near future those would be alternative protein sources in monogastric animal nutrition.

  • Experiment of quality properties of dehydrated fruits

    The lyophilization is the joint application of freezing and drying. It is an up-to-date conserving procedure, the point of which is that the humidity existing in the frozen humid material is transferred from the solid state directly into the gaseous state at a temperature below 0 oC under vacuum. Out of the procedures applied nowadays, this is the most tolerant drying process.
    With regard to the high investment and operational costs, freeze drying is applied only for valuable, heat-sensitive materials when the technological aim is to preserve such properties as aroma, taste and colour as well as such components as proteins and vitamins. This procedure is suitable for drying and conserving certain foodstuffs, stimulants, organic chemicals, medicines and similar sensitive and valuable materials.
    In our institute, we have been conducting freeze drying experiments with regional fruits and vegetables since the year 2005. During the first phase, we examined the heat- and material transfer as well as the abstraction of humidity, while during the second phase we analysed the rehydration ability and nutrient content of the freeze-dried materials as compared to those dried with the method of convection. Moreover we have conducted penetration measurements with a portable hardness tester.
    To sum up the results gained so far, we can state that the quality of the lyophilized materials is better than those dried in the traditional way. It originates partly in the fact that the temperature and pressure applied for the freeze drying are smaller and the drying period is far longer than for the convection drying.
    In contrast to convection-dried materials, freeze dried materials set in close to their original water-content, keep their original shape and size after being rehydrated. The reason of it the porous, spongy structure (flexible cell wall) of the lyophilized products which is able to take up moisture quickly. In addition, the lyophilized products can be rehydrated faster than those dried in the traditional way.
    Regarding the results of the chemical analyses, the following conclusion can be drawn: the vacuum freeze drying results a small decrease of nutrient content and nutritive value for the lyophilized products.
    The results of the hardness tests support the statement that the majority of agricultural materials cannot be considered as an ideal flexible body, because during the experiment the flexibility coefficient changed when going from the surface of the material inwards. In addition, the penetration tests also confirm that the surface of the convection-dried vegetables is at least 1.5-3 times harder than that of the freeze-dried products. The reason of it that it takes place during the drying denaturation processes.
    The article summarizes the results of our research work listed above, in accordance with our experiments conducted by using the characteristic fruits (apple, plum) of the Nyírség Region.

  • Development of seed analyses by means of various matrix solutions and the MALDI-TOF MS technique

    The earth's population is growing steadily, currently accounting for about 7.3 billion people. Population growth causes food demand to rise, approximately 36 million people die each year due to starvation or related diseases. One solution to this problem is the continuous examination and development of the agricultural economy. In this study, matrix-assisted laser desorption/ionization time-of-flight mass spectrometer (MALDI -TOF MS) were used to analyse of sunflower, soybean and hemp. In order to analyse the protein of maize, this method has already been applied. However, for sunflower, soy and hemp, it is necessary to develop a sample preparation method. Choosing the optimal matrix solution for ionization the traget molecule is an essential part of developing the method. Our aim is to compare two different matrix solutions (α-HCCA, SA matrix), based on the properties (intensity, noise ratio, value of spectra) of the spectra.

  • Alternatives of synthetic chemicals – chemicals derived from foods and related materials

    Foods are composed of numerous chemicals, each of them work individually or synergistically inside the living organism for a specific reaction supporting the life processes. To date, the efficacy of these chemicals in industrial processes has been explored to a limited extent. In fact, chemicals derived from food have great potential to be used in multi-disciplinary sciences and in numerous applications. Synthetic chemicals might be replaced by the natural resources in the future which will also reduce the environmental impact of hazardous and toxic chemicals. The present article gathers a few examples of chemicals derived from food or related materials and shows how worthy they are in various fields, ranging from radiopharmaceuticals to alternative mining of precious metals. In a nutshell, the article puts forward a few simple ways through which research may adjoin the greener mandates and help in maintaining the sustainability of our environment.

  • Effect of agrotechnological treatment on nutritive value of common millet

    The cultivation of common millet shows an increasing tendency due to its adaptability to extreme poor circumstances. In addition, millet do not contain gluten forming proteins such as gliadin and glutenin, therefore people with gluten intolerance could consume it. The vitamin B and mineral content increase the importance of millet. The aim of our experiment was to measure the quality of flour of two millet varieties treated with different nitrogen fertilizer doses.

  • Application of the SDS-PAGE method for the characterisation of winter wheat flour quality

    The principle, development and importance of the SDS-PAGE method are presented in this article. The SDS-PAGE method has become one of the basic methods of molecular biological research, because it is widely applicable and its sensitivity is excellent in the separation of wheat storage proteins.
    We have shown the application of this method with a concrete example. It was also tested whether, it was possible to obtain a better baking quality product from a large amount of poor quality less valuable wheat by fractioning the flour according to particle sizes after grinding. We studied the rheological properties of flours with different particle size fractions from the original flour. The baking quality of the original flour was B2. The 125-90 and 90-63 μm fractions have significantly better baking quality (B1) than the original flour. The protein contents of these flour fractions were also significantly higher than the protein content of the original flour. We had a question: what has influenced the baking quality: the protein content or other factors? We searched for an explanation on these results in the protein composition of the flour samples. We studied the distribution of glutenin-fractions by SDS-PAGE method and evaluated them. We found with correlation analysis that the amount of LMW-Glutenin D-group (52-60 kDa) is in a strong, negative correlation to the baking quality (r = – 0.855*). Therefore, the baking quality of flour samples was influenced by this glutenin fraction.

  • Application of yeasts fortified with microelements – Review

    Microelements are increasingly becoming into the focus of interest from both a point of view of nutrition science and feeding. An always growing care must be paid to the microelements coverage both in human and animal organisms because of incorrect alimentation habits and
    unsatisfactory feedstuff nutrition value. For the increased supply of the micro-nutrients, enrichment or fortification with microelements can not only be realized with traditional foodstuffs and forage but there are already alternative ways such as single-cell proteins from yeasts directly enriched or fortified with microelements for the purpose.
    We would like to draw the attention that the production of these items is more favourable in comparison with traditional foodstuffs or forage since yeasts are capable to multiply microelement levels compared to their original state, and establish organic bonds with them.
    For this purpose, we explored and analysed the scientific literature, studies and research results on this subject, that is why we stressed the significance of yeasts, the features and health effects of certain microelements, as well as the possibilities for use of yeasts enriched with micro-nutrients.

    The food allergy is a hypersensitivity reaction against naturally occurring proteins in food. These types of disease can cause not only personal inconvenience to the patient but serious health, food safety and food analysis, social-economic problems. The only effective treatment for these illnesses is a life-long diet avoiding the allergenic foods or components of food. In the interest of the patients’ health 14 allergenic components must be labeled on the food packaging. To meet the requirement of regulation reliable and valid analytical methods are necessary which for the most allergenic foods are not available.

  • How does the S-locus determining self-incompatibility in stone fruits work in self-compatible peach?

    The majority of stone fruit species are self-incompatible, a feature that is determined by a specific recognition mechanism between the S-ribonuclease enzymes residing in the pistils and the F-box proteins expressed in the pollen tubes. Failure in the function of any component of this bipartite system resulted in self-compatibility (SC) in many cultivars of Prunus species. Peach (Prunus persica (L.) Batsch.) is the only species in the Prunoideae subfamily that is traditionally known to be self-compatible, but its molecular background is completely unknown. Isoelectric focusing and S-gene specific PCR revealed that SC is not due to functional inability of pistil ribonucleases. We hypothesize that SC may be a consequence of a kind of pollen-part mutation or the action of one or more currently unknown modifier gene(s). Only two S-alleles were identified in a set of peach genotypes of various origin and phenotypes in contrast to the 17–30 alleles described in self-incompatible fruit trees. Most important commercial cultivars carry the same S-allele and are in a homozygote state. This indicates the common origin of these cultivars and also the consequence of self-fertilization. According to the available information, this is the first report to elucidate the role of S-locus in the fertilization process of peach. 

  • Impact of lameness on the milk production of ewes

    The aim of this study was assessing the impact of lameness on the milk production, somatic cells count and component of milk. We assess also impact of lameness on the order of entry into the milking parlour.

    The experiment was carried at the farm, located in northern Slovakia. The farm keeps sheep Improved Valachian. Samples of milk were taken during two periods: May, July. It was taken 428 samples together. We recorded three groups by lameness- strong lame, slightly lame, non-lame ewes. We recorded also the order of entry of ewes into the milking parlour in milking row. The results were mathematically processed using the Microsoft Excel program and statistically evaluated by SAS.

    We found significant statistical differences between months (P<0.0001) in all the above mentioned indicators. In July we recorded 26 ewes with slightly lameness and 18 ewes with strong lameness. Other ewes were non-lame. Non-lame sheep had in July the highest milk yield (356±148 ml) and the lowest decrease in milk yield from May to July (-206±131 ml) compared with slightly (317±116 ml, -223±163) and strong (319±122 ml, -219±151 ml) lame ewes. However, these differences were not statistically significant. We have not identified statistically significant differences between groups in somatic cells count (logxSCC for non-lame: 4.83±0.608 in ml, slightly lame: 4.76±0.653 in ml, strong lame 4.71±0.787 in ml). Milk composition (fat, proteins, lactose) nor changes in the composition of milk that occurred between May and July were not affected by lameness of ewes. But lameness in July affected the change the order of entry of ewes in the milking parlour in July compared with the order of entry recorded in May.
