  • Genetic relatedness among Asian Cotoneaster species investigated with DNA marker analysis

    The widespread genus Cotoneaster has its centre of diversity in the Himalayas and surrounding areas. Most taxa appear to be polyploid and apomictic, and many of them have become popular ornamentals due to their attractive foliage and berries. One of the taxonomically most critical groups is Section Alpigeni which contains several important ornamental plants. The number of species belonging to this section varies widely between different taxonomic treatises, depending on whether 'splitting' or 'lumping' of species is preferred. Using a rather narrow species definition, we have investigated 13 different species using RAPD analysis. A simple matching (SM) coefficient-based principle coordinate analysis (PCO) was calculated from the RAPD data. Some species were clearly more similar to each other than to other species in the analysis. The levels of similarity did, however, not correspond very well to the lumping together of several taxa under the same species name as performed e.g. in the recent Flora of China. Obviously, the complex hybridogenous origination and, in some cases, still ongoing recombination with sexual species or among the apomictic taxa themselves, produces a genetic variability structure that cannot be properly reflected in a hierarchical taxonomy.

  • Comparative analysis of sour cherry cultivars on their ecological and biological indicators

    Sour cherries developed in the northern hemisphere, an alloploid hybrid of dwarf sour cherries (Prunus fruticosa) and bird cherries (P. avium), born in the confluence of the two species. However, the ecological and, above all, cold tolerance of the ancestor of cultivated sour cherries is higher than that of wild cherries (De Candolle, 1894; Rehder, 1954; Terpó, 1974; Iezzoni et al., 1991; Faust & Surányi, 1997). The cultivation limits are in the northern hemisphere 38-44. degree. The Carpathian Basin, the Balkans and Asia Minor are considered to be the main birthplaces for sour cherries. The genetic and morphological diversity of sour cherries is greater than that of the basic species (Iezzoni et al. 1991; Faust & Surányi, 1997). In the study, 472 sour cherry cultivars were compared based on 7 relative ecological indicators and 3 biological values. Compared to other Prunus species, we mostly found less variability in sour cherries - not counting their salt tolerance (SB). The partial similarity between open pollination (OP), frost tolerance (FR) and disease resistance (DR) - partly true in terms of varieties, but also reflected the effects of purposeful breeding and selection. The cultivars together - in comparison, showed balance, but in the highlighting, the differences of the 3 cultivar groups became significant. Indeed, the differences between the species of the former Hungarian cultural flora are clearly different (Surányi, 2004), which is also the case when comparing a large number of apricot (Surányi, 2014), plum (Surányi, 2015) and peach (Surányi, 2020) varieties.

  • Evaluation of fruit quality parameters of Rosa taxa from the Carpathian basin

    From the wide range of genetic sources available in Hungary, we have chosen as objective the evaluation of those rose taxa, which — on the base of their quality and morphologic features — are suitable for selecting fruit production varieties or parent varieties for further breeding. We examined physical and inner parameters of fruits of Rosa taxa, and evaluated the correlations by mathematical statistic methods. Namely, if a correlation can be found between physical and inner parameters, fruit production value can be determined by less examinations in the future. According to our results, there is a large variability in some physical parameters (weight, diameter of fruits; weight, number of achenes) as well as in inner content values (vitamin C, glucose, fructose, K, Ca, Fe, Zn and Cu content) of rosehip, regarding production year, habitat and even the individual of examination. Twice as much ascorbic acid can be found in achene-free fruits, and nearly five times as much in their dry product as in fresh whole fruits. It was established repeatedly, that vitamin C concentrates in fruit flesh, and ascorbic acid content can be preserved better in achene-free flesh. There is no essential difference in citric acid and carbo-hydrate content of whole and achene-free rosehips. However, macro- and microelement content of whole fruits is 30-40% higher than that of achene-free fruit flesh. The connection between fresh weight and diameter, as well as achene number and seed weight of fruits can be described by the function y=axb. A significant relationship can be found in case of K-Fe, Ca-Mg, Ca-B, Ca-Mn and Zn-Cu, between fresh weight and B content of fruits. According to our examinations, fresh weight of rose species with a higher citric acid content is usually higher, while their Fe content is smaller. Glucose content of fruits of rose species is directly proportional to their Ca and Zn contents. Zn content of rose species with higher fructose content is usually smaller.

  • Development of microsatellite markers for Rhodiola rosea

    Rhodiola rosea L. is an important adaptogen medicinal plant. In this study two new microsatellite markers were developed. The assessment of the genetic diversity of R. rosea has recently started with molecular markers, but only a few species-specific microsatellite markers have been published so far. However the small number of markers allows only a limited insight into the genetic variability of the species therefore the aim of our work was to develop new microsatellite markers for R. rosea with a microsatellite enrichment library technique. Genomic DNA was cleaved with an endonuclease enzyme followed by adaptor ligation and PCR amplification. DNA fragments that contained microsatellites were first isolated using a biotin-streptavidin linkage based magnetic selection and then cloned into plasmids. Out of forty-three sequenced clones three contained  microsatellites, in these cases primers were designed for the amplification of the microsatellite repeats. The newly developed primer pairs were tested on individuals from distant R. rosea populations and the variability of the amplified fragments was estimated by fragment-length analysis. The locus RhpB14a was found to be monomorphic while RhpB14b and RhpB13 were polymorphic. As a result of the present study, two novel variable microsatellite loci were identified in the genome of R. rosea.

  • Antioxidant capacity and total polyphenol content of Lavandula cultivars at different growing areas in Hungary

    Lavandula species are worldwide grown essential oil producing medicinal plants with considerable economic value. Beside volatile oil, lavender also contains different phenolic compounds which have been less widely studied till now. Cultivation of lavender has become widespread in Hungary in the recent years, however, growers have limited knowledge on the productivity of cultivars available. In our research we were aimed to studying the antioxidant capacity and total polyphenol content of samples collected in two growing areas (Dörgicse and Szomód) from flowers of two L. angustifolia (‘Hidcote’, ‘Munstead’) and two L. × intermedia (‘Grosso’, Grappenhall’) varieties, during the full blooming period of 2017. Antioxidant capacity of the samples was determined by FRAP assay (Benzie and Strain, 1996), while total polyphenol content (TPC) was measured by a modified method of Singleton and Rossi (1965). According to our results, varieties of L. × intermedia showed higher values of antioxidant capacity and of total polyphenol content, than those of L. angustifolia cultivars. Among them, both the antioxidant capacity (179.6 mg AAE/g DW) and total polyphenol content values (152.4 mg GAE/g DW) of ’Grosso’ from Dörgicse were the highest. Concerning FRAP values of all the cultivars investigated, larger variability were found in Dörgicse, than in Szomód. Regarding the effect of growing area, each cultivar represented similar FRAP values in Dörgicse and in Szomód, except for ’Grosso’. However, in the case of TPC values, higher variability was observed between the growing areas, especially in the case of ’Grosso’. In the future our studies on lavender polyphenols will be completed with qualitative evaluation of the values obtained by HPLC analysis.

  • Results in the determination of some Hosta varieties by the method of isoelectric focusing

    Mass propagation of 5 newly introduced Hosta varieties was carried out by the method of micropropagation. Our aim was to determine exact variety specificity after the micropropagation period in the pattern of peroxidase isoenzymes by isoelectric focusing in pH 3-9 range and to determine that phenological phase of mother plant in which the isoenzyme pattern of mother plant can safely be comparable to the isoenzyme pattern of micropropagated descendants. The isoenzyme patterns of descendants were similar to the mother plants of the same hybrid lines. The older leaves seemed to be not so suitable for examination than newly developed ones despite of the higher activity of peroxidase enzymes. There were big differences in isoenzyme patterns of leaves in different phenological phases. With this quick and easy method Hosta varieties could be selected already in the very early stage of micropropagation.

     

  • Introduction of alkanet (Alkanna tinctoria (L.) Tausch), a traditional dye plant into cultivation

    As a part of the research project to establish natural sources of plant pigments, possibilities of introduction of Alkanna tinctoria (alkanet) into cultivation were studied.

    As a result of the germination experiment, the relevance of 21 days' duration of germination procedure was proved. To get high germination rate alkanet fruits are proposed to pretreat by gibberellinic acid (GA3) in the concentration of 400 ppm, overnight before sowing. This method results approximately 50 % germination rate.

    The morphological and production properties of alkanet roots are characterized during ontogenesis. Transplanted populations can be characterized by numerous, thick and heavy roots comparing to the spontaneous ones. Thus, seed sowing and transplantation proved to be an effective method for cultivation of the species.

    According to our results it can be concluded that in cultivation the optimal harvesting time of roots is at the end of the second vegetation cycle, when the dry root mass of the individuals is about 10-20 g with 3,0-3,5 % accumulation level of active substances.

    Considerable seasonal variability have been found influencing not only the root masses, but also the accumulation levels of alkannin derivatives. In a more humid vegetation cycle the root size and mass as well as the content of active substances are much higher.

  • Large variations in antioxidant capacity and contents of Hungarian sour and sweet cherry cultivars

    Sour cherry (Prunus cerasus L.) cultivars grown in Hungary are of local origin while most sweet cherry cultivars in Hungary are introduced from other countries.A great phenotypic variability is displayed by both species. In the present study, we analyzed 10 sour and 9 sweet cherry cultivars for their antioxidant capacity, total polyphenolics (TPC) and total anthocyanin (TMAC) contents. In general, sour cherries showed higher levels of antioxidant capacity, TPC and TMAC. The anthocyanin contents varied from 0.16 to 6.85 and 1.41 to 127.56 mg/100 g for sweet and sour cherries, respectively. However, TMAC generally seems to have a limited influence on the antioxidant capacity of cherries.An amarelletype sour cherry, ‘Pipacs 1’ showed the highest antioxidant capacity (21.21mmolAA/l) and TPC (44.07mgGA/l) in contrast to its lowanthocyanin content. The detected diversity presents a choice that can satisfy different consumer preferences, and meet specific nutritional requirements.

  • Nutritional and seasonal requirements for callus growth in Taxus baccata

    Callus cultures derived from young stems of two varieties of Taxus baccata cv. aureovariegata (genotype I) and Taxus baccata L. (genotype II, III) were induced. Gamborg's B5 medium was supplemented with different concentrations of auxin (2,4-D) in combination with cytokinins (kinetin or topolin) and with a phenolic-binding compound (PVP) to prevent callus darkening and growth inhibition. Stem explants displayed different responses to in vitro culture depending on plant genotype and on the season. Genetic variability was observed in the growth rate of calli initiated from all three genotypes of the same Taxus species. We found the best growth of callus cultures originated from the genotype ill in defined media. After the first subculture the majority of the cream-coloured primary callus turned brown and ceased its growth. However, the long-term culture was initiated.

     

  • Inhibition and recovery of germination and growing ability of seedlings under and after osmotic stress induced by polyethylene glycol in 8 pea genotypes

    Germination and early seedling growth of eight pea genotypes were examined under and soon after different (5%, 10%, 15%, and 20%) PEG treatments. Seeds were germinated on PEG solution for 3 or 6 days and then further germinated and cultured on filter paper moistened by water for 3 and 6 days. The length and fresh weight of shoots and roots of seedlings were measured and used for evaluation of genotypes. Roots were less inhibited by osmotic stress than shoots similarly to other plant species. The variability among the genotypes was the greatest in the case of shoot growth at 5% PEG treatment and in the case of root growth at 15% PEG treatment. Results suggest that growing responses of genotypes after cessation of stress are more suitable for the evaluation of their osmotic tolerance, than their responses expressed during in vitro stress conditions. Genotypes with relatively high or low osmotic stress tolerance, respectively, could be distinguished with 6 days after recovery from 3-day-long 15% PEG treatment concerning the rate of shoot weight to root weight.
