Relation between the germination and infection ratio on Sida hermaphrodita L. Rusby seeds under hot water treatment
117-121.Views:210Sida hermephrodita or virginia mallow is a perspective perennial herb in the Malvaceae family able to yield a biomass crop through between ten and twenty years. Additionally, the plants have a lot of uses and benefits for instance it can use it as a fodder crop, honey crop, ornamental plant in public gardens. It has favorable features like fast growing and resistance against the disease and climatic fluctuations, etc. Sida is in base stage of domestication therefore has a serious disadvantage the low and slow germination as a big part of wild plants. Due to the expressly low germination percent the need of seed showing of driller is should tenfold, 200 thousand seed/acre instead of 10-20 thousand what is not available and expensive Therefore practical purposes of our research of seed physiology was to increase the seed germination percent in a disposable ,basically wild Sida population. We examined two factors relating to seed germination percent and seed germination power during our research: the influence of hot water treatment and the effect of exogenus or endogenus infection of seed. However, in our germination tests, utilizing scarified seeds with hot water (65, 80 and 95 oC), 29,33 to 46% germinated of the seeds collected from the population of S. hermaphrodita in Debrecen. The average germination for all season was 5-10 % wihitout treatment and rised using hot water up to almost 50%. When physically scarified used, the oldest seeds showed the best germination (46 %) after the hot water operation in spite of the previus studys (Spooner 1985; Chudik et al. 2010; Doliński R. 2009.). We discovered that there are a distinguished close relationship between the seeds collecting time and the infection, as well as germination percentage. Thus, 2009 season was the most favourable in case of contamination (control:17,33 and 80 oC treatment:0%) as well as germination percent. It could be concluded that, the best season for our findings was 2009 due to autumn harvest of Sida seeds. In our oppinion, the autumn harvesting should be the best time to overcome the problem of the low germination and high infection percentage.

The development and reproduction of grape phylloxera (Daktulosphaira vitifoliae Fitch) strains of Hungarian vine-districts in root-bioassays
67-71.Views:142The root bioassay method allows for 4-6 week continuous observation of grape phylloxera feeding on the grape root. 10 Hungarian phylloxera strains were compared on susceptible Vitis vinifera cv. Chardonnay and the resistant rootstocks of V. berlandieri x V. riparia Teleki 5C and V. berlandieri x V. rupestris Georgikon 121 in in vitro observations. The strains originated from Villany and Eger (Figure 3) had higher reproduction on the root of V. berlandieri x V. riparia Teleki 5C (201 and 119 eggs) and were more aggressive than the others (average production 10 eggs). The continuous high level of survival, development and reproduction of the Eger strain on Teleki 5C (V. berlandieri x V. ripuria) until day 46 may be due to adaptation.

The Effects of Some Parameters on Agrobacterium-Mediated Transformation in Muskmelon
46-49.Views:184Some parameters involved in Agrobacterium-mediated transformation in muskmelon Hales best (HBS) were studied. Cotyledon explants excised from 3.5-day-old seedlings were co-cultivated with Agrobacterium tumefaciens harbouring binary vectors which contained GUS and BAR genes. After co-cultivation on a low pH medium, explants were transferred to selective medium, with higher pH, containing Claforan and Finale. The medium was changed every two weeks till shoots were induced. All shoots rooted on MS medium supplemented with 0.3 mg/L IBA. These parameters combined as a whole led to successful transformation. The expression of the introduced gene construct was confirmed by GUS staining of shoot segments.

Genetic transformation of bean callus via Agrobacterium- mediated DNA transfer
49-53.Views:130Callus cultures were induced from hypocotyl of young bean seedlings. Callus developed and maintenaned on B5 medium supplemented with 2mg/1 2,4-D and 1 mg/1 kinetin. The results demonstrate that A. tumefacins-mediated transformation is a convenient method to obtain transient gene expression in callus of bean. The results have shown that the bean callus co-cultivated with A. tumefaciens can be transformed to get heibicide Finale (glufosinate-ammonium) resistant GUS positive tissues. Southern blot analysis of transformed calli showed integration of gusA marker gene carried by a binary vector. Transformed calli were selected on herbicide containing media. Data of molecular analysis (Southern blotting) confirmed the insertion of gusA gene in the genome of herbicide resistant calli with bar gene. There are three evidences that calli are stable transformants: (1) herbicide resistance, (2) GUS activity which is indicative since the coding region containing an intron, (3) the results of Southern hybridization technique.

Vegetative Indicators of sour cherry cultivars and resistant clons
55-58.Views:195Sour cherry is a hungaricum. Hungary has several excellent and self fertile sour cherry cultivars. Hungarian sour cherries have a name for fruit quality and they are suitable for fresh consumption. Today breeding is focused not only on fruit quality, but also on disease resistance. The new, investigated cultivars have these characteristics too. But growth characteristics are also very important because they contribute to successful production. The vegetative and generative properties of the cultivars are different. If a cultivar is very productive, the ignorance of cultivar-specific pruning may result in the decrease in tree condition and fruit quality. The properties of producing fruiting wood greatly determine the renewal capabilities of cultivars. Knowing these properties is essential in case of sour cherries, the fruit species that requires a lot of sunlight and tends to produce bare wood. The research of candidate cultivars until now focused on fruit quality. Our aim was to investigate the vegetative characteristics of the resistant ‘bosnyák’ cultivar group in detail, defining the growth characteristics of these cultivars.

Traditional farming within the Carpathian basin - pomaceous fruits
15-18.Views:175In the Carpathian basin there are still maintained the main historical fruit varieties and traces of traditional farming. The Department of Fruit Science considered the utilisation of obsolete varieties in breeding as source of resistance to local adversities, moreover, the practice of traditional .farming using those varieties should be also explored. In the present study, the farming methods and variety structure of two substantially different growing sites are described. One is an alluvial plain along a river were an ancient form of farming based on the control of sluices. The second is practised by clearing the forest on the slopes of the central range and of the Carpathian chain. During the course of our research we succeeded to find 8 pear, 29 apple and a few quince as well as medlar varieties. Further endeavour would be the conservation of those varieties, preferably on the spot together with their growing techniques as relicts, also as aesthetic components of that particular landscape. Our committed partners in this work are the National Parks.
