Aloe vera is one of the most famous medicinal plants. Aloin and aloe emodin are the most important active compounds in this plant. The purpose of this research was the comparison of aloin and aloe emodin production after the elicitation by methyl jasmonate, salicylic acid, and phenylalanine in diploid and tetraploid Aloe vera...plants in greenhouse conditions. The plants were treated with the concentrations of 25, 50, and 100 µM. The amounts of aloin and aloe emodin were determined 24 and 48 hours after application of the treatment. HPLC analysis showed that the leaves of the control diploid plants (without applying elicitors) had more aloin (1.20 fold) and aloe emodin (1.14 fold) than the control tetraploid plants. The maximum concentration of aloin (1.15 ± 0.07 µg mg-1 dry weight) was obtained after the elicitation by 25 µM methyl jasmonate, 24 hours after treatment, in diploid plants) 6.36 fold compared to the control (0.18 µg mg-1 dry weight (. In addition, the maximum concentration of aloe emodin (0.28 µg mg-1 dry weight) was obtained after the elicitation by 25 µM salicylic acid, 24 hours after treatment, in diploid plants) 6.18 fold compared to the control (0.04 µg mg-1 dry weight)). The long-term effect of three studied elicitors (after 240 days) on plant health and survival was also studied. This investigation showed that only methyl jasmonate at a concentration of 100 µM was resulted in the death of Aloe vera plants.
The content of different auxins of soft-wood cutted plum rootstock 'Marianna GF8/1' (Prunus cerasifera x P. munsoniana) was determined during the rooting period. The level of auxin-concentration (exogenous and endogenous) of basic and intemodal part of cuttings was determined by WATERS HPLC equipment every 7 days during rootin...g period. The lengths soft wood cuttings were app. 30 cm long. The basal part of shoots were treated with 2000 pg/g concentrated indole-butiryc acid in talcum powder. After treatment the cuttings were placed in propagation green-house under intermittent mist. The plant hormones were extracted by methanol the solution was cleaned by paper-filter, and further cleaned by centrifuge. The effluent was examined by reversed phase High Performance Liquid Chromatography, with WATERS 2487 dual detector at 220 am on Symmetry C18 4,6x 150 column. Recovery and reproducibility assessment indicates good accuracy and acceptable relative standard deviation (RSD) 5%. Linear responses (r20.997) for calibration curve was obtained with IAA, IPA and IBA standard in range, with a limit of quantification of 0.15 g•m1-1. The concentration of IAA, IPA and IBA in the basal part of cuttings were measured, during the rooting period. We proved the external IBA was taken up by the plants. In the plants were found the IBA, and the IAA concentration of IBA treated cuttings was higher, than the untreated one.
In our work the effectiveness of supercritical carbon dioxide extraction was studied on volatile and nonvolatile components of Lamiaceae herbs. The aim of our investigations was to determine the optimal extraction parameters yielding high amount of volatiles and nonvolatiles in a desirable composition. As plant materials, dried and pow...dered cude drugs of Satureja hortensis, Ocimum basilicum and Melissa officinalis were chosen. Pressure (8-50 MPa), temperature (35-60 °C), time (5-60 min) and the ratio of modifier (5-50% methanol) of extraction were regulated. Results obtained by supercritical fluid extraction (SFE) were compared to that of the conventional extraction procedure, hydrodistillation (HD). In the case of Satureja hortensis, Ocimum basilicum and Melissa officinalis extract yield of SFE was comparable to the essential oil amount obtained by hydrodistillation from the same drug. Essential oil rich extracts were analysed by GC-FID, while extracts rich in nonvolatile compounds were analysed by HPLC. We have found that yield and quality of SFE extracts highly depend on the conditions of extraction.
The allelopathic activity of the different extracts of the lichen Cladonia verticillaris and fumarprotocetraric acid on the early growth of A cepa (IPA 6) seedlings depends on their chemical composition and concentration, respectively. It was observed that the length of the radicle was significantly stimulated by fumarprotocetraric acid at high... concentrations and by the total extract of C. verticillaris thalli, which contained a high level of fumarprotocetraric, acid confirmed by HPLC-technique. In addition, it was found, that the phosphate buffer extract, which contained high level of methyl (3-orcinol carboxilate measured by HPLC, reduced the length of the hypocotyls significantly. Under our experimental conditions there was no influence of the different types of extract and fumarprotocetraric acid on the ratio of seed germination, in relation to the control. From the study of HPLC it was found that fumarprotocetraric acid and methyl 13-orcinol carboxilate were present in all extracts at different concentrations, depending on the method of extraction.
There has been a growing tendency in organic farming in recent years, in which organic grape growing and the production of biowine are also significant. The literature on bio-wine is rather limited, that is why we consider it really important to contribute to the data. As a result of the special conditions in organic farming we can expect bio...-wines to have a different composition of nitrogen compounds from that of traditional wines. Biogenic amines could thus specially characterize bio-wines. In our research we compared the biogenic amine content of wines produced using organic methods and of those where traditional procedures were used. Our measurements have proved that it is not possible to clearly distinguish bio-wines from wines produced with traditional methods on the basis of amine content.
Lavandula species are worldwide grown essential oil producing medicinal plants with considerable economic value. Beside volatile oil, lavender also contains different phenolic compounds which have been less widely studied till now. Cultivation of lavender has become widespread in Hungary in the recent years, however, growers have limit...ed knowledge on the productivity of cultivars available. In our research we were aimed to studying the antioxidant capacity and total polyphenol content of samples collected in two growing areas (Dörgicse and Szomód) from flowers of two L. angustifolia (‘Hidcote’, ‘Munstead’) and two L. × intermedia (‘Grosso’, Grappenhall’) varieties, during the full blooming period of 2017. Antioxidant capacity of the samples was determined by FRAP assay (Benzie and Strain, 1996), while total polyphenol content (TPC) was measured by a modified method of Singleton and Rossi (1965). According to our results, varieties of L. × intermedia showed higher values of antioxidant capacity and of total polyphenol content, than those of L. angustifolia cultivars. Among them, both the antioxidant capacity (179.6 mg AAE/g DW) and total polyphenol content values (152.4 mg GAE/g DW) of ’Grosso’ from Dörgicse were the highest. Concerning FRAP values of all the cultivars investigated, larger variability were found in Dörgicse, than in Szomód. Regarding the effect of growing area, each cultivar represented similar FRAP values in Dörgicse and in Szomód, except for ’Grosso’. However, in the case of TPC values, higher variability was observed between the growing areas, especially in the case of ’Grosso’. In the future our studies on lavender polyphenols will be completed with qualitative evaluation of the values obtained by HPLC analysis.
The influence of the foliar nutrition on the pear fruit quality (Pyrus communis L.) cv. 'Williams' was studied in years 1997 and 1998. We determined the contents of individual sugars (glucose, fructose, sucrose and sorbitol) and organic acids (malic, citric, fumaric and shikimic) by HPLC (High Performance Liquid Chromatography). The si...zes of the fruits (diameter, length, weight) were measured as well as the amounts of soluble solids and titrable acids. The experiment comprised two treatments: the foliar nutrition and the control. In the treatment of foliar nutrition the trees were sprayed five times (from May 22 to July 7) with a foliar fertilizer, which contained 15% of P205, 20% of K20, 0.1% of Mn, 0.1% of B and 0.1% of Mo. The foliar nutrition influenced higher quantities of sugars (glucose, sorbitol, soluble solids) and organic acids (malic, citric) but had no effect on the contents of shikimic acid, pH juice and titrable acids. At the treatment of foliar nutrition a trend of decrease in the contents of fructose, sucrose, fumaric acid, boron, and zinc was noticed.
Our general conclusion was that the thiamine amounts of 0,6 and 3,0 mg/1 added prior to wine fermentation, resulted in higher free-S02 level in comparison to the control. Furthermore, among the yeast strains examined in our experiments, Uvaferm BC strain produced the highest free-S02 content under the conditions mentioned...before. As regards the dynamics of reductone and SO, production, the concentration of the previous one was practically identical in the middle and at the end of the fermentation while the SO2 content — both free- and total — was lower at the end. Thiamine addition did not cause any difference in the organoleptic properties of wines as proved by sensory analysis.
Lycium barbarum is known exclusively as an ornamental plant in Hungary, and is planted so, as popular belief deem it a toxic plant. The plant’s fruit receives great respect in countries abroad, thus, due to its favourable content values the Lycium has achieved the title of ‘biological gold mine’. The last couple of years has seen the impo...rt of Lycium shrubs and its corresponding products, dried goods or in the form of various processed products which have been marketed and sold at extreme prices (under name of Goji, Wolfberry, Lifeberry). Our goal was to examine the similarities of the content values of wolfberry found in Hungary and those cultivated abroad. Along with Miklós Józsa the domestic Lycium population was surveyed based on foreign examples, between the years 2009 and 2011. Those defined sweet and largefruited were selected for further investigations to be set into a clone repository. This clone repository– which contains 67 different clones from a number of regions of Hungary – was established in the nursery of Dr. Miklós Józsa, located in the city of Szombathely. The phenological and morphological characteristics and the fruit ripening and quality indicators of plants in the clone repository were investigated. Six ‘best’ clones – selected based on flavour, disease-resistance and vegetative characteristics (plant size, fruit set, yield and fruit size) were analysed based on their content values. The control plant was a cultivar imported by a delivery service, found also in growing. In addition to the results of the selection, the results of the content values of the six selected ‘best’ clones (total soluble sugar content, glucose-fructose ratio, carotene content, FRAP value) is documented in this paper. Further, based on the resulting information the possibilities of the fruit’s utilization are suggested. Significant differences were measured in the vegetative characteristics of the clones (plant size, fruit set, yield and fruit size) and of those of
its inner content values. The resulting properties are considerable in regards of the plant’s introduction into cultivation and also utilization. Based on growing characteristics, more favourable clones were selected than the foreign varieties already in growing. Those clones selected by us have had similar or better content values than foreign breeds. The investigation of these content values is still in progress.
Oxygen free radicals play an important role in the development of different disorders like inflammatory-immune injury, carcinogenesis, hepatic toxicity and artherosclerosis. The antioxydant role of a wide spectrum of natural products has been established. Flavonoids and other phenolic compounds (proanthocyanidins, rosmarinic acid, hydroxicinnam...ic derivatives, catechines, etc.) of plant origin have been reported as scavengers and inhibitors of lipid peroxidation.
We have studied the antioxidant activity as well as content and composition of natural phenolics in a series of medicinal plants with phytotherapeutical significance. Thus we determined the total phenol contents and studied the composition of flavonoids, polyphenols, phenolic acids of different vegetative and reproductive organs of medicinal plants: Anthriscus cerefolium (L.) Hoffm., Petroselinum crispum L., Cichorium intybus L., Helichrysum arenarium D.C.„cempervivum tectorum L., Taravacum officinale Web.
Characteristic constituents in the various crude drugs were determined by chromatographic (TLC, HPLC) and spectroscopic (UV, UV-VIS) methods. The non specific scavenger activities of the medicinal plant extracts were studied by the chemiluminometric technique. The changes of chemiluminescence intensity of the H,G,•0H-luminol system at increasing concentrations of the H702/ -OH were measured. Inhibitory effects of selected standardized fractions from plants were tested on ascorbic acid induced lipid peroxidation in rat liver and homogenates.
The best correlation were established with total phenolics in some medicinal plants (S. tectorum, T. officinale) while activities in other cases seem to be influenced by flavonoids (P. crispum, H. arenarium, A. cerefolium) and by hydroxicinnamic derivatives (C. intybus).
A study was conducted to analyse the carotenoids by high-performance liquid chromatography (HPLC) using reversed-phase cross linked end-capping and to study the pigment content and composition in sea buckthorn products as well as in freshly harvested fruits from peach thees grown under organic and integrated farming conditions. It was found tha...t carotenoids in fruits of both crops occur mainly esterified with fatty acids in form of mono- and di-esters. The major carotenoids were esters of zeaxanthin, lutein, β-cryptoxanthin and β-carotene. The major carotenoids showed different response to processing of sea buckthorn being highly sensitive to thermal treatments such as blanching, cooking and drying. Significant differences were found between peach varieties in their carotenoid content, whereas the impact of organic farming on carotenoids formation was found to be variety-dependent. Two of the three varieties examined in this work, when cultivated under organic farming conditions contained lower carotenoid level as compared to that found in the fruits of the same varieties but produced in integrated farms.
It has been established, that thiamin content added in 3 mg/I concentration to musts before fermentation resulted in the enhanced storability of sparkling wine base at lower SO2 levels. Fermentation rate is not increased by this concentration and it causes no "thiamine taste" in organoleptic evaluation.
Influence of a three-year-long foliar fertilization on mineral composition of leaf, content of sugars (glucose, fructose, galactose and sucrose) and organic acids (citric, malic and fumaric acid) of sweet cherry (Prunus avium L.) fruits was studied. The experiment was conducted during 2005-2007 in West Hungary on mature cv. `Germersdor...fi 3' grafted on Prunus mahaleb rootstock, planted in 1999. Trees spaced 7 x 5 m, and growing in a calcareous chernozem soil. Trees were foliar-fertilized with potassium (K) as KNO3 and calcium (Ca) as Ca(NO3)2. Potassium spraying was carried out 3 (K1) and 5 (K2) while calcium was applied at 3 (Ca 1), 5 (Ca2) and 6 (Ca3) weeks after full bloom. Beside fruit analysis, complete soil and leaf analysis were done to study the rate of nutrient uptake and its effects on fruit quality. Contents of nutrients of soil and leaf were determined by atomic absorption and spectrophotometric method, while sugars and organic acids in fruit were determined by HPLC. The applied treatments (except K1) had been increasing leaf K significantly compared to the control till ripening. Most of treatments had no significant effect on Ca content of leaf till ripening. From applied treatments only the boron treatments had significant increasing effect on contents of all examined sugars, compared the control. Furthermore, the effect of calcium spraying on the contents of organic acids was significant.
