Grapevine and apple replant disease in Hungary
57-61.Views:154Field experiment was conducted to study the replant problems of grapevine and apple. Plantings were in three different fields: on virgin soil, on apple replant soil and on vine replant soil. Each field was planted with 60 pieces of grafted vine (variety Bianca on rootstock Berl. X Rip. T.K. 5BB) and 60 pieces of grafted apple (variety Gloster on rootstock MM. 106). Fungicide (BUVICID K with 50 % captan agent, 0.5 g/1 1 soil) and nematocide (VYDATE 10 G with 10 % oxamil agent, 0.03 g/1 I soil) treatments were used in the soil in order to identify the causal factor of the problem.
Biological soil test was conducted to test 17 soil samples of II wine districts and vine growing fields in plastic pots, under shading net. No root pieces were left in the soil. Two bud-cuttings of the Bed. X Rip. T 5C rootstock varieties were used as test plants. In each case, samples were taken from the vineyard and from the virgin soil. One fourth of the soil from the vineyard was left untreated and the other three part was treated with nematocide, fungicide or heat.
The results of the field experiment suggest that there was no problem growing grapevine after apple and apple after grapevine, but both species had been inhibited growing after itself. The fungicide and nematocide treatments did not succeed in determining the casual factor of the problem. Heat treatment of replant soil (in pot test) was useful in AS and VNS soils.
Results of biological soil test suggest, that grapevine replant problem do not occur in every vineyard. In fifty percent of soils, no significant differences between the treatments for shoot length, weight of cane, length, diameter and wood:ratio of the fourth internode were observed. In one case, difference was not found in any of the measured characters. However, fruiting bodies of Roesleria pallida (Pers.)Sacc. and the mycelium of Rosellinia necatrix Prill. were observed in this sample. In other samples, there was no significant difference between the treatments, but nematode and fungus infection appeared to be involved in increased shoot growth in nematocide and fungicide treated plants (mycelium of Rosellinia necatrix was detected). In other samples, the fungus infection caused significant difference between the virgin, untreated and fungicide treated soils and infection of Rosellinia necatrix was observed.

Reactions of some cucumber (Cucumis sativus L.) lines and hybrids to zucchini yellow mosaic virus (ZYMV) and selection of tolerant breeding lines
66-68.Views:172In the past years zucchini yellow mosaic virus (ZYMV) has been appeared as a new pathogen of cucurbitaceous plants in Hungary. It caused severe disease outbreaks on the fields of pickling cucumber hybrids which are highly tolerant to cucumber mosaic virus (CMV). Preliminary inoculation tests have showed that all of the important pickling cucumber hybrids produced in Hungary were susceptible to ZYMV-P. However, a selection of Chinese Long and the breeding line I- KS 10C proved to be highly tolerant. The Fl plants of the crosses between the tolerant lines and the susceptible genotype H I were susceptible to ZYMV-P. F2 populations segregated for susceptible and tolerant individuals at a ratio of 3:1. The results strongly suggest that the resistance to ZYMV in our sources is controlled by a single recessive gene. Inoculations of the ZYMV and CMV susceptible cultivar Budai csemege and the CMV tolerant hybrid Perez Fl with the complex of ZYMV+CMV resulted extremely severe symptoms (strong mosaic and necrotic spotting of the leaves) on both cultivars. On the Chinese Long line, which is tolerant to ZYMV and CMV, respectively, the complex of the two viruses caused mild symptoms. The results show a synergistic pathological effect of ZYMV and CMV on differrent cucumber genotypes. More detailed studies on the interactions among the plant genotypes, viruses and virus strains are needed to develop cucumber hybrids that are highly resistant to the ZYMV+CMV complex.

Recent developments in biochemical characterization of Vitis vinifera L. varieties in Hungary
57-61.Views:179Isoelectric focusing is an effective and well reproducible method to provide information for identification of various plant species and clones if breeding or other genetic modification(s) for a given species are reflected in changes of an isozyme pattern. The method has been used for characterization of plant proteins and enzymes and for identification of various species and varieties. Our aim was to continue our several-year-work carried out on a wide variety of grapevine varieties and to reveal whether analyses of esterase and peroxidase isozyme patterns are suitable to distinguish various grapevine varieties. Therefore we compared esterase and peroxidase isozyme patterns of various species. Plant samples were obtained from Szigetcsep, Kecskemet, Tokaj and Eger. The following samples were analyzed: Pinot gris, noir, blanc, Chardonnay, Riesling Theses, Chasselas from Szigetcs6p, Bianca and his parents Eger2 and Bouvier from Kecskemet, Furmint and Hárslevelű from Tokaj, Kékfrankos and Zweigelt from Eger. To identify various species according to their esterase isozyme patterns the after blooming phenological phase while according to their peroxidase isozyme pattern the dormant phenological phase was found as optimal sampling time.

Molecular analysis of strawberry cultivars using RAPD, AP-PCR and STS markers
24-28.Views:136Seventeen strawberry (Fragaria x ananassa Duch.) cultivars representing the national list of Hungary, were subjected to RAPD, AP—PCR and STS analysis. Of the 31 decamer and oligomer primers tested 26 primers produced polymorphic patterns. 45 polymorphic fragments were analysed, ranging between 200-2800 by in size. Based on the data, similarity coefficients (Jaccard index and Simple matching coefficient) were calculated, and dendrograms were constructed using the unweighted pair group method of arithmetic averages (UPGMA). The dendrograms only partly reflect the known pedigree data. Specific RAPD markers were identified for cultivars F5, Pocahontas and Rabunda.

Investigation of the in vitro regeneration of mericlones in the caribe variety of carnation
87-89.Views:137In vitro culture conditions were experimented for the relatively sensitive, but very esthaetic "Caribe" variety of carnation with uniformly dark violet flowers. Regeneration of new plants from shoot apex meristems can be significantly improved by the combined addition of very low amounts of indolebutiric acid, benzyladenine and gibberelic acid, dissolved in the Murashige-Skoog nutrient medium. Callus formation as a prerequisite for the induction of somaclonal variability can be achieved successfully with certain molar ratios between 2,4-dichlorophenoxyacetic acid and benzyladenine. Acclimation of the obtained mericlones to the ex vitro conditions was also evaluated.

Microsatellite based identification of grapevine cultivars traditional in Hungary and in the Carpathian Basin
71-73.Views:149Grapevine cultivars and clones traditional in Hungary and in the Carpathian Basin are maintained in the genetic collection of the Corvinus University of Budapest. Mostly ancient varieties and clones were genotyped using microsatellite loci. The investigated 6 loci were sufficient to distinguish all cultivars. Some clones could also be separated based mostly on the variable VVS2 loci. For 17 out of the 31 investigated cultivars this is the first report on characterization of the polymorphism of the allele lengths by microsatellite markers on loci: VVS2, VVMD7, VVMD27, vrZAG62.

The effect of bud density on the composition of colour in red wines
63-68.Views:170Authors investigated the effect of fruit load on the composition of colour substances and of reservatrol in red wines of some grape varieties grown in Hungary. The content of both, anthocyanin compounds and reservatrol showed practically the same responses to fruit load and the same profile in all the varieties studied. Determination of these compounds solely is not sufficient to identify the respective grape variety.

Identification of ripening-related genes in strawberry fruit by cDNA-AFLP
33-41.Views:165An RNA fingerprinting study of strawberry receptacle and achene tissue was performed to identify candidate genes involved in fruit ripening. Quantitative cDNA-AFLP was used to detect differential gene expression in green, white, pink and red stages of fruit ripening. Based on hierarchical average linkage clustering the differentially expressed genes formed three major groups, genes expressed only in green receptacle, genes expressed mainly in white, pink and red receptacle, and in achene. 130 transcript-derived fragments (TDFs) were isolated and sequenced. Most TDFs did not show any homology to sequences with known functions, others were homologous to genes involved in oxidative stress response, signal transduction, regulation of development and cell-wall metabolism. Novel genes, so far not associated with strawberry ripening and ripening in general, were identified, such as genes encoding a bHLH protein, putative nitrilase-related protein, putative HD-zip protein. The differential pattern of gene expression draws the attention to the significance of ripening induced-or repressed promoters in strawberry fruit, whose isolation and characterization can be useful tool for functional genomics. For this purpose nine cDNA-AFLP fragments related either to ontogeny or senescing were completed with 5'UTR aiming at more precise annotation and future promoter isolation. Although tens of potentially important transcriptome changes were identified, the function of many ripening induced genes remain unknown.
