Comparison of alveohraph and extensigraph properties of winter wheat samples

The rheological properties of dough are determined by the amount and proportion of gliadin and glutenin proteins. Extensibility and resistance to the extension of doughs which can be measured using the extensigraph or alveograph allow good assesment of the baking behavior under realistic conditions. In our study, we compared the extensigraph an...d alveograph parameters of 87 flour samples. The results showed that the alveograph parameters were different from the extensigraph parameters. The classification of wheat varieties was different, and the order of varieties, as well. The alveograph properties were affected by other factors than the extensigraph properties, except the W value. The two rheological tests did not give similar results, therefor neither test are replaceable.

Parameter optimization of an inductively coupled plasma mass spectrometer for measuring arsenic and selenium

In the last decades, an increased interest has evolved in arsenic and selenium. The aim is to understand the environmental, agricultural and biological roles of these elements. In the case of arsenic, the major reasons are the relatively high concentration of arsenic in marine biota (mg kg-1) and the arsenic contaminated drinking bases of some Asian countries, as well as Hungary. The toxicity of higher level selenium content is also known; nevertheless, selenium is essential for several biological functions. Considering its essentialness, in our country, the insufficient selenium intake rate causes a lack of selenium. Measuring the concentrations of these elements provides crucial, but unsatisfactory information, as the speciation, i.e. the form of an element presented in a sample is also required.
In both cases, the most suitable method to determine concentration is inductively coupled plasma mass spectrometry (ICP-MS). Our objective was to optimize the variable parameters of the ICP-MS to attain the lowest (the best) detection limit. For this purpose, we investigated the effect of parameter change on net signal intensity and relative signal intensity. With the optimized parameter settings, the limits of detection for arsenic and selenium were determined, which are 0,032 ng dm-3 for arsenic, and 0,097 ng dm-3 for selenium.

Cytochrome b diversity of Hungarian Botrytis cinerea strains

In the mitochondrion of eukaryotes, cytochrome b is a component of respiratory chain complex III. Cytochrome b is encoded by the
cytochrome b (CYTB) gene located in the mitochondrial genome. The fungicidal activity of QoIs relies on their ability to inhibit mitochondrial respiration by binding at the so-called Qo site (the outer quinol-oxida...tion site) of the complex III. Since their introduction, QoIs (like azoxystrobin) have become essential components of plant disease control programs because of their wide-ranging efficacy against many agriculturally important fungal diseases like grey mould on various crops. QoI resistance primarily arises from a target-site-based mechanism involving mutations in the mitochondrial CYTB. As the management of grey mould is often dependent on chemicals, the rational design of control programs requires the information about the diversity of genes connected with resistance in field populations of the pathogen.
Monospore B. cinerea field isolates has been collected during 2008-2009 from different hosts in Hungary. PCR fragment length analysis
indicated the high frequency presence of type large intron in the isolates while in a few strains G143A substitution could also be detected.
These results indicated the heterogeneity of CYTB in the Hungarian B. cinerea populations, which possibly involve the heteroplasmy of this
mitochondrial gene, moreover indicates the existence op azoxystrobin resistant populations in Hungary.

Effect of molybdenum treatment on the element uptake of food crops in a long-term field experiment

Molybdenum, as a constituent of several important enzymes, is an essential microelement. It can be found in all kind of food naturally at low
levels. However, environmental pollution, from natural or anthropogenic sources, can lead to high levels of the metal in plants. Our study is based on long-term field experiments at Nagyhörcsök, wher...e different levels of soil contamination conditions are simulated. Plant samples were collected from the experiment station to study the behavior of elements: uptake by and transport within the plants, accumulation in different organs, phytotoxicity and effects on the quantity and quality of the crop. In this study, we present the effect of molybdenum treatment on the uptake of other elements. Molybdenum is proved to be in an antagonist relationship with copper and sulphur, while molybdenum-phosphorus is a synergist interaction. However, in most of the plants we studied, increasing molybdenum-treatment enhanced cadmium uptake. We found the most significant cadmium accumulation in the case of pea, spinach and red beet.

SSR based characterization of peach (Prunus persica L.) and apricot (Prunus armeniaca L.) varieties cultivated in Hungary

The SSR (Simple Sequence Repeat) markers allow the discrimination of the cultivars and determination its specific DNA fingerprints. The aim of this research was to evaluate fifteen apricot (Prunus armeniaca L.) and fifty-one peach (Prunus persica L.) genotypes cultivated in Hungary to obtain their DNA fingerprints in 6 SSR (Simple Sequence Re...peats) loci by allele numbers and sizes.
DNAs were extracted from leaves. PCR was carried out with CY-5 fluorescent labeled Prunus microsatellite markers and the products were separated on polyacrylamide gel with ALF (Automated Laser Flourometer)-Express II.
According to our results, in the case of peach genotypes, all 6 SSRs were able to amplify alleles. UDP 96 005 was the most informative marker and UCDCH 17 was the least due to its monomorphic pattern. Regarding the apricot samples BPPCT 041 did not amplify any allele. In the case of P. armeniaca UDP 96 005 had the highest heterozygosity index as well and the highest number of alleles. The least informative marker was the UCDCH 17. Since the 6 SSR were not enough to discriminate the apricot and peach genotypes, it is suggested to use more SSR primers.

The effect of bentonite on the quantity change of soil microorganisms, the CO2-production and the activity of saccharase enzyme

We examined the impact of bentonite – the perspective improving material of sandy soils – and treatments of livestock manure composted with bentonite on sandy soils, within the framework of a small-plot experiment.
The adjustment of the experiment was made on the Experiment Site of the Nyíregyháza Research Centre of the University of D...ebrecen, Centre of Agricultural Sciences (UD CAS). We collected soil samples from parcels treated with increasing bentonite doses (5, 10, 15, 20 t/ha) on the one hand, and from the parcels treated with livestock manure composted with increasing doses of bentonite, on the other.
We performed laboratory research in the soil microbiology laboratory of the Soil Science Faculty of UD CAS DAS, during which we determined the total number of bacteria, the quantity of microscopic fungi, the number of cellulose-decomposing bacteria, the CO2-production of the soil and the activity of saccharase enzyme.
During the evaluation of the examinations, we made a statistical analysis using SPSS 9.0. We determined the average of measurements, the standard deviation of controls, the standard deviation, the significance value and we also performed a correlation analysis.
Concerning the impacts of bentonite treatment and the treatment of livestock manure composted with bentonite on the examined microbiological features of sandy soil, we can summarize the following:
• Our results prove that the microbiologic activity of the soil has increased owing to the impact of bentonite treatments regarding total number of bacteria and the quantity of microscopic fungi. ”Pure bentonite” treatments – although not significantly in every case – increased these values, but larger doses decreased them. The treatments of livestock manure composted with bentonite resulted in a larger increase regarding both parameters.
• The number of cellulose-decomposing bacteria was increased by the low doses of both series, and was decreased by the higher doses. Higher bentonite doses decreased it in a higher – significant – degree than those of treatments of livestock manure composted with bentonite, whose low dose caused salient number of bacteria.
• Regarding the carbon-dioxide formation, we have experienced an increase even in the case of low dose treatments (nevertheless, the increase did not prove to be significant), but – similarly to the quantitative changes in the number of cellulose-decomposing bacteria – the large doses of both series of treatment decreased the CO2-production of the soil.
• Bentonite also increased the activity of saccharase enzyme significantly. We learned that”pure bentonite” treatments increased the activity of the enzyme to a higher degree than composted treatments. Moreover, it can be stated that the treatments of larger doses of both bentonite and livestock manure composted with bentonite have decreased the enzyme activity – not significantly, though.
• Based on the correlation analysis, it can be stated that as an impact of the treatments, the microbiological activity of the soil has also increased with the increase of the number of soil microbes, as in both treatment series we have experienced a tight positive correlation (r=0.81-0.82) between the change of total number of bacteria and the CO2-production of soil. In both treatments, there was a medium correlation between the total number of fungi and soil respiration (r=0.63-0.63). Furthermore, it can be stated that the usage of organic manure had a positive effect on the activity of cellulose-decomposing bacteria, as in this treatment series there was a positive correlation (r=0.65) between this physiological group and CO2-production. Both treatments prosperously impacted the activity of saccharase enzyme, because there was a medium correlation (r=0.62-0.64) between the activity of the enzyme and soil respiration.

Element content analyses in the Institute for Food Sciences, Quality Assurance and Microbiology

The role of chemical elements to ensure and promote our health is undisputed. Some of them are essential for plants, animals and human, others can cause diseases. The major source of mineral constituents is food, drinking water has a minor contribution to it, so the knowledge of elemental intake through food is crucial and needs continuous moni...toring and by this way it promotes the food quality assurance and dietetics.
With the evolution of spectroscopic methods increasingly lower concentrations could be determined, so the elemental composition of a sample could be more precisely and fully described. Due to the results the gathered knowledge up to the present is supported and new observations can be done helping us to understand such complex systems as biological organisms are.
The quality of a food is determined by the full process of its production, consequently it starts with agricultural production so elemental-analysis usually cover the whole soil – plant – (animal) – food chain, by this way the „Fork-to-Farm” precept is true in elemental analysis field also.
The history of elemental analysis in the University of Debrecen, Centre for Agricultural and Applied Economic Sciences, Faculty of Agricultural and Food Sciences and Environmental Management, Institute of Food Processing, Quality Assurance and Microbiology goes back to 1980s when the so called Regional Measurement Central gave the background for research. The continuous deployment resulted in an obtain of an inductively coupled plasma atomic emission spectrometer (ICP-AES) in 1988, which extended the scope of examinations due to its excellent performance characteristics
compared to flame atom absorption (FAAS) and flame emission spectrometers (FES). The instrumental park retain up to date correlate to the developing analytical techniques due to acquiring a newer ICPAES in 1998 and an inductively coupled plasma mass spectrometer in 2004 – which sensitivity is three order of magnitude better compared to ICP-AES. The Institute supports the work with its own ICP-AES and ICP-MS since 2011.

Comparison of the sample preparation methods worked out for the examination of the element content of wine

The examination of the potentially toxic elements content of the wines is not easy task, because the most elements are in little concentration (mg kg-1 or μg kg-1) in the wine and the wines contain great amount of organic matrix. The efficient sample preparation is essential for the accurate determination of element conte...nt. The eim of our research was to determine which sample preparation method will be the most efficient in examination of wines with ICP technology. The examined wine sample was a 2008 Chardonnay from the Eger wine region. We did the sample preparation and analysis examination in University of Debrecen, Centre for Agricultural and Applied Economic Sciences, Institute of Food Science, Quality Assurance and Microbiology.
We did the analysis examinations with ICP- MS (inductively coupled plasma mass spectroscopy). We always did the sample preparations and the examinations in three times rehearsal. The applied sample preparation methods: dilution with distilled water, open digestion and microwave digestion. 
We were able to measure B, Al, Mn, Fe and Zn with only dilution and open sample preparation. In the smaller quantity present Sr and Ba were measurable in the wine in the case of all three methods well. We were able to measure the Co with dilution and open digestion method,  while Cr, Ni, and Te with only dilution method. In the case of arsenic we were not able to measure reliable result with dilution and open digestion method because of organic matrix and other components
(alcohols, monosaccharides, polysaccharides, polyalcohols and inorganic salts). On the whole we are able to say that in the case of certain elements (B, Mn, Fe, Zn, Sr, Ba) the open digestion and dilution sample preparation is applicable well, however, in the case of certain elements (As, Al, V, Cr, Se, Mo, Cd, Hg, Pb) we have to develop the methods. It may be development of one of the way, if we develop sample preparation methods to examined element specifically and not
to wine generally.

Effects of some herbicides on the microbiological characteristics of soil nitrogen cycle under maize plantation

Nitrogen is a key element for the living organisms and influence not only for the quantity but for the quality of the yield, considerable. Availability of nitrogen from the soil is influenced by several microbiological processes of the Nitrogen-cycle. Among the intensive agricultural production the herbicide application cannot be omitted more i...nformation needs therefore about the inhibitor effect of herbicides on the different microorganisms.
An experiment was set up on calcareous chernozem soil under maize culture. Effect of four different herbicides (Acenit, Frontier, Merlin, and Wing) was investigated. The effect of herbicides was measured to four microbiological parameters of the Nitrogencycle (abundance of nitrifying bacteria, nitrate solubilisation, biomass nitrogen and urease enzyme activity). There were singledouble-  and five times of recommended doses of herbicides applied for two onsecutive vegetation periods.
From the results of the different doses of herbicides, the following can be stated:
– The Acenit has a stimulating effect on nitrifying bacteria in general. The Frontier and Merlin also influenced the quantity of nitrifyers, however in certain cases decreased in another cases increased the number of bacteria.
– The double doses and five times doses of herbicides was found to be increasing the nitrate content of soil, -especially in 2006.
– The quantity of microbial biomass nitrogen increased in the 60% of treatments and decreased in the 40% of the treatments.
– Except of the result of Wing in 2006 and Merlin in 2005, the effect of simple dose herbicides was the smallest on the urease enzyme activity. According to the results the effect of Merlin was positive; the effect of Wing was negative on the soil enzyme’s activity.
Regarding the application of four different herbicides in three  different doses on the microbiological parameters of soil (at two consecutive years-) in 39% of the treatments has resulted a significant inhibitory effect, 28% of the treatments, however have significant stimulating effect on the parameters measured. More than 50% of the inhibitory effect was measured in case of the Wing, at more than 50% of the Frontier the microbiological parameters have not changed.

Definition antioxidant activity of selenium-enriched food sprout, as well as their microbiological analysis

We took into account the fact, we choose micronutrients to our treatment, that selenium can be delivered to the body by a small amount with the most widely consumed food.

We focused during our research to determine that the increasing concentrations of selenium treatment, in which we used sprouts, knowing fully well that it has an impact on aboriginal antioxidant capacity of sprouts, which is mainly due to high vitamin content of sprouts.

Furthermore, we think it is important to make microbiological analysis, because germination conditions, for example temperature, pH, all this will create an ideal environment for the growth of microorganisms. So we had goal to determine, how the used selenium concentration affect the total plate count, coliform bacteria count and Staphylococcus aureus count of sprouts.

We determined the aboriginal water-soluble and lipid-soluble antioxidant capacity of sprout with the PHOTOCHEM chemiluminometer and we applied pour plate technique for the mapping of the mycrobiological state of sprouts.

Experimental results are evaluated, we state that increasing concentrations of selenite or selenate treatment, is primarily water-soluble antioxidant capacity of sprouts was affected. The water-soluble antioxidant capacity of wheat sprout was much higher than the measured values in pea sprout, this may be linked to what we measured. That is much higher ascorbic acid content in case of wheat sprout, which is well known as one of the most important antioxidant properties compounds of wheat sprout.

We conclude from the results of the microbiological, that the highest concentrations of selenite or selenate treatment has a relative significant anti-microbial effect in case of wheat sprouts. Coliform and total plate count showed no clear decreasing tendency, although the values of treatments in both cases obtained were below the control values.

The effect of different herbicide on the number and activity of living microorganisms in soil

Sustainable plant growth, considering the difficulties of weed elimination, cannot be effective without the application of herbicides. However, these chemicals have enormous ecological implications, including effects on the microbiological communities of soils. It is advisable to use herbicides that have minimal secondary effects on the environ...ment and soil-living microorganisms. In contrast, herbicides with prolonged growth stimulating or inhibiting effects are not suitable, because both types have strong influences on the number and activity of bacteria, thus causing changes in the ecological equilibrium.
Preceding small plot experiments, laboratory tests were carried out to study the effect of herbicides used in maize cultures on the number of bacteria and growth of microscopic fungi.
Substances that were observed to have stronger influences were applied in small plot experiments set up in the experimental garden of the Department of Plant Protection of the University of Debrecen. We studied the effects of four herbicides (Acenit A88EC, Frontier 900 EC, Merlin SC and Wing EC) on the microbiological properties of the soil. These herbicides were used in different concentrations in maize culture, and we investigated the effects in different soil layers.
In the laboratory experiments, we determined the total number of bacteria and microscopic fungi and examined the growth of Aspergillus niger, Trichoderma sp. and Fusarium oxysporum on peptone-glucose agar containing herbicides.
During the small plot experiments, soil samples were collected 3 times a year from 2-20 cm depth. The total numbers of bacteria and microscopic fungi were determined by plate dilution method, while the method of most probable number (Pochon method) was used to determine the numbers of nitrifying bacteria and cellulose decomposing bacteria. To evaluate the microbiological activity of the soil samples we measured carbon-dioxide release (after 10 days incubation), nitrate production (after 14 days incubation) and the concentration of C and N in the biomass.
We can summarize our results as follows:
• In laboratory experiments, herbicides caused a decrease in the number of bacteria and inhibited the growth of microscopic fungi.
• Frontier 900 EC and Acenit A 880 EC had the strongest inhibiting effect on microorganisms.
• In small plot experiments, herbicide treatment decreased the total number of bacteria and microscopic fungi.
• Herbicides caused a significant increase in the number of nitrifying and cellulose decomposing bacteria.
• Different herbicides containing the same active compound had similar influences on soil microoorganisms.
• A significant increase was observed in the physiological processes of tolerant microorganisms surviving the effects of herbicides

Comparative examination of a mineral fertiliser and a bacterial fertilizer on humic sandy soil

In our pot experiment, the impact of a bacterial fertilizer, Bactofil® A10 and a mineral fertilizer Ca(NO3)2 applied in different rates was studied on some soil chemical and microbiological characteristics of a humic sandy soil (Pallag). Perennial rye-grass (Lolium perenne L.) was used as a test-plant. Samples were collec...ted four and eight weeks after sowing in each year. The experiment was set up in 2007-2009 in the greenhouse of
the UD CASE Department of Agrochemistry and Soil Science. The available (AL-extractable) nutrient contents of soil, among the microbial parameters the total number of bacteria, the number of microscopic fungi, cellulose-decomposing and nitrifying bacteria, the sacharase and urease enzyme activity, as well as the soil respiration rate were measured.
Statistical analyses were made by means of the measurements deviation, LSD values at the P=0.05 level and correlation coefficients were calculated. Results of our experiment were summarised as follows:
− The readily available nutrient content of humic sandy soil increased as affected by the treatments, in case of the available (AL-extractable) phosphorus and potassium content the higher value was measured in high-dosage artificial fertilizer treatment.
− The treatments had also positive effect on several soil microbial parameters studied. The higher-dosage mineral fertilizer treatments had a beneficial effect on the total number of bacteria, cellulose-decomposing and nitrifying bacteria. No significant differences were obtained between the effect of treatment in case of the total-number of bacteria, the number of microscopic fungi and nitrifying bacteria.
− On the sacharase enzyme activity the artificial fertiliser treatments proved to be unambiguously stimulating, the urease activity significantly increased on the effect of the lower-dosage Ca(NO3)2 artificial fertilizer treatment. 
− The soil respiration increased in all treatments in related to the amounts applied, significantly increased in the highest rate of Ca(NO3)2 fertilizer addition. 
− Some medium and tight positive correlations were observed between the soil chemical and microbiological parameters studied in case of both nutrient sources. 
Summarizing our results, it was established that the organic and all the mineral fertilizer treatments had beneficial effects on the major soil characteristics from the aspect of nutrient supply. In majority of the examined soil parameters (AL-extractable phosphorus- and potassium, total number of bacteria, number of cellulose-decomposing and nitrifying bacteria, activity of sacharase enzyme) the high rate of Ca(NO3)2 mineral fertilizer treatment proved to be more stimulating, but at the same time the high rate bacterium fertilizer resulted in significant increases in
the nitrate-N content, the AL-potassium content of soil, the total number of bacteria, the number of cellulose-decomposing and nitrifying bacteria and the urease enyme activity. 
Our examinations showed that the mineral fertilizer treatments proved to be more stimulating on most of the soil parameters studied but according to our results, it was established that Bactofil is efficiently applicable in the maintenance of soil fertility and the combined application of
mineral fertilizer and bacterium fertilizer may be a favourable opportunity – also in aspect of the environmental protection – in maintaining soil fertility.

The application of bentonite and zeolite for soil amelioration in acidic sandy soil

In a pot experiment, we have studied the effect of bentonite and zeolite in different dosages [control; 5; 10; 15; 20 g kg-1] on acidic (pHH2O=5.65) humus sandy soil. The experiment was set up in 2007 and 2008 in the greenhouse of the UD CASE Department of Agrochemistry and Soil Science. As a test plant, perennial ryegrass... (Lolium perenne L.) was used. 
In laboratory examinations, pH(H2O), pH(KCl), hidrolytic acidity, nitrate-N content, readily available phosphorus and potassium content were determined. Among soil microbial parameters, the total number of bacteria, the cellulose-decomposing bacteria, the carbon-dioxide production, the microbial biomass-C content of soil, and the saccharase enzyme activity were measured. In the experiment the biomass of the test plant was determined.
The effect of bentonite and zeolite in different dosages can be summarized as follows:
− The pH increased under the effect of low dosages. With the increasing of the pH the hydrolytic acidity - at the bentonite treatments significantly – decreased. 
− Regarding the readily available nutrient content of the soil, low and medium dosages proved to be effective. High dosages of bentonite treatments reduced the nitrate-N content, the readily available phosphorus, and potassium content of soil, by zeolite treatments the high dosages reduced the nitrate-N content of soil. 
− Regarding the measured soil microbial parameters in both treatments low and medium dosages proved to be also effective, but the high dosages didn’t cause decreasing at the total number of bacteria, and by zeolite treatments the biomass-C content of soil.
− Also the bentonite and zeolite treatments enlarged the biomass of the test plant. We experienced significant increasing by bentonite treatments by the effect of medium and high dosages, while in zeolite treatments only the high dosage caused significantly increasing in plant biomass. The largest dosages decrease the plant biomass. 
− Under the statistical analysis we found many medium and tight correlation between the studied parameters.

The dynamics of biodiversity structure of soil microorganisms under the impact of biopreparations during potato growing season

The use of biological preparations such as Phytotsid and Planryz contributes the increase of the general number of soil bacteria population by 13.0–36.1% in the case of potato variety Scarbnytsya and by 4.5–24.6% for potato variety Oberig compared with control. It also increases the number of saprophyte microflora, which compete with plant ...pathogens, micromycetes, and causes 1.2–1.8 times reduction in the number of soil fungi – Fusarium and Alternaria. During the application of Rovral Akvaflo the Shannon ecological index of species biodiversity is lower than during the biopreparation use. The decrease of species biodiversity was observed as well as strengthening the dominance of some species (dark pigmentation in fungi).

The examination of presumed Escherichia coli count of raw milk samples on several milk production farms

For dairy farms, it is of great importance to insure the appropriate hygienic status of milk and to examine it regularly. Escherichia coli, belonging to the coliform bacteria type of, is a good indicator of contamination, and therefore suitable for characterising the hygienic condition of milk production.
The aim of our research was to the connection between the Escherichi coli count in bulk tank milk and housing and milking technologies of different-sizes farms. We examined the relation using various statistical methods.
Analysing the connection between the E. coli count and the farm size we found no significant difference between the farms. On the basis of the mean values of the E. coli count, we can say that the hygienic conditions are appropriate for mid-sized farms, and tolerable for large farms. We found differences in the hygienic status among the small farms. Half of the eight small farms, had no adequate hygiene. The results of the analysis of the quality categories show that the probability of inadequate quality milk was the largest on small farms (37.5%).
Comparing the various housing and milking methods with each other, there were numerical differences in the E. coli count, but these differences were not significant. We got higher E. coli count values on those farms using tied stall barn and bucket milking installation. The reason for this could be that in cases of farms using bucket milking installation, it is harder to meet the requirements.
After forming groups by farm size, housing and milking methods, we found that the E. coli counts are adequate on mid-size farms using various housing and milking methods; and tolerable on those large farms using loose housing stable and a milking parlour. At the same time, we found inadequate E. coli counts on the smaller farms using tied stall barns and bucket milking installation.
The results show that if there is suitable attention, independent of farm size, housing and milking procedure, it is possible to produce milk with low E. coli counts, and to insure appropriate hygienic conditions.
Further detailed examinations are needed to decide which factors of housing and milking technologies influence the E. coli count of bulk tank milk.

Comparison of supplier quality assurance methods for compound feed manufacturing from efficiency, product consistency and economical point of view

The food industryparticularly has intensively implemented the recent quality management principles and developed systems that facilitate the continuous improvement and efficiency of the industrial production. Feed production has taken similar appr...oaches however the intensity of deployment at the manufacturer and the rollout towards its supply chain has shown slower progress. The methods, that the feed manufacturer manages the supplier chain of mainly primer agricultural products according to, have a certain impact on the efficiency of the inbound operations, feed product quality and its consistency as well as on other resources. These methods have been built on sound quality management principles that are stated not only in quality standards but also in relevant regulations. Current study addresses the questions related to the link between supply chain quality management and feed product. The objective of the frontier research was to highlight the  theoretical possibilities and benefits of the robust design methodimplemented into animal feed manufacturing dealing with highly variable ingredients.

Identification of the Slovak traditional cheese “Parenica” microflora

Numerous studies have demonstrated the higher accuracy, faster time-to-results and lower costs provided by MALDI Biotyper systems compared to classical methods. In this study, the culturable population of total count of bacteria, enterococci, coliforms bacteria, lactic acid bacteria (LAB) and microscopic fungi and yeasts from cow’s dairy prod...ucts was identified using the MALDI-TOF MS Biotyper. Altogether, 50 samples of the Slovak cheese “Parenica” were examined. Total numbers of bacteria were cultured on Plate count agar at 37 °C for 24–48 h, aerobically; enterococci were cultured on Enterococcus selective agar at 37 °C for 24–48 h, aerobically; coliforms bacteria were cultured on Violet Red Bile lactose agar at 37 °C for 24–48 h, aerobically. The LAB were cultured on MRS (Main Rogosa agar), MSE and APT agar at 30 °C in microaerophilic conditions. The microscopic fungi and yeasts were cultured on Malt extract agar at 25 °C for 5 days, aerobically. Isolated strains (total 669) were subjected to identification by the MALDI-TOF MS. Among total count the identified bacteria mostly were Acinetobacter baumannii, Bacillus cereus, Micrococcus luteus and Staphylococcus warneri. Escherichia coli and Enterobacter cloacae were the most abundant coliform bacteria representatives identified. Coliform bacteria included Citrobacter, Hafnia and Klebsiella. Altogether three genera belonged to the LAB – Lactobacillus, Lactococcus and Leuconostoc were identified with Lactococcus lactis, Lactobacillus plantarum, Lactobacillus coryniformis, L. fructivorans and Leuconostoc mesenteroides were considered as the dominated LAB species in dairy products. Among yeasts, Kluyveromyces lactis, Candida zeylanoides and Yarrowia lipolytica were among the most isolated.

Examination the effects of different herbicides on the soil microorganisms of a calcareous chernozem

Pesticides play a key role in fighting weeds, pests and parasitic fungi. According to surveys, pests reduce the yield of agricultural crops by 35% worldwide. Pests, fungi and weeds account for 14%, 12% and 9% yield loss, respectively (Gáborjányi et al., 1995). Chemicals have contributed to increasing and maintaining the yields of crop product...ion for decades. Today, agricultural production (in spite of many efforts) is unthinkable without the use of pesticides (herbicides, insecticides and fungicides). On the other hand, these chemicals contribute to the pollution of the atmosphere, surface and underground waters, and agricultural soils, especially if they are applied improperly.
The sustainable agricultural production pays attention to environment-friendly cultivation-technologies; but at the same time it makes an effort to produce good quality and economical products. The examination of the herbicides’ secondary effects, fits into this chain of idas namely, how the herbicides affect – stimulating or inhibiting – the soil microbiological processes, prevention of soil fertility.
In the course of the experimental work the effect of herbicides on soil biological properties were examined in different maize (Zea mays) cultures. We wanted wished to know that how the herbicides affect the quantity change of soil microorganisms, the life of different physiological groups of bacteria and the activity of microorganisms. A small pot experiment was set up in 2008 with the application of two herbicides - Acenit A 880 EC and Merlin 480 SC – in the breeding house of the Department. The moisture content and nutrient supply were at optimal level in the experiment.
On the basis of results the following can be stated: 
1. It can be stated that the two herbicides and all their doses affected negatively the number of total soil bacteria, the
inhibiting effects were significant. The quantity of microscopical fungi increased by the effect of Merlin 480 SC and decreased in the treatments of Acenit A 880 EC.
2. The Acenit A 880 EC had stimulating effect on the nitrate mobilization. The CO2-production was stimulated by the basic doses of herbicides; the other treatments did not influence the CO2-production significantly.
3. The quantity of microbial biomass-carbon –except for only one treatment- decreased significantly by the effect of herbicides. Besides it, the quantity of microbial biomass-nitrogen increased significantly in the treatments of Acenit A 880 EC.
4. The biomass of test plant decreased in the treatments of herbicides, their quantities were smaller than in the control. In the pots treated by Merlin 480 SC, parallel with the increase of doses decreased the quantity of plant-biomass.

Improved soil and tomato quality by some biofertilizer products

The use of microbial inoculums is a part of sustainable agricultural practices. Among various bioeffectors, the phosphorus-mobilizing bacteria are frequently used.

The objective of this study is to investigate the effect of some industrial biofertilizer inoculums, of containing P-mobilizing bacteria on the quantity and some quality para...meters of tomato fruits. Spore-forming industrial Bacillus amyloliquefaciens FZB42 (Rhizovital) as single inoculums and combinations with other Bacillus strains (Biorex) were applied on Solanum lycopersicon Mill. var. Mobil test plant. Soil microbial counts, phosphorus availability, yield and fruit quality, such as total soluble solids (TSS) content and sugars (glucose, fructose) were assessed. The results found that single industrial inoculums of FZB42 product had positive effect on P-availability and fruit quality in the pots. Fruit quality parameters, TSS content, soluble sugars were significantly improved (p<0.05). Such better fruit taste was correlated significantly by the most probable number (MPN) microbial counts. Use of such bioeffector products is supported by the positive interrelation among measured soil characteristics and inside healthy quality parameters of tomato fruits.

Variations in major quality parameters of forage and medium quality winter wheat varieties in storage

We analysed five parameters (moisture-, protein content, Hagberg’s falling number, wet gluten content and alveographic W (10-4 J) values) and the microbiological changes of four forage and milling III. quality winter wheat varieties (Magor, Hunor, Róna and Kondor) during storage, to determine the tendency, type and volume of the o...f the change of this five qualitative parameters during storage.
We found that the examined winter wheat varieties retained their moisture, protein content and their Hagberg’s falling number, they did not change during storage.
A slight growth could be experienced in the values of wet gluten content for all the four winter wheat varieties in terms of the duration of storage (129 days). This result proved the theory of after-ripening, when gluten percentage improves qualitatively and quantitatively as well. The value of the quantitative growth was about 10% for all the four winter wheat varieties.
We placed a special emphasis on measuring the alveographic W (10-4 J) values during storage. All the four winter wheat varieties showed decreasing values of about 20-40%.
Microbiological examinations on the four winter wheat varieties showed that mould, mould flora and total germ count remained balanced with some slight variations and they did not change in terms of time under optimal storage conditions.

Comparative examination of a bacterium preparation (BACTOFIL® A10) and an artificial fertilizer [CA(NO3)2] on calcareous chernozem soil

In a small-pot experiment a bacterium preparation, Bactofil® A10 and an artificial fertilizer containing Ca(NO3)2 in different dosages were studied on calcareous chernozem soil, concerning the readily available nutrient content of soil (nitrate-nitrogen, AL-phosphorus, ALpotassium content of soil, some soil microbial char...acteristics (total number of bacteria and fungi, cellulose-decomposing and nitrifying bacteria, CO2-production of soil), and the amount of the plant biomass.
The readily available nutrient content of the calcareous chernozem soil increased due to the treatments, except for the change in the soil nitrate-nitrogen content, which did not measure up to the control due to the effect of high-dosage Bactofil.
The treatments also influenced the examined microbial characteristics of the soil positively. The artificial treatments significantly increased the total number of bacteria and the number of cellulose-decomposing and nitrifying bacteria. The low-dosage Bactofil significantly increased the number of cellulose-decomposing bacteria and both Bactofil dosage significantly increased the number of nitrifying bacteria. The measure of the soil respiration grew in all treatments, but significantly only in Ca(NO3)2 fertiliser treatments.
The quantity of the plant biomass was grew in a low-dosage Bactofil and significantly in the artificial fertiliser treatments. The highest plant biomass quantity was measured in the high-dosage artificial fertiliser treatment.
In the correlation analyses we found some medium positive correlation between the soil chemical, microbiological parameters examined, and the plant biomass in the case of both treatment-forms. 
Based on our results Ca(NO3)2 artificial fertiliser treatments on calcareous chernozem soil proved to be more stimulating regarding the
examined soil characteristics and the amount of the plant biomass, but the low-dosage Bactofil also positively influenced the majority of the
soil characteristics examined in terms of nutrient supply.

Comparative analysis of Staphylococcus aureus strains by molecular microbiology methods

Staphylococcus aureus is a very important pathogen for dairy farms and milk processing plants. Subclinical mastitis is often caused by this species, and it can contaminate bulk tank milk when milking cows are suffering from mastitis. Additionally, thermostable enterotoxins (SE) produced by some types of this bacterium can cause food poisoning.<>The aim of our research was to examine the number of S. aureus in bulk tank milk in two dairy farms and the enterotoxin-producing ability, genetic relation (pulsotype) and antibiotic resistance of S. aureus strains from different sources (bulk tank milk, udder quarter milk and environment).
The results show that the mean number of S. aureus of bulk tank milk of two farms significantly differed (P<0.05). Fourteen isolates were selected for further molecular genetic studies (five isolates were from bulk tank milk and nine isolates were from udder quarter milk). S. aureus was not recovered from the environmental samples. Three of the fourteen isolates (21.4%) tested by multiplex PCR were positive for SE genes. Two isolates carried one gene (seb) and one isolate carried two genes (seg and sei). The fourteen strains were classified into three pulsotypes and two subtypes at 86% similarity level. Isolates from bulk tank milk (n=5), were divided into 2 pulsotypes (A, C) and one subtype (C1). The isolates from udder quarter milk (n=9) belonged to three different pulsotypes (A, B, C) and two subtypes (A1, C1). The distribution of pulsotypes in the present study revealed genetic relationship between S. aureus isolated from udder quarter milk and bulk tank milk. This could be explained by the fact that in farms with a high number of infected cows, these cows could represent the main source of contamination. The results of the antibiotic resistance investigations show, that all strains were susceptible to methicillin, cefoxitin, lincomycin, tetracycline, erythromycin and sulfamethoxazole/trimethoprim. Thirteen out of fourteen strains were resistant to penicillin (A and C pulsotypes, A1 and C1 subtypes) and just one isolate was susceptible (B pulsotype) to all antibiotics tested.

Microbiological status of bulk tank milk and different flavored gomolya cheeses produced by a milk producing and processing plant

The microbiological quality of milk is important not only for food safety, but it can also influence the quality of dairy products. In this study, our aim was to assess the microbiological status of the bulk milk of a milk-producing farm, and some natural and flavored (garlic, dill, onion) gomolya cheeses made from pasteurized milk produced by ...their own processing plant. We determined the number of coliform bacteria, Escherichia coli, Staphylococcus aureus, and molds of three milk and eight cheese samples. The tests were conducted between July and September, 2017.

In bulk milk, the mean coliform count was 3.83±0.17 log10 CFU/ml; the mean E. coli count was 1.38±0.14 log10 CFU/ml; the mean mold count was 3.74±1.30 log10 CFU/ml; and the S. aureus count was <1.00 log10 CFU/ml, respectively. The mean coliform count in gomolya cheeses was 3.69±1.00 log10 CFU/g; the mean E. coli count was 2.63±0.58 log10 CFU/g; the mean S. aureus count was 3.69±1.35 log10 CFU/g and the mean mold count was 1.74±0.37 log10 CFU/g. The amount of coliforms detected in different flavored gomolya cheeses were significantly different (P<0.05). More than 10 CFU/g of E. coli was found only in the dill flavored cheeses, and S. aureus was found only in dill (3.66±1.86 log10 CFU/g) and onion (3.71±0.52 log10 CFU/g) flavored gomolya cheeses. Based on the obtained results, it was found that the amount of coliform bacteria and E. coli in bulk milk exceeded the limit set in regulation of the Hungarian Ministry of Health (MoH) 4/1998 (XI. 11.) and the amount of S. aureus was below the limit. For gomolya cheeses, the S. aureus count exceeded the limit. The amount of coliform bacteria remained above the limit in cheeses, except for the garlic flavored gomolya cheese. In cheeses, a larger E. coli count was detected than in the bulk milk, but there is no specific limit for cheeses in the regulation. The mold count exceeded the limit specified in the regulation in cheeses, but a lower value was detected relative to milk.

The results show that, in the case of bulk milk and gomolya cheeses, certain detected quantities exceeded the limit values set forth in regulation of MoH 4/1998 (XI. 11.). The results indicate an inadequate microbiological state of the raw material and the finished products. The reasons for these are due to reduced technological hygiene or the inappropriate handling of raw material and finished products. In this study, we have summarized the results of our preliminary studies, which can provide a basis for further hygiene studies.
