  • Xanthomonas resistance in Hungarian 4 spice pepper varieties

    With a view to further enhance the reputation of Hungarian spice pepper it was necessary to improve resistance to the bacterium Xanthomonas campestris pv.vesicatoria, the most dangerous pathogen of pepper varieties. From among the familiar resistance genes in Hungary only the gene Bs-2 could provide sufficient protection against the aggressiveness spectrum of the bacterium species X.c.pv. vesicatoria. The first results of the resistance breeding are the spice pepper varieties Kaldom and Kalorez. In addition to the Bs-2 gene attempts are also being made at building in a gds gene into pepper, a gene creating a general defense system, a different strategy towards Xanthomonas campestris pv. vesicatoria.

     

  • Comparison of Xanthomonas arboricola pv. juglandis isolates from walnut trees grown in Romania and Hungary

    Among diseases that affect walnut, bacterial blight is considered the most important one in all walnut growing areas both from Hungary and Romania. For the determination of susceptibility/resistance of walnut cultivars in our posterior work planned, 61 bacterial isolates were collected from walnuts showing symptoms of blight, with the purpose of isolating Xanthomonas arboricola pv. juglandis (Xaj). The characteristic Xaj colonies, frequently present as saprophytes in the infected plant tissues, were separated from other bacteria according to their morphology, yellow colour, hydrolysis of starch, and oxidation of glucose. All isolates were tested for pathogenicity by hypersensitive reaction on tobacco leaves (Nicotiana tabacum L.), bean pods (Phaseolus vulgaris L) and unripe nuts (Juglans regia L.). Determination of taxonomy of the selected isolates denotes a possible subdivision (races, biotypes) of Xaj occurring in different geographical areas, API 20NE and API 50CH kits were used. Hungarian and Romanian strains showed a high degree of similarity of carbohydrate utilization but slightly differed from the type strain. All were Cu-sensitive.

  • The genetic background of resistance to common bacterial blight in newly identified common bean lines on the basis of inheritance studies

    Common bacterial blight (CBB), caused by Xanthomonas campestris pv. phaseoli (Xcp). is a major disease problem of common bean (Phaseolus vulgaris L.). The inheritance of resistance in Xrl and Xr2 lines to two isolates of Xcp was studied in the F2 and F3 popu­lations from the crosses between these lines and the Masay variety (susceptible to Xcp). Segregation patterns indicated that different single recessive genes presumably in coupling phase linkage determined the resistance to the HUN and EK-1 1 strains of Xcp in both lines. The presence of some minor, modifying genes beside the monogenic genetic background of resistance was also observed. Xrl and Xr2 lines represent valuable new monogenic genetic sources in resistance breeding to CBB.

  • Epidemiological survey of Xanthomonas arboricola pv. juglandis and Gnomonia leptostyla on natural population of walnut (Juglans regia) in eastern Transylvania

    In Romania, walnut Juglans regia L. is an important fruit crop, although most of the fruit production comes from non-grafted walnut trees, which are natural hybrids. Breeding programs have been launched during last 30 years to develop new cultivars with uniform fruit quality. In addition, foreign cultivars have been introduced and tested to establish a valuable walnut genepool. To improve the present assortment of generative rootstocks in walnut and to examine the infestation level, a long term survey was carried out in Eastern Transylvania. The main physical characteristics of fruits and its variation to the infestation level were considered. The cumulative distribution of Xanthomonas arboricola pv. juglandis and Gnomonia leptostyla were relatively low and the maximum value was around 15%. The highest infestation of husk with X. arboricola pv. juglandis was observed for roundish forms and differences were statistically significant comparing with other phenotypes. Infestation with G. leptostyla was similar for roundish, elliptic and thwarting egg-shaped phenotypes, while the husk infestation for egg-shaped phenotypes was not observed. The walnut population studied in our experiment can be considered as a genetically valuable population. More than 20% of them have upper class quality fruits with at least or more than 50% of nutmeat. Do to the large scale climate variation in Eastern Transylvania and the high humidity favourable for pathogen infestations, these population can be considered resistant and well adapted to abiotic and biotic factors.

    In ecological farming systems farmers can't use chemicals against pests. In ecological plant protection the aim is to prevent diseases; if it is not possible the use of allowed materials are permitted. Until now there haven't been enough effective and environmental friendly materials for seed treatment in organic farming. Seed borne diseases of tomato and pepper can cause serious losses in yield, so finding appropriate inhibitors has a great importance. Different materials were tested against these bacterial strains for seed treatment in this study. In vitro trials have shown that vinegar, cider vinegar, red wine vinegar and white wine vinegar have inhibiting effect against the causative agent of bacterial canker (Clavibacter michiganensis subsp. michiganensis), bacterial speck (Pseudomonas syringae pv. tomato) of tomato. These materials also have inhibiting impact on the causative agent of bacterial spot of pepper (Xanthomonas campestris pv. vesicatoria). Seed treatment with (natural alkaline material) sodium hydrogen carbonate (NaHCO3) had no effect on the examined bacterial strains. Among examined essential oils cinnamon oil seemed to be the most effective, but all oils decreased the germination ability. Thyme and savory teas were effective against Pseudomonas syringae pv. tomato. Other examined materials had insufficient bactericide impact (sucrose, NaCI, ethanol, valerian extract, peppermint tea). The germination test has shown that examined vinegar types don't decrease germination ability.

  • Long term investigations of flowers and leaves on mainly non-domestica plums

    The author dealt with plum species representing different eco-geographic areas by their genetic adaptation and their hybrids, as European (P. domestica, P. italica, P. cerasifera), Asian (P. salicina, P. simonii, P. ussuriensis), American (P. americana, P. besseyi, P. munsoniana, P. tomentosa). The rootstocks of the trees examined were seedlings of C. 679 myrobalan with the exception of Laroda and Santa Rosa II, which were grown on three different stocks: seedlings of C. 174 myrobalan, C. 449 bitter almond and C. 471 sweet almond. The size of peduncle, length of pistil, stamen number per flower, relative stamen number (SN/PL) have been suitable for description and distinction of varieties. Similarly shape of leaves, length of petiole, length and width of blade helped the identification.

    The ratio of the dimensions of leaves, length of petiole and of leaf blade, also contributed to the distinction of European, Asian and American plum species, notwithstanding their relations with ecological conditions as well as historical, technical properties, pomological features, etc. Computed indicators (relative stamen number and shape-index of leaves) also have been useful data.

    Significant correlations have been found between colour of nectaries and mean values of variety-groups. The potential values of non-European varieties for purposes of commercial production could be forecasted from the point of view of quality, ecological, pomological as well as market value. It is important, however, to know the effect of the rootstock and growing site as well as their interaction, on the one hand, whereas the resistance or tolerance of the varieties as limiting factors, at least to the sharka (Plum pox) virus, Xanthomonas pruni, on the other hand (cf. Surányi & Erdős, 2004a and 2004b).

  • General defense system in the plant kingdom III.

    Our observations regarding the symptoms not fitting into, significantly differing from the hypersensitive defense system, which we noticed during the judgment of several plant species, symptoms provoked on several million plants have constituted a unified entity. They have provided evidence for the existence of a different plant defense system. We called this so far unknown basic response of plants to biotic effects as general defense system. This system defends them from the attack of numerous microbe species in the environment.

    The evolutionary intermediate phase between the general and the specific, the two defense systems is the susceptible host—pathogen relation. The vertical resistance system of plants escaping from the susceptible host—pathogen relation, based on specific hypersensitive reaction also suggested the existence of a more original, general defense system and the susceptible host—pathogen relation developed as a result of the collapse of that system.

    The evolutionary relation of the two defense systems is proved by the only recessive inheritance of the older general defense system and in the majority of cases dominant hereditary course of the specific defense system. In our experiences, the modifying genes of the recessive general defense system, in most cases, are behind the specific defense systems, which are known to have monogenic dominant hereditary course and react with hypersensitive tissue destruction. This seemingly striking genetic fact is explained by the following: the general defense system less dependent on environmental effects regulates much faster pathophysiological reaction than the specific resistance genes strongly dependant on environmental effects coding dominant hypersensitive reaction.

    The general and specific defense reactions, the processes excluding the microbes attacking plants with compacting of cell growth and tissue destruction, which mean two opposite strategies, building on and regulating each other constitute the entity of resistance to plant disease.

    In addition to successes achieved in certain varieties in resistance breeding based on a defense reaction of host plants involving hypersensitive tissue destruction, resistant varieties putting a very strong selection pressure on pathogens have selected more and more aggressive types of pathogens. The never-ending race between plant and pathogen resulting from this can only be controlled by a defense system characterised by a different strategy. In each of the plant species that we bred a defence system was found, which contrary to hypersensitive reaction strives to keep the tissues at all costs and is not pathogen specific. This is implied in the term general defense system.

     

  • Examinations of potential environmental friendly materials against tomato and pepper patogens

    In organic farming systems the focus is on prevention with regards to plant protection. To follow the rules of Good Agricultural Practice one is able to avoid serious yield losses; if it is not possible the use of allowed materials are permitted. Organic farmers have less material to protect their plants so it is necessary to find effective potential materials. Bacterial and fungal diseases of tomato and pepper can cause serious losses in yield. Different materials were tested against some plant pathogen bacterial (Clavibacter michiganensis subsp. michiganensis, Pseudomonas syringae pv. tomato, Xanthomonas campestris pv. vesicatoria and fungal (Phytoptora infestans, Rhizoctonia solani, Sclerotinia sclerotium) strains in order to find potential materials in the field of organic seed treatment. In vitro trials have shown that vinegar, cider vinegar, red wine vinegar, white wine vinegar, cinnamon and thyme oil have inhibiting effect against the causative agent of bacteria and fungi. Germination test has shown that examined vinegar types do not decrease germination ability if the concentration is low, but in higher (more than 5%) concentration it ruins the germination ability. Even in 0,5% concentration of red — and white vine vinegar have good effect on germination capacity.
