  • Sequence stability at SSR, ISSR and mtDNA loci of common millet (Panicum miliaceum) from the middle ages

    Seed remains of medieval millet, recovered from a 15th century layer (King’s Palace, Budapest, Hungary), showed reddish yellow grain color after rehydrating on tissue culture medium that was close to grain color of modern cultivar Omszkoje. aDNA of medieval c. millet was extracted successfully, analyzed and compared to modern common millets by ISSR, SSR, CAPS and mtDNA. Analyses of fragments and sequences revealed
    polymorphism at seven ISSR loci (22 alleles) and at the 5S-18S rDNA locus of mtDNA. CAPS analysis of the 5S-18S rDNA fragment revealed no SNPs in the restriction sites of six endonucleases TaqI, BsuRI, HinfI, MboI, AluI and RsaI. Sequence alignments of the restriction fragments RsaI also revealed
    consensus sequence in the medieval sample compared to a modern variety. Morphological characterization of twenty common millet (Panicum miliaceum L., 2n=4×=36) cultivars and landraces revealed four distinct clusters which were apparently consistent with the grain colors of black, black and brown, red, yellow, and white. In the comparative AFLP, SSR and mtDNA analysis modern millet cv. ‘Topáz’ was used. AFLP analysis revealed that extensive DNA degradation had occurred in the 4th CENT. ancient millet resulting in only 2 (1.2%) AFLP fragments (98.8% degradation),
    compared to the 15th CENT. medieval millet with 158 (40%) fragments (60% degradation) and modern millet cv. ‘Topáz’ with 264 fragments (100%). Eight AFLP fragments were sequenced after reamplification and cloning. Microsatellite (SSR) analysis at the nuclear gln4, sh1, rps28 and rps15 loci of the medieval DNA revealed one SNP (single nucleotide polymorphism) at the 29th position (A to G) of rps28 locus compared to modern millet.
    Mitochondrial (mtDNA) fragment (MboI) amplified at the 5S-18S-rDNA locus in the medieval millet showed no molecular changes compared to modern millet. The results underline the significance of survived aDNA extraction and analysis of excavated seeds for comparative analysis and molecular reconstruction of ancient and extinct plant genotypes. An attempted phenotype reconstruction indicated that medieval common millet showed the closest morphological similarity to modern millet cultivar Omszkoje. 

  • Investigation of the decomposition and leaching dynamics of Salix, Populus and mixed leaves in the area of Lake Balaton and Kis-Balaton Wetland

    In lakes and wetlands, leaf litter input from the coastal vegetation represents a major nutrient load and plays a basic structural and functional role in several ecosystems. In Hungary, at the banks of lakes and wetlands, Salix and Populus trees are the most common species. In an experiment in Lake Balaton and Kis-Balaton Wetland between 16 November 2017 to and 3 June 2018, the decomposition rates and leaching dynamics of Salix, Populus and mixed leaves (50% Salix and 50% Populus) were investigated. Total nitrogen and phosphorus content of biomass samples were measured at the beginning and end of the experiment for the leaching dynamics experiment. We found that litter mass losses (Salix, Populus and mixed leaves) were not significantly different between the two mesh size litterbags and between Lake Balaton and Kis-Balaton Wetland. Different amounts of the total nitrogen and phosphorus leaching from Salix, Populus and mixed leaves were detected. The total nitrogen contents of the plant samples were around 8-18% at the end of the investigated period. Slightly higher values were measured compared to phosphorous (27-29%).

    The experiments were performed in the University of Debrecen Centre for Agricultural Research Institute of Nyíregyháza of 1500 m2 asparagus plantation in 2011. We were determind the number of shoots, plant height, fold thickness and yield of asparagus hybrid. I studied to the effects of various cropyear is addition Grolim hybrid asparagus growth and development. Compared to previous years, 2014 was unusually wet and cold weather conditions which causes the plantation has developed outstanding. The soil temperature early and dynamic increase due to, the harvest had started a one month earlier 2014 than the 2013. The experiment asparagus harvested from lower yields as a result of the extremely dry spring in 2015.

  • The effects of agrotechnological factors on winter wheat yield in humid cropyear

    The effects of crop rotation, nutrien supply and crop protection technologies, as well as the appearance of the main ear- and leafdiseases
    (powdery mildew, helminthosporium leaf spot, leaf rust, fusarium) were studied on the crop yields of winter wheat variety MV
    Pálma during the 2009/2010 crop year. The experiments were conducted in triculture (pea – wheat – corn) and biculture (wheat – corn), at
    five nutrition levels, with the use of three crop protection technologies (extensive, conventional and intensive) at the Látókép Research Site of
    the University of Debrecen, Centre of Agricultural Sciences. Our results proved that the appearance of leaf- and ear-diseases were
    significant in the wheat cultures during the 2009/2010 crop year, because of the rainy, warmer than usual weather, the lodging, and the huge
    vegetative mass developed. The most severe infections by the four examined diseases after pea and corn pre-crops were observed at
    extensive crop protection levels, when fertilizers were used at the highest dose.
    Following corn pre-crop, in the case of all the three crop protection technologies the maximum rate of wheat yield results were achieved
    at N150+PK level. The highest yield was reached at intensive crop protection level (6079 kg ha-1). In triculture, in case of all the three crop
    protection technologies the maximum yields were achieved at N50+PK level; in extensive technology 5041 kg·ha-1 yield, in conventional
    technology 6190 kg ha-1 yield was realised, while in the intensive technological model the yield was 7228 kg ha-1.
    The relationship between yield and fertilizer amounts, the rate of pathogen contaminations, crop protection technologies and pre-crops
    was defined with correlation analysis in case of different crop rotations during the 2009/2010 crop year. Based on the results of the
    experiment, we found that in stands after corn pre-crop strong positive correlation was established between the crop protection level and the
    crop yield (0.543), the nutrient levels and the emergence of the four examined pathogens, and between the nutrient levels and the yield
    (0.639). Extremly strong positive correlation was observed between crop protection and yield (0.843) in triculture. Strong positive
    correlation was detected between the nutrient levels and the presence of the four examined pathogens, as well as between nutrient and
    lodging (0.688). Strong negative correlation was between the crop protection level and the four examined diseases both in biculture and
    triculture.

  • The Effect of Grazing Intensities on Magnesium Contents

    Research was carried out on two areas of grassland in Hortobágy National Park, Hungary. Two herds of Hungarian Grey Cattle were kept in free range grazing and the effects of grazing pressure on the magnesium content of soil and ryegrass (Lolium perenne L) were determined.
    Changes of plant available and total soil magnesium content under different grazing intensities did not show any evident tendency on the investigated grasslands. Different amounts of cattle faeces, urine and trampling had no effect on the magnesium concentration of ryegrass. We conclude that the magnesium content of ryegrass on both grassland sites as moderate grazing and overgrazing matches the requirements of cattle. Symptoms of magnesium deficiency of cattle will likely not appear.

  • Comparative analysis of soil analysing datas on different sempling-plots

    Hibrid maize is cultivated on larger plots, therefore the sown areas of hibrid maize are heterogeneous from a pedology aspect. Heterogenity causes problems during tasseling, chemical plant protection and harvest. The heterogenity of sown areas can be compensated by fertilization which is based on soil analysis. We carried out research into change of the soil on four soil types from 1987 to 2005.
    There were no significant changes in pH, hydroiodic acidity, CaCO3-content, humus-content on meadow chernozem soil. We detected equalization of salin content in the examined soil layers. There were no significant changes in the measured values on chernozem meadow soil and solonetz meadow soil in 2005. We discoverd equalization of saline content on chernozem meadow soil, but the changes were not as obvious as the changes on meadow chernozem soil. We found salinization in the 30-60 cm soil layer on type meadow soil that may be due to water movement.

    In the frame of a common “Hungarian-Ukrainian Intergovernmental S&T Cooperation Programme” which title is “Change of soils ecological characteristics of Ukraine and Hungary in the conditions of anthropogenic transformed ecosystems and optimization of biological processes of plants primary feeds elements mobilization” a Workshop was held in Debrecen. The member institutes of project participated with different presentation in this program.
    The title of Workshop was: “Anthropogenic effect on the properties of Middle and Eastern European chernozem soils and on the sustainable agricultural production”.
    The aim of the Workshop was to give relevant information about the present situation of the Middle and Eastern European Chernozem soils, especially emphasize the effect of different loading on the quality (properties) of chernozem soils. With the Workshop we would like to create a tradition for discussion about the anthropogenic effect on the soil properties and through it on the productivity of different soils. It was a forum for discussion of research results related to problems and possibilities for prevention of soil quality. With this possibility we would like to contribute to the sustainable agricultural production.
    The papers were read for the publisher and we would like to show them in a separate supplement of Journal of Agricultural Sciences, Acta Agraria Debreceniensis as one of the results of the project.
    The papers comply with the requirements of the scientific issue except those two which show the university and the department of the Ukrainian partner taking part in this project.

  • Q-PCR analysis of the resistance of Hungarian Botrytis cinerea isolates toward azoxystrobin

    The genes being in the mitochondrial DNA primarily encode the enzymes of cellular respiration. Fungicides belonging to the family of quinol oxidase inhibitors (QoIs) play on important role in the protection against several plant diseases caused by fungi. These fungicides bind to the cytochrome bc1 complex so they block electron transport between cytochrome b and cytochrome c1. This way these fungicides inhibit the ATP synthesis consequently they inhibit the mitochondrial respiration. The QoI resistance has two mechanisms. One of them is the point mutation of the cytochrome b gene (CYTB), e.g. the substitution of a single glycine by alanine at position 143 results in high-resistance. The other is the cyanide-resistant alternative respiration sustained by the alternative oxidase.
    In a cell there are several mitochondria. The phenomenon when the genomes of all mitochondria in the cell are identical is called homoplazmy. If in the cell there is wild and mutant mitochondrial DNA this is called heteroplasmy. Whether the mutation in the mitochondria causes fenotypical diversity or does not depend on the dose, i.e. it depends on the percentage of the changed mitochondrials. During our work we investigated Botrytis cinerea single spore isolates which have been collected in 2008-2009 on different host plants. Our goal was to decide whether heteroplasmy influences the level of resistance. We managed to detect the change of the level of heteroplasmy, so the change the level of the resistance due to the treatment with fungicide.

  • Estimation of the vine-shoot yield in the Gyöngyös district

    Every year, a significant amount of vineyard prunings is generated in Hungary. Instead of utilization them, it is burned directly on the field in spite of the limitations of legalization highly pollute the environment in recent decades. The vineyard pruning itself or with other by-products from pomology and forestry can be significant amount of fuel source. However, before the planning to utilization the essential task is to estimate the potential. This study examine that how many vineyard is generated in the micro-region of Gyöngyös in every year on appointed vineyard.

    In order to vine-branch potential calculate more accurate data was necessary so the products had to be determined for all plant. Thus, the biomass resource and the utilization become plannable. As the result of the experiment we can estimate that 1,5 t/ha vineyard pruning are generated. It means more than 9 thousand tons pruning in the micro-region of Gyöngyös. This amount can be convert into fuel source more than 150 thousand GJ heating value.

    The results evince that the vineyard pruning is important fuel source in every year in the micro-region of Gyöngyös. Further usage is suggested for other regions in other size.

  • Effects of the development of production factors on productivity

    In every economic sector, the enacted and expected quality of the products strongly affects its market entry and subsistence. The aim of management is success, competitiveness and subsistence that require adequate human and material resources and enough production stock. We must also notice that the quality of tools and instruments significantly influences work productivity. This applies to both plant production and animal husbandry.

    The author researched milk production factors with methodical observation, document analysis and interviews. Data were collected with a ten-year interval, thus this assay can highlight the field, volume and sources of investments during this period and that the production stock increased in most farms and decreased in few. The data also shows the influence of investments on work productivity - there are differences between farms of equal size, so there are still ways of development.

  • Improved soil and tomato quality by some biofertilizer products

    The use of microbial inoculums is a part of sustainable agricultural practices. Among various bioeffectors, the phosphorus-mobilizing bacteria are frequently used.

    The objective of this study is to investigate the effect of some industrial biofertilizer inoculums, of containing P-mobilizing bacteria on the quantity and some quality parameters of tomato fruits. Spore-forming industrial Bacillus amyloliquefaciens FZB42 (Rhizovital) as single inoculums and combinations with other Bacillus strains (Biorex) were applied on Solanum lycopersicon Mill. var. Mobil test plant. Soil microbial counts, phosphorus availability, yield and fruit quality, such as total soluble solids (TSS) content and sugars (glucose, fructose) were assessed. The results found that single industrial inoculums of FZB42 product had positive effect on P-availability and fruit quality in the pots. Fruit quality parameters, TSS content, soluble sugars were significantly improved (p<0.05). Such better fruit taste was correlated significantly by the most probable number (MPN) microbial counts. Use of such bioeffector products is supported by the positive interrelation among measured soil characteristics and inside healthy quality parameters of tomato fruits.

  • Fungi detected in trunk of stone fruits in the Czech Republic

    This study was focused on detection of the spectrum of fungi in the wood of stone fruits using molecular genetic methods. Samples were obtained from apricots, plums and sweet cherry trees from region of Moravia, one sample was obtained from Myjava (Slovakia). Segments of symptomatic wood were obtained from dying stone fruit trees with very significant symptoms. This study describes detection of the fungi in the wood of 11 trees in general in 5 localities. The cultivation of the fungi from symptomatic wood and sequencing of ITS was carried out. Eleven fungal genera were determined in the stone fruits wood, particularly Irpex lacteus, Fomes fomentarius, Neofabraea corticola, Calosphaeria pulchella, Cytospora leucostoma, Phellinus tuberculosus, Stereum hirsutum, Collophora sp., Pithomyces chartarum, Aureobasidium pullulans,Fusarium sp. The results of this study demonstrate that the reason of declining of stone fruit trees in Moravia is caused probably by trunk pathogens.

  • Effects of fermented chicken manure products on the N mineralization rate of the soil using the incubation method

    In our study, the effect of fermented and specially added poultry manure products (superabsorbent polymer (SAP), bentonite and Aegis as a mycorrhizal inoculum) were investigated in a short soil incubation experiment – at 60% water capacity level - on sandy soil. Soil samples were collected from two layers of the incubation pots after the second and fourth week to check the status of the tested products and the processes in the soil. The pH and the electric conductivity (EC) of the samples were measured using an electrochemical method, while the ammonium and nitrate content of the samples was determined with a photometric method. Soil pH and EC values slightly were decreased during the experiment. Our results pointed out that the increasing dose of SAP caused lower soil pH. The nitrate content of the soil did not change significantly during the experiment. It was found that the increasing SAP content in the products, due to its cross-linked structural property, protected the nitrate ions from leaching. Our results suggest that applied SAP does not bind the nutrient ions so tightly in its structure that it competes with the plant for uptake.

  • Evaluation of long term experiments from a new aspect

    During our work, we developed a new, simple method to show the effects of fertilization on yield, which can both be applied over the long term as well as in series of independent experiments.
    During the testing of this method, at the experimental farm of the Debrecen University Center for Agricultural Sciences at Látókép on a chernozem soil with lime deposits, we examined the fertilizer reaction of maize hybrids between 1989 and 1994. The treatments were: winter tillage, plant density of 70-80 thousand, unfertilized, N 120, N 240 kg/ha fertilized treatments, long term experiments using Dekalb 524 and Volga SC hybrids in long term experiments.
    Four parameters are shown in the model. In the examined period TRmax represents the greatest yield in the fertilized treatments, NT the yield in unfertilized treatment, k the „efficiency of fertilizer” to NT and b the depression-coefficient, where the expected value is zero. The expected grain yield of the fertilized treatments (Y), in the function of the unfertilized grain yield (x) is the following:

    The parameters were determined using the Monte Carlo method, in the optimizing process the sum of deviation square was minimized. The correct conformation of the functions was determined by the greatness of the R-value and the standard error. We found that during six years of testing, the tendency of fertilization efficiency was similar in the case of both hybrids. There was an unfavorable weather interval and, in these years, the yields were low, fertilization did not have an effect and moreover, in extremely bad conditions resulted in an obvious yield decrease. With the  improvement of conditions, which in the case of our country means an increase in precipitation, the efficiency of fertilization increases and reaches its peak at 13-14 t/ha. At this point, the yield increasing effect of fertilization is 4-4,5 t/ha. If the yield of the unfertilized treatments increases from 8-9 t/ha, then the efficiency of the applied fertilizer decreases.
    Most likely, the k and b parameters depend on the soil of the experimental location (nutrient and water management) and on the amount of  pplied fertilizer and the characteristics of the  hybrid. With the increase of fertilizer dosage the k-parameter also increases. The greater value though does not obviously mean a more favorable situation. It is true that in medium and good years this means great fertilizer efficiency, but in low or extreme precipitation conditions it also means greater risk. With the increase of the k-parameter, the yield deviation also increases which, from a cultivation point of view, is quite unfavorable. If the value of the b-parameter is other than, zero then the effect is clearly unfavorable, because with the increase of this value, the yield decrease is also greater. The fertilizer reaction of the two examined hybrids can be well characterized by these two hybrids.
    Examining the six years, our created model estimated the effect of fertilization on the yield accurately and with a high degree of safety. Both in highly unfavorable and extremely good years, it gave an exact estimate. In our opinion, it can be used well to evaluate the effects of fertilization on yield in the future.

  • Microbiological and Chemical Characterization of Different Composts

    Composting of agricultural waste is considered particularly important from the point-of-view of environmental protection. Degradation of organic substance results in a significant reduction of waste volume.
    The end product of the composting process, mature compost, can be used as soil coverage against excess loss of wastes, for mulching, for organic manure etc. The problem of composting has come into limelight in environmental studies and in agriculture.
    The quality of the mature compost is determined by physical, chemical and biological parameters of the composting process which, in turn, depend on initial composition of the raw materials, the technology, e.g. regular mixing and moistening and on environmental factors. Quality is the key question in compost use.
    We studied the composting process in compost windrows of different raw material composition. We measured temperature, humidity content, pH, organic substance content, nitrogen and carbon content.
    We counted the number of bacteria, microscopic fungy, ammonifying and cellulose decomposing microorganisms. We directed the composting process with turning weekly (to provide oxygen) and watering (to provide humidity content 40-60%).
    We set up windrows of 1 m3 volume from dry plant substances (cornstalk, pea straw, tomato stalk and crop, weeds) and cow manure not older than 1 week. The cow manure was used at ratios of 0%, 35%, 50%, 65% and 100%, respectively.
    We measured changes in compost temperature relationship with outside temperature until they were almoust the same. Humidity was 40-60% in most cases.
    At the beginning of the process, pH was slightly acidic-neutral; it later becomes neutral-slightly alkaline (pH: 6.93-8.02) as ammonia is liberated from proteins.
    At the end of the process, pH decreased again, due to humification.
    Organic substance content decreased as microorganisms mineralized them. Organic carbon content decreased gradually due to microorganisms used it as an energy.
    Total nitrogen content increased until middle of july and decreased gradually until than.
    The carbon/nitrogen rate were higher in the beginning, it decreased until july-august and increased by smaller degree until end of the process.
    The number of bacteria was higher in the first three weeks and between june-september. The number of cellulose degrading bacteria was the highest in the first three month, the number of ammonifying bacteria was the highest from the end of may until sepember.
    The number of microscopic fungy was significant in the second part of process, after july.

  • Hydro-cultural growing of cut flower in greenhouse

    The closed system hydro-cultural growing is environmentfriendly,  the chemical fertilizer used for nourishing material supply does not contaminate the soil water and it is possible to eliminate the chemical materials accumulating during the disinfection of the soil getting into the environment. It can be mechanized and regulated well so the optimum growing conditions can be fixed for the plant and as a consequence higher yield can be reached both for the carnation and for Zantedeschia and for Rosa. Growing method with saving both the nourishing material and the water. Taking all the above into consideration timing is easier and better, it can be programmed better than the traditional chemo-cultural growing.

  • Effect of plant extract against opportunist human pathogen soil bacteria

    Our experiments have repeatedly shown that the extract of ragweed (Ambrosia artemisiifolia) is an antifungal and antibacterial component.
    In our paper we tested of the biological activity of the herbal substance against opportunistic human pathogenic bacteria strains (Staphylococcus
    aureus 110003 and 25923; Staphylococcus saprophyticus 110008). Our laboratory tests show that the extract is bacteriostatic and in several cases bactericid. We can assume that from the the agricultural and public health aspects can be extremely dangerous weed, contains biologically active components and it may be suitable for the prepare of antibacterial agents.

  • Comparative study of special honey products and herbhoneys

    Honey has a positive effect on human body due to its high content of biologically active substances (e.g. monosaccharides, vitamins, enzymes, amino acids, polyphenolic compounds). The properties of honey depend on its botanical origin due to the bioactive plant components, mainly secondary metabolites that are included in honey made by bees from nectar. Herbally infused honeys are delicious products that combine the therapeutic action of herbs and honey. Additionally, herbs can provide nutrient fortified syrup for honey bees and protect them against diseases and other ecological threats.
    The aim of this study was to define the physicochemical properties of multifloral honey, herb enriched natural honey and herbhoney samples. We measured the moisture content, pH value, electrical conductivity and proline content. Although great diversity was observed in the basic properties of the examined products. In our study, we found that the electrical conductivity shows the significant differ between the groups. All the samples, including the herbhoneys passes the quality standards of honeys.
