Mutation induction in sweet basil (Ocimum basilicum L.) by fast neutron irradiation

Basil species are highly sensitive to exterior environmental conditions and its consequences lead to great economic and agronomic losses. In this research, a mutation method was optimized out for creating a new variety of Ocimum basilicum L., which could tolerate the extreme/extraordinary climatic circumstances or biotic stresses, such... as fungal diseases. Fast neutron irradiation was performed on the Hungarian commercial variety seeds with doses of 5 to 60 Gray and grown into fully developed plants. Numerous phenotypical changes like deformed congestion, leaf mutation, and low growth occurred, especially at higher dosages. Then to confirm whether the plantlets had mutation or not, and to detect the molecular variation and relationship, fingerprinting profiles of the developed mutant regenerants and donor plant have been assessed using ISSR markers. 115 loci were yielded, ranging from 0.2 to 1.5 kb, out of which 110 loci were polymorphic in nature, representing 95.6% polymorphism. The most suitable primer to determine the genetic diversity within the Ocimum species was the UBC-856 with 0.42 PIC and 4.1 MI values.

Fruit coloration of apple cultivars

One of the most important parameter of apple cultivars is the scale and the intensity of the coloration of fruits. Nowadays among the bicolour cultivars those are the most saleable which have the highest and darkest fruit coloration. In our three years old study cultivars born with bud mutation and cross breading were examined, which have super...ior coloration peculiarities than their parental cultivars. According to our results among the observed cultivars the cvs. ’Gala Decarli-Fendeca’, ’Galaval’, ’Wilton’s Red Jonaprince’, ’Early Red One’ and ’Red Cap Valtod’ reached favourable high fruit surface colour (80-100%) regardless the vintage effect. Concerning the colour intensity, that is the darkness of the red colour most of the cultivars (‘Gala Venus Fengal’, ‘Gala Fendeca-Fendeca’, ‘Galaval’, ‘Jeromine’, ‘Red Topaz’, ‘Early Red One’, ‘Crimson Crisp’, ‘Wilton’s Red Jonaprince’) showed excellent values (4,5-5,0). Based on the fruit surface colour and the fruit colour intensity a new index number, the fruit coloration index was created by us, which describes the coloration features in a complex way.

S-locus genotyping on stone fruits in Hungary: a review of the most recent achievements

Central Europe can be taken as a geographical and historical connection zone between the western growing countries and Asian gene centres of Prunus tree fruits. The determination of the S-genotype of stone fruit (mainly almond, plum, cherries and apricot) cultivars and landraces has both practical and theoretical significance. Our group has all...ocated complete S-genotypes for more than 200 cultivars and selections of almond, Japanese plum, sweet cherry and apricot. Among Eastern European almond cultivars, two novel cross-incompatibility groups (CIGs) were identified. S-alleles of a related species were also shown in P. dulcis accessions; a fact seems to be indicative of introgressive hybridization. Our results with Japanese plum clarified and harmonized two different allele nomenclatures and formed a basis for intensive international studies. In apricot, a total of 13 new S-alleles were identified from Eastern European and Asian accessions. Many Turkish and North African cultivars were classified into new CIGs, III–XVII. Results suggest that the mutation rendering apricot self-compatible might have occurred somewhere in south-east of Turkey and we were successful to confirm the presumed Irano-Caucasian origin of North African apricots based on the geographical distribution of S-alleles. In sweet cherry, new alleles have been identified and characterized from Turkish cultivars and selections. In addition, wild sweet cherry and sour cherry S-alleles were also shown indicating a a broader gene pool in Turkey as compared with international cultivars. We also used S-genotype information of Ukrainian sweet cherry cultivars to design crosses in a functional breeding program. Our results exhibit an increased number of S-alleles in tree fruit accessions native to the regions from Eastern Europe to Central Asia, which can be used to develop S-genotyping methods, to assist cultivation and draw inferences for crop evolution.

The importance of clonal selection of grapevine and the role of selected clones in production of healthy propagating stocks

Genetical alterations and phytosanitary status promote the variability and modify the appearance of vine. Old vine varieties in old vineyards are highly variable and well adapted to selection. Clonal selektion is based on a visual performance: valuable individuals (clones) are picked out according to visible symptoms or characters. The genetica...l stability of clones is proved by testing the vegetatively propagated progenies on the basis of morphological and molekular (SSR, AFLP, SMPL, RAPD) markes. Authors take great care of the visual phytosanitary selection as part of the clonal selection being the oreliminary step to develop pathogen-free propagation stocks. In Serbia (Vojvodina) the selection breeding has been carried on for several decades resulted in comparative clone trials with home and imported clones of Welsch Riesling, Chardonnay, Pinot gris, White Riesling. Among the clones of home selection SK.54 Welsch Riesling clone is the most valuable. Its clearing from pathogene is being carried on in an interregional IPA programme (HUSRB/0901/214/123) in Kecskemét. In Kecskemét, the centre of the Hungarian Danube vine region 5 vine clones have been registered (Cegléd szépe K.73, Irsai Olivér K.11, Kövidinka K.8, Hárslevelû K.9, Pannónia kincse K.56). Besides them 18 virus-tested clones have also been qualified.Works aiming at their complete exemption are going on in order to obtain clones free of propagation wood-borne diseases.

Test of the utility of apple retrotransposon insertion patterns for molecular identification of 'Jonathan' somatic mutants

Up until today, apple sport mutants proved to be indistinguishable from each other and their progenitors at the molecular level using random amplified polymorphic DNA (RAPD), amplified fragment length polymorphism (AFLP) and simple sequence repeat (SSR) marker techniques. This is not surprising, since the genomes of these somatic mutants differ... only in one or a few small regions that affect economically important characteristics, such as improved fruit colour, size, or flavour. In most cases, these genome differences are probably caused by retrotransposons which are able to convert their RNA transcripts to DNA with reverse transcriptase enzyme prior to reinsertion, but unable to leave the genome and infect other cells. Retrotransposon insertions can alter the expression of other genes and/or the structure of encoded proteins. The sequence-specific amplified polymorphism (S-SAP) technique is capable of revealing the genetic distribution of retrotransposable elements over the whole genome. The present study used this approach to try to characterize and distinguish 'Jonathan' somatic mutants via fingerprinting, which is an unsolved problem.

Comparative study of some nutritionally important components on different cabbage (Brassica oleracea L.) genotypes

Vegetables of Brassicaceae are especially important because of their inner values, which play a provably positive role in curing and preventing diseases. Cabbage (Brassica oleracea L. convar. capitata provar. capitata) is still today one of the most frequently consumed green vegetables, which is known to brak...e the absorption of carcinogenous substances and to obstruct cell mutation. Different cabbage cultivars were compared growing under the same circumstance. The level of different carbohydrate fractions and some biologically active quaternary ammonium compounds were determined. In all cases we analyzed the antioxidant capacity of samples characterizing the free radical scavenging capacity.

Clearly detectable significant differences were found between the varieties tested, which appeared suitable for selecting the most precious varieties for human nutrition.
