Several plant bug species (Miridae) are important pests of crops and vegetables, thus monitoring them is of essential importance for effective pest control. During the current, preliminary study synthetic plant volatile combinations were tested in field conditions in Hungary in alfalfa fields. Beside semiochemical baited traps, sweep-netting wa...s also performed. In the experiments three plant bug species were found in higher numbers: Adelphocoris lineolatus, Lygus rugulipennis and L. pratensis. As a novel, interesting finding L. pratensis was attracted to phenylacetaldehyde baited traps. For all species, both males and females were trapped in all combinations. Sweep-netting and semiochemical baited traps showed different efficacy in case of the three species, as sweep-netting catches were highly biased for A. lineolatus, which indicates the higher efficacy of this method as compared to the tested semiochemical-baited traps. On the other hand, semiochemical baited showed better performance for L. rugulipennis and L. pratensis. For these species none of the tested combinations performed better than phenylacetaldehyde baited traps. The potential implication of results in view of monitoring are discussed.
The most limitating factor for successful transformation is the absence of high-yielding regeneration protocols. However, the anther-derived embryogenic culture is an optimal technique for genetic transformation and it has been widely applied in many important cultivars, but the necessity of further development of regeneration systems has been...proved. We attempted to produce somatic embryos on a wide range of genotypes from various tissues; leaves, petioles, stem segments. We started the examination of grapevine regeneration via organogenesis, succeeded in inducing shoot from the meristematic tissue of the base of bud by testing induction medium contained different concentrations of two types of hormones. To optimize the conditions of the Agrobacterium-mediated transformation, we studied the effectiveness of different Agrobacterium-treatments, the use of antioxidants and the sufficient quantity of kanamycin for selection of transformed cells.
The study examined the health status, growth type, berry morphology, berry quality, and gastronomical value of 60 heirloom tomato lines. In the middle of June, the level of virus infection was medium in the plant stock according to the assessments. 21% of the lines did not show symptoms of virus infection. During the vegetation period, the majo...r fungal diseases were Alternaria by the middle of August and Phytophthora from the beginning of September. In spite of the fact that no fungicides were applied, 13% of the lines were healthy and 36% showed medium infection at the end of August. Half of the lines had a strong, 23% of the lines had a medium growth vigour. The majority (74%) of the lines showed strong or medium lateral shoot growth and only 7% of them proved to be less prone to branching. In spite of the rapid growth, 20% of the lines had a weak main stem, the majority of the lines had a main stem medium strength. The different shapes occurred in different colours, in addition to the traditional red colour (53%), there were orange, yellow, wine-red (blackish), lilac, green, almost white or multicoloured varieties. Unfortunately, most of the lines were prone to longitudinal or circular cracking of the berries. Based on the first experiences, all tested variety types had more favourable utilization characteristics then the traditional tomatoes. Varieties with a thick flesh and harmonic acid/sugar ratio such as some of the ox-heart varieties are especially valuable. For drying, the elongated types proved to be the best. The hollow tomato is the most special type which can be utilized most diversely. It is especially suitable for making white tomato soup and, “Concasse" due to its easily removable placenta rich in jelly materials and its thick flesh. In addition, it can be used for preparing stuffed tomato.
In vitro cultures have widely been used in horticulture for rapid multiplication of new varieties and clones as well as to produce pathogen-free stock material. To improve efficient hardening and transfer in vitro grown grapevine plants were multiplied by cutting them into single-node internodes with the whole leaf. Microcutti...ngs including the shoot tips were rooted in granulated perlite moisted with tapwater under sterile conditions. After 2-3 weeks the rooted microcuttings were supplied by nutrients and hardened by gradual opening and finally by complete removal of the lids of jars or plastic boxes used for growth. Using this method microcuttings of Vitis vinifera cvs. „Chardonnay", „Cabernet franc", „Riesling" and „Sauvignon blanc" and the rootstock varieties Vitis riparia x Vitis cinerea cv. „Barrier" and Vitis berlandieri x Vitis rupestris cv. „Richter 110" formed new roots and shoots and 100% of the tested plants survived the acclimatization procedure. Similar results were obtained when perlite was replaced with rockwool-, or pit-pot blocks. This method may highly increase the efficiency of producing pathogen-free propagating material and new transgenic lines.
The VirEl protein plays a key role in the transport of VirE2 protein from the bacterium to the plant cell during crown gall tumor induction by Agrobacterium. The virEl gene of A. tutnefaciens pTiA6 was cloned into the plant transformation vector pTd33 yielding pTd93virEl that was introduced into A. tuniefaciens...em>EHA101 and used for tobacco transformation. The presence of the foreign DNA in the putative transgenic plants was confirmed by PCR analysis. Nine of the 41 transformed plants formed only small tumors following infection with the wild-type A. vitis octopine strain AB3. This property was inherited into the T1 generation. The expression of virEl gene in TI plants was demonstrated by Northern blot analysis.
Producing high quality apples in mature trees of the cv. 'Golden Delicious' is rather difficult because of the russeting of it's fruits, especially in seasons of high relative humidity and/or wetness.
Earlier experiments proved the possibility to decrease russeting by treatments of GA4.7 during a period after petal... fall.
Phyl-Gold, a product of Phylaxia Co. (ai. 10 g1-1 GA4.7) was applied to inhibit russet formation in fruit skin of 'Golden Delicious' apples. Four consecutive sprays were carried out (with 10 ppm a.i.) in weekly intervals, starting at petal fall and tank-mixed with the current pesticides of the plant protection program.
Due to the GA4.7 application the russet formation decreased at a rate of economical importance. As for the side-effects of treatments, fruit set was reduced slightly in trees treated, however, there was no consequent influence on return bloom.
