Storage of ram semen in gelatin supplemented extender
7-10Views:147The aim of our study was to examine how different gelatin concentrations affect ram semens viability in liquid storage at 5 oC for five days. Our hypothesis was if we add gelatin to the semen extender, than the viability of ram semen will be better in the extenders containing gelatin, than the control. We used two different semen extenders:1.5% UHT milk and 1.5% UHT milk + 5% egg yolk. We added 0; 0.5; 1.0; 1.5; 2.0% Dr. Oetker gelatin to the semen extenders. We stored the semen for five days at 5 oC and in every 24 hour we made sampling.
We stained the smears with Kovács-Foote staining and evaluated them with light-microscope. We categorized the cells in five groups like: live and intact cells, live cells with injured acrosome, dead cells, live head with dead tail and live tail with dead head. We used one-way analysis of variance (ANOVA) to assign how gelatin concentration affects the number of the categorized cells. On the fifth day, the viability was the best in the following semen extenders: 1.5% fat UHT milk + 1.0% gelatin and 1.5% fat UHT milk + 1.5% gelatin, but it was not significant (p>0.05). -
Examination for Susceptibility of CAE
33-36Views:160Caprine Arthritis Encephalitis (CAE) is a group of diseases in goats caused by a retrovirus, namely the Caprine Arthritis Encephalitis Virus. CAEV belongs to the lentivirus group within the retrovirus family. The CAEV is intimately associated with white blood cells, therfore any body secretions which contain white blood cells are a potential source of virus to other goats in the flock. Once goats are infected with CAEV, it remains infected for life and has to be eliminated.
Since not all goats infected with CAEV progress to disease, very important to test goats for infection using a serology test (ELISA, AGID, IDT). These serological tests demonstrate the presence of antibodies to CAEV in goat serum or detect the virus genome in the white blood cells. However, Swiss results (Dolf and Ruff, 1994) pointed out individual and species differencies in predisposition for this disease. This result was considered as a starting point to our examination. Microsatellite analysis was used in order to find whether there was any association between genotype and serological results, and to look for a marker associated with this disease. To date, altogether 135 goats have been examined. Unfortunately, a significant association between serological results and genotype was not found using the Chi2 test -
Impact of lameness on the milk production of ewes
5-7Views:138The aim of this study was assessing the impact of lameness on the milk production, somatic cells count and component of milk. We assess also impact of lameness on the order of entry into the milking parlour.
The experiment was carried at the farm, located in northern Slovakia. The farm keeps sheep Improved Valachian. Samples of milk were taken during two periods: May, July. It was taken 428 samples together. We recorded three groups by lameness- strong lame, slightly lame, non-lame ewes. We recorded also the order of entry of ewes into the milking parlour in milking row. The results were mathematically processed using the Microsoft Excel program and statistically evaluated by SAS.
We found significant statistical differences between months (P<0.0001) in all the above mentioned indicators. In July we recorded 26 ewes with slightly lameness and 18 ewes with strong lameness. Other ewes were non-lame. Non-lame sheep had in July the highest milk yield (356±148 ml) and the lowest decrease in milk yield from May to July (-206±131 ml) compared with slightly (317±116 ml, -223±163) and strong (319±122 ml, -219±151 ml) lame ewes. However, these differences were not statistically significant. We have not identified statistically significant differences between groups in somatic cells count (logxSCC for non-lame: 4.83±0.608 in ml, slightly lame: 4.76±0.653 in ml, strong lame 4.71±0.787 in ml). Milk composition (fat, proteins, lactose) nor changes in the composition of milk that occurred between May and July were not affected by lameness of ewes. But lameness in July affected the change the order of entry of ewes in the milking parlour in July compared with the order of entry recorded in May.

Investigation of intersexual polled goats
11-15Views:186The sex reversal may occur in all mammalian species, but is connected to a favourable trait – the polledness – only in the goat. Later abnormal sex ratio was noticed in these goat populations, in which a part of the phenotypically male individuals was sterile. These males have two X chromosomes. In goats the PIS (Polled Intersex Syndrome) mutation is responsible for the absence of horns in homozygous and heterozygous individuals. This same mutation causes a female-to-male sex reversal, but only in the homozygous polled genetic female goats. The PIS mutation inhibits the expression of the FOXL2 gene which is responsible for ovary development, and a protein encoded by this gene inhibits the activity of the Sox9 gene. The Sox9 gene stimulates the development of the cells of the testis. When the FOXL2 gene is inhibited, the Sox9 gene is activated and transforms the ovary cells into testis cells. In our article we briefly introduce the morphological and chromosome investigations of three intersex individuals we found.

The effect of apoplastic pH on the nutrient uptake

The exudates organic acids of plant able to modify the pH, as well as the microorganisms also take part in mobilization of nutrients. The nutrient solve mostly in mildly acidic and neutral pH. The either assumption of utilization of nutrients is the uptake by roots and of course uptake to the cells to take part in metabolism. The pH of apoplast fluid determines the solubility and uptake of nutrients to the cells.
The aim of this study was to examine the effect of nutrient solution and apoplastic pH together with a bacteria based biofertiliser (Phylazonit MC®) on nutrient uptake and pH of apoplast fluid in case of nutrient solution grown plants in laboratory experiment. According to my results, the bicarbonate increased the pH of nutrient solution in due to influence the solubility and uptake of nutrients. The given bicarbonate to the nutrient solution and infiltrated into the apoplazma also modified the pH of the apoplast fluid of the test plants. The effect of bicarbonate and biofertilizer were different on the pH of the apoplast fluid and nutrient solution in nutrient solution experiment. -
Effect of anthocyanin-rich sour cherry extract on the level of IL-8 in LPS-induced endothelial cell
27-30Views:243The anthocyanin content of the Hungarian sour cherry is remarkable. Nutraceutical and pharmaceutical effects of the anthocyanins and their role in disease prevention have been studied extensively. Endothelial cells are involved in the pathogenesis of several inflammatory diseases. The objective of this work was to investigate pure sour cherry extract on human umbilical cord vein endothelial cells (HUVECs) as an inflammatory model. HUVECs were treated with 100 ng/mL lipopolysaccharide (LPS) and 50 mg/mL sour cherry extract or M199 medium as control. The optimal concentration range of the sour cherry extract was investigated and selected based on MTT assay measuring the conversion of the tetrazolium salt to formazan by mitochondrial dehydrogenases. The level of interleukine-8 (IL-8), a pro-inflammatory cytokine, was measured in Luminex MagPlex assay. LPS treatment significantly increased the secretion of IL-8. The pure sour cherry extract was able to attenuate this increment indicating the potent anti-inflammatory effect of pure sour cherry extract. Our results emphasize that pure sour cherry extract could reduce the LPS-induced inflammatory response thereby may improve endothelial dysfunction.

Effect of allithiamine on the level of hyperglycaemia-induced advanced glycation end products
41-44Views:308Diabetes mellitus is a rapidly growing public health burden in both developed and developing countries. Diabetes mellitus is accompanied by hyperglycaemia, which can cause tissue injury by several mechanisms. One of these is the formation of advanced glycation end-products (AGEs). In this study, the effect of allithaimine, a fat-soluble thiamine derivative, was investigated on hyperglycaemia-induced AGEs levels using human umbilical cord vein endothelial cells (HUVECs) as a hyperglycaemic model. HUVECs were isolated by enzymatic digestion, characterized by flow cytometer and treated 30 mM glucose plus allithaimine or thiamine or cell maintenance medium as control. Allithiamine was synthesized and purified. The structure of the synthesized and isolated compound was verified by reverse phase HPLC and MALDI-TOF. AGEs were evaluated by ELISA. Collectively, our results indicate that allithiamine can reduce level of the hyperglycaemia-induced AGEs similar to thiamine.

33-37Views:110This study was carried out at the Hungarian National Artificial Insemination Centre in order to evaluate the seasonal effects on the quantitative and qualitative parameters of sperm production using sperm analyser equipment. The volume of ejaculate (VE), percentage of living sperm (PLS), spermatozoa concentration (SC), motility score (MC), percentage of motile spermatozoa (PMS), speed of movement of sperm cells (SMS) and the percentage of sperm cells moving straightforward (PSMS) were collected and observed in three seasons (summer, autumn, winter) in Holstein-Friesian breeding bulls (n = 15). The sperm collection was done for three successive weeks in every season on the same day (summer: n = 41, autumn: n=39, winter: n = 42, altogether: n = 122). The same sperm samples were measured by sperm analyser equipment (HTM version 7.0, Danvers) for PMS, SMS and PSMS. The seasonal and the number of sperm collection effects were confirmed on semen characteristics by multiple analysis of variance (two ways MANOVA, Type III) using the program package of STATISTICA 4.5. The average values of VE were similar in the three seasons (summer: from 4.42 to 6.28 cm3, autumn: from 4.08 to 6.86 cm3, winter: from 5.43 to 5.71 cm3). The average values of the MC were similar in each of the three seasons (summer: from 3.66-4.00 M; autumn: from 3.66-3.77 M; winter: from 3.86-4.07 M). The summary of all effects for season (P<0.001), repeated sperm collections in the same season (P<0.05) and interaction of two traits (P<0.01) were established on the measured characteristics. The special effects (P<0.05) of a given season were calculated, excepting the VE and MC, on all of the measured characteristics. The special effect (P<0.01) of the repeated sperm collections in the same season was verified in only one case (SMS). It is not surprised at the interaction was established on the VE (P<0.001), PMS (P<0.05), and SMS (P<10.0). The results of present study suggest that seasonal effect on sperm quality of breeding bulls cannot be eliminated even at standard feeding and keeping conditions. Considering our results, we can recommend that the average values of three successive weeks in every season be used, to take advantage of seasonal effect on the quantitative and qualitative parameters of sperm production.

The effect of sunburn damage on the fruit quality parameters of apple (Malus domestica cv. Idared)
55-64Views:216In the present study the authors investigated the effect of sunburn injury on fruit quality parameters (cover colour, depth of tissue damage, fruit flesh firmness, dry matter content) of apple.
The symptoms of sunburn injury appeared as concentric rings, differing in colour from each other and the cover colour. This can be connected with the ratio of the injury. The authors observed the following colours on the fruit surface (from the epicentre of spots on the surface of the fruit) dark brown (strongly damaged), light brown (moderately damaged), pale red transition (weakly damaged), red surface cover colour (not damaged).
Sunburn of apple fruits is a surface injury caused by solar radiation, heat and low relative humidity. In the initial phase, a light corky layer, golden or bronze discolouration and injuries of the epidermal tissue appear on the surface exposed to radiation. Thus, it detracts from the fruit’s appearance, but in most of the cases it would not cause serious damages in the epidermal tissue. The depth of tissue damage is not considerable, its values are between 1.5-2.0 mm in general. It is commonly known, that tissue structure of the apple fruit is not homogeneous. Accordingly, the degree of injury shows some differences under the different parts of the fruit surface.
On the basis of the flesh firmness studies, it can be stated that the flesh firmness of the damaged parts increases due to the sunburn effect. This is due to the fact that the damaged plant cells die, the water content of the tissue decreases and it hardens. However, due to this reduction in the water content the dry matter content will increase. -
The effect of gossypol acetate on the fertility of boars
47-55Views:191This study evaluated the effect of gossypol acetate, a potential antifertility compound, on semen quality and testicular histopathology of boars. Six boars were allotted into two experimental groups. Half of the animals were fed a diet that was supplemented with 2 mg of gossypol acetate per kg body weight per day for 9 weeks, control animals received a gossypol acetate-free feed. During the experimental period, semen was collected weekly from each boar and semen parameters were recorded. The animals were then euthanized, testicular samples were collected and histopathological examination of the testicular cells, as well as morphometrical analysis of the seminiferous tubules, were performed. The percentage of spermatozoa showing tail abnormalities increased significantly (P=0.017) in the semen of boars fed gossypol acetate-supplemented feed, while several other semen parameters deteriorated without showing statistical significance. Gossypol acetate supplementation also led to a decrease (P=0.042) in the number of spermatogonia in the seminiferous tubules and an increase (P=0.020) in the number of vacuoles in the seminiferous epithelium, consistent with changes seen in cases of male reproductive toxicity. In conclusion, gossypol acetate negatively affected a number of semen characteristics and also had detrimental effects on the histopathology of the testes.

Basic Research for the Development of Fertiliser Spreaders
52-57Views:118The knowledge of the physical characteristics of fertiliser particles is essential for the constructors and operators of fertiliser distributors. Among physical characteristics, the most important are the frictional and aerodynamic properties for the description of particle movement. Adjustable angled slopes, shearing boxes and various rotating disks are used to identify frictional properties. We have developed a high precision shearing box with digital force measuring cells and a distance signaller (incremental transducer) that we use for slide tests efficiently. We measured the frictional characteristics of 6 different fertilisers: the inner coefficient of friction and the coefficient of friction on ten test surfaces most commonly used in machinery, and we specified the relationship between displacement, loading and the coefficient of friction. We can conclude that the material of the frictional surface significantly influences the force of friction.
However, our experience tells us that the shearing box is not suitable for the measurement of the inner friction, since the examined particles slide on the metal surface of the shearing box in a growing extent in the course of displacement, so it does not measure the real inner friction. Therefore, in our experiment we have developed rotating shearing equipment with a constant shearing surface to identify the inner friction. We tested the equipment with fertilisers and we identified the inner frictional characteristics of 6 different fertilisers. With the developed rotating shearing apparatus we could measure the real inner friction of the particles.
To identify the aerodynamic characteristics of granules, wind tunnels and free-fall tests are used. An elutriator have been developed for our investigation. We have used fertilisers for testing the measuring equipment and we have identified the aerodynamic characteristics of 6 different fertilisers. -
The effect Benefit PZ biostimulant on the fruit size and yield production of apricot varieties (Prunus Armeniaca)
16-18Views:147Fruit size is determined by the genetic makings of varieties the pomology and enviromental factors. The climate and the previous year’s harvested yield have a great effect on yield volume. It is not easy to harmonize the qualitative and quantitave parameters of the fruit. High yield usually causes smaller fruits.
The aim of our trials was to evalaute the effect of Benefit PZ biostimulant on fruit size. We examined two apricot varieties (Bergeron, Magyarkajszi C235) for the efficacy of the product. The three applications were made with 400 l/ha water with 2500 ppm. The optimal interval of application with Benefit PZ is during in the early phases of development. The size of the final fruits depends on the number of cells making up the fruits. The later application could not increase fruit size growth, there are no significant differences between the Benefit PZ sprayed and the control plots. The totally harvested fruit weight was 13.5% more on the trees of cv. Bergeron, it was caused by the higher fruit set. On the sprayed trees were 12.2% more fruit. Therefore were no differences between the average fruit weight. Fruit weights of the treated trees were 44.37 g on cv. Bergeron, 44.15 g on cv. Magyarkajszi C235 and on the no treated trees were 43.83 g as well as 45.33 g. -

Data on the bumblebee assemblages (Apidae: Bombus spp.) lives in lands under agri-environment commitment
31-35Views:257The goal of agri-environmental schemes (AES) and greening programs are protecting and increasing biodiversity in agricultural lands. The evaluation of effectiveness of AES needs further investigations. For the purpose of investigations, species and species groups should be selected which can indicate the effects of changes in landscape use on biodiversity. Bumblebees are good indicators for this purpose.
The role of bumblebees in pollination is well studied but in the case of different crops, much less detailed data are available. In 2018, bumblebee assemblages of 44 sites belonged to 8 different agricultural and semi-natural habitat types were studied in the surroundings of Sajószöged, Tiszaújváros and Derecske.
This study provides new distribution data of 8 bumblebee species in three 10×10 km UTM cells covering the sampling area. According to our results, the alfalfa and red clover fields and semi-natural grasslands has more species rich and abundant bumblebee assemblages than different crop fields (sunflower, oilseed radish and vegetable morrow) and can help protect bumblebee assemblages of agricultural lands. Based on the collected distribution and abundance data, the role of the bumblebees in pollination of the studied crops should be re-evaluated.

Effects of paraffine oil on leaf and berry mycobiota on two grape varieties
61-66Views:223Application of fungicides have advantages and also some direct or indirect disadvantages, such as imbalance and/or fungicide resistance in microbe population. To avoid these problems the development of alternative, eco-friendly methods like mostly spraying with oils are in the focus nowadays. The investigations of the effects of fungicides on microbiota in some cultivations can give a more complex view to this topic and developmental possibilities. In the present study, our aim was testing of the effects of paraffine oil (as alternative fungicide) on microbial properties (CFU and rate of filamentous fungi and yeasts) of Chardonnay and Kékfrankos leaves and berries.
Our results from 2014 showed that the application of paraffine oil as sole spray agent can decrease the presence of saprophytic filamentous fungi on the berries of Chardonnay (susceptible for fungal infections). In the case of Kékfrankos berries opposite properties were observed, which may be the result of the absorption of oil by the thick wax layer of this variety. The oil treatment did not affect the yeast population of Chardonnay and Kékfrankos berries contrary to negative effect of the regular pesticide treatment. The selective fungicide effect of paraffine oil against filamentous fungal population caused the accumulation of yeast cells in the mycobiota of grape berries. The careful use of this yeasts in spontaneous fermentation can improve the aroma profile of wines. The year of 2015 did no prefer the growth of fungi, therefore no interesting properties were detected in the mycobiota of grape varieties. The occurence of the harmful saprophytic filamentous fungi predicted to be increased in mild climate agricultures as the result of the climate change.
In summary, the paraffine oils are seem to be promising tools for the eco-friendly control of harmful fungi of grapes.

Tenability of ram semen
63-66Views:202It could be stated that the diluted semen of Awassi rams taken in the breeding season preserved its fertilizing abilityat different temperatures for different periods of time. The motility of spermatozoa kept at 23 vs. 8oC was checked daily. The largest spread of data was observed 24 hours after taking the semen, then the motility rate of cells showed a linear decrease. Motility results of fresh and frozen-thawed samples were compared also after heat resistance test and significant differences were found between these groups. Significant individual differences were observedin the sperm motility after heat resistance test.

Growth regulators influence on stability of shoots and ascorbic acid content at cadmium and nickel joint action
9-12Views:141It is shown that for a maize the most effective protector of cadmium and nickel influence was zeastimulin, for a pea – agrostimulin.A protective function of emistim C for both cultures was insignificant. It is set that zeastimulin is influential in the increased metals absorption of a root system, however substantially (on 15% for a nickel and twice for a cadmium) reduces their translocation to above-ground part of plants. Previous treatment of pea seed by agristimulin also intensified the accumulation of nickel roots on 60% and did not influence on cadmium absorption and toxicants translocation to tissues of assimilatory organs. The protector role of growth regulators to cadmium and nickel joint influence shows up in the increase of ascorbic acid maintenance in the roots cells of both species to 34%, where as in leaves – at a maize resulted in the lowering of vitamin С amount (on 28%), but at a pea – increase on 20%.

Species-specificity of the glutathione reduced form accumulation in the arboreal plants, testing influence of industrial emissions
105-110Views:89Conducted are researches of dynamics of glutathione reduced form maintenance in the leaves of arboreal plants which grow in the zones of different level of contamination by industrial emissions of“Red Lead Factory” LTD and ArselorMittal Kryvyi Rig. For comparison, similar investigations were conducted in the intact Botanic Garden in s.c.t. Sofievka. Findings testify to absence of negative influence of industrial emissions and, thus, the arboretum of botanical garden also can be examined as conditional control. It was stated that it is set a few strategies of active oxygen forms and their metabolites deactivation, appearing in cells under action of environment negative factors. To our opinion, fluctuation of glutathione reduced form maintenance can examine as an index of plants stability, on which investigational species can be distributed on steady and moderato steady.
