Polyphenol- and anthocyanin content changes effected by different fermentation- pressing and aging technologies

Different grape processing, fermentation and aging technologies were compared in our study on the white wine-grape variety Grüner Veltliner between 2012 and 2014 in Hungary,Cserszegtomaj. The vines are grown on brown forest soil on dolomite bedrock, stocks were planted 3x1 m row and vine space, respectively in our experimental area. The soil h...as slightly alkaline pH, the orientation of the vine rows are East-West. The training system is modified Guyot cordon, with 1 m trunk height and cane pruning method. After the harvest half of the yield has been put into the de-stemmer crusher before pressing while the other half has been pressed immediately (whole bunches). From the filtered and bottled wine anthocyanin, and polyphenol content was measured in 2013 and 2014. Another enological technology testing experiment has been set on aging of Grüner Veltliner in 2013. The wine was fermented with addition of fine lees from juice sedimentation. Traditional (racking only), battonage and fast ready-made aging technologies have been set together, each treatment in three replicates were observed.

Sampling experience in a cherry plantation

Experiments in a cherry plantation were performed to find out that samples taken from various parts of the foliage of the tree what extent represent the whole tree. One tree from three different cherry varieties was selected. In the selection, we tried to form a good representation of the foliage of the orchard. 8 sampling locations were design...ated on each tree, in two different heights and four different directions according to the points of the compass.We measured the main sizes of the fruits in three orthogonal dimensions with a digital slide gauge with an accuracy of 0.01 mm; the mass of each cherry by an analytical balance, with an accuracy of 0.001 g; the mass of the stones with the same analytical balance, and then calculated the stone-pulp ratio. The measured and calculated data are used to characterize the sample in question.

Production of transgenic carnation with antisense ACS (1-aminocyclopropane44-carboxy late synthase) gene

Dianthus chinensis and Dianthus caryophyllus varieties were tested for shoot regeneration from leaf and petal explants and transformed with Agrobacterium tuniefaciens strains (EHA 105 and LBA 4404) harbouring an apple derived ACS cDNA in antisense orientation in order to reduce ethylene production and influence the et...hylene dependant traits in carnation. After transformation regenerating shoots were selected on MS medium containing 50-75-100-125-150 mg/1 kanamycin and supplemented with 1 mg/1 BA, 0.2 mg/1 NAA. Transgene integration was proved by PCR analysis with npt II spcific primers followed by Southern hybridisation of DNA isolated from green shoots on medium containing 150 mg/1 kanamycin. Several putative transformants were subjected to RT-PCR in order to examine the npt 11 expression at mRNA level. Both the transformant and the non-transformant plants were potted into glasshouse to observe the effect of changed ethylene production on flowering time, petal senescence and vase life.
