Development of seed analyses by means of various matrix solutions and the MALDI-TOF MS technique

Population growth causes food demand to rise, approximately 36 million people die each year due to starvation or related diseases. One solution to this problem is the continuous examination and development of the agricultural economy. In this study, matrix-assisted laser desorption/ionization time-of-flight mass spectrometer (MALDI -TOF MS) were used to analyse of sunflower, soybean and hemp. In order to analyse the protein of maize, this method has already been applied. However, for sunflower, soy and hemp, it is necessary to develop a sample preparation method. Choosing the optimal matrix solution for ionization the traget molecule is an essential part of developing the method. Our aim is to compare two different matrix solutions (α-HCCA, SA matrix), based on the properties (intensity, noise ratio, value of spectra) of the spectra.

Analyses of a few physiological parameters of hybrid wheat in the case of different nitrogen supply levels
49-53Views:498The winter wheat is one of the most determinant crops because its role was always important in human’s life. To increase the average yield there are several possibilities, which are still not clear fields of agricultural plant production. Our main goal was to examine the responses of winter wheat genotypes to different amounts of nitrogen supplies. The sowing area of hybrid wheats are increasing, they may have different nutrient nitrogen utilization compared to varieties, and the question arose if it is possible to achieve same yield at lower nitrogen fertilizer application or not.
The present study analyzes the results of winter wheat (Triticum aestivum L.) from tillering growing stage. Under controlled conditions three different wheat hybrids were grown (Hywin, Hystar, Hybiza) with two different amounts of nitrogen supplies (optimal and the fourth part). The dry matter accumulation, relative chlorophyll content and nitrogen content were measured in order to draw conclusions from the different supplies of nitrogen for winter wheat genotypes and their physiological plasticity.

If the increase in stock size, specific yields, genetic improvement and modernisation are not realised, several thousand shepherds will lose their jobs and possibilities to work, and their families will lose their only source of livelihood.
Several hundred hectares of grassland will become weedy and the costs of crop protection will increase in these regions. Pollen pollution, the number of allergic ailments and medical costs will increase, mentioning only the main disadvantages. Its financial requirement is nearly equal to the one required for development without any essential compensation or production.
In order to develop a competitive branch, a subsidisation of 10-20 billion HUF is needed until the accession of Hungary to the EU, depending on the scale and progressivity of development. The subsidisation and realisation of the strategic steps would result in:
– the establishment of a registered livestock with 1.5 million ewes, which would ensure better market position for the branch in the EU (5%),
– the establishment of a competitive and marketable branch with a production value of 15-25 billion HUF, calculated using the present price level (after accession, the branch would be self-operating and profitable, with the 6-8 billion HUF/year normative subsidisation by the EU),
– the establishment of a genetic potential which would ensure our participation in profitable international breeding animal market, which provides extra profit,
– the establishment of a sheep husbandry with better genetic potential and phenotypic performance, producing a quality of international level,
– the maintenance of the workplace of more than 10.000 people (saving 3-5 billion HUF/year of the budget),
– the maintenance of the condition of growing areas of grasslands with their advantages (crop protection and public health) whose financial effect is considerable. -
Comparative Evaluation of the Temperaments of Charolais and Hungarian Grey Steers
14-19Views:277Animal breeding increasingly lays claim to the theoretical and practical knowledge of applied ethology. The authors’ aim was to evaluate and compare the temperaments of Charolais (CH, n= 10) and Hungarian Grey (HG, n= 10) steers, and also to determine the correlation between their temperament scores and flight speed scores. Temperament was evaluated by the results of the scale test (assessing of behaviour in a 1-5 score system, while the animal is standing on a scale for 30 seconds) and flight speed test (minutes it takes the animal to move a set distance of 1.7 m when leaving the scale), on three occasions (1, 2, 3). Data management was done by SPSS.10 (ANOVA, Mann-Whitney-test, Spearman-correlation). Results of the scale test differed significantly between breeds at the third measurement (CH: 2,9 scores; HG: 1,4 scores; P<0,01) and when evaluating the three measurements together (CH: 2,0 scores; HG: 1,37 scores; P<0,05). Concerning of flight speed score, there were significant differences between breeds of steers at each measurement (1. measurement CH: 2,77 s; HG: 4,09 s; P<0,05; 2. measurement CH: 2,89 s; HG: 5,01 s; P<0,01; 3. measurement CH: 2,46 s; HG: 5,33 s; P<0,01) and overall (CH: 2,71 s; HG: 4,81 s; P<0,001). In the case of both breeds, evaluated by measurements and overall, a negative correlation was calculated between temperament score and flight speed score, but this was significant only in three cases: CH1 (n=10) r= -0,75; P<0,01; CH1+2+3 (n=30) r= -0,44; P<0,05; CH+HG1+2+3 (n=60) r= -0,33; P<0,01). Results indicate that Hungarian Grey steers are calmer than individuals of Charolais. Animals behaving calmer on the scale left the scale, more slowly. The authors propose the use of these temperament tests in Hungarian breeding practice, in order to select too temperament animals.

Studying the reproduction-biological effect of the GPR54 receptor gene in sheep

Today, genes, mutations that may develop aseasonality in sheep are extensively researched in order to make the establishment of populations capable of aseasonal estrus possible.
According to the current knowledge, the GPR54 gene participates in the GnRH release from the hypothalamus, and thus in the alteration of the reproduction state of the organism. Our research is aimed to determine whether the GPR54 gene has allele variations that influence the proneness to aseasonal estrus in sheep in a different extent. Therefore, the GPR54 gene sequence of sheep is first examined. -

That optimum point can be reached, if we achieve maximum possibility of recovery of material risks. There are more than one procedures exist for researching of optimum point or risk assessment in other word, and they are needed to use even parallel. In order to take into consideration effects of risk we need to measure them somehow. At risk assessments risks are usually some kind of result of impact and possibility. Knowledge and measurement of impacts and possibilities are far not enough for assessment of certain risks. In this study I will present a possible usable risk assessment methodology for financial audit activities with using Multi Attribute Utility Theory (MAUT in the followings).

Agricultural land use and food safety
55-60Views:545The world’s food production needs to be doubled in order to cover the need of the population by 2050 even if it exceeds 9 billion. The output of agriculture is expected to increase by 1.7% every year until 2020 (OECD, FAO 2011) which is a major decrease in comparison with the average 2.6% increase of the previous decade. At the same time, the meat, dairy, sugar and vegetable oil consumption is likely to increase by 2020 to a higher extent than so far. Due to the increasing food prices, the amount of starving people will increase and food consumption will decrease – especially in developing countries – as people will be able to buy less and only cheaper food products. Also, obesity may become a more severe problem and the inequality of the population’s health status could increase.
One of the most important elements of adapting to global climate change is food safety; therefore, it is especially important to breed new biological bases and to introduce production systems which contribute to adapting to changed circumstances.

Testing laboratory parameters of compost tea
31-36Views:989During the industrial production of broiler chicken, a large amount of manure is produced, of which easily contained nitrogen content (without pre-treatment) is released into the atmosphere as an air pollutant. In our experiments, we aimed to prepare compost tea, also known as water extract of compost, from pre-treated poultry manure in order to create a product can be utilized as liquid nutrient supply. The poultry manure source was the Baromfi-Coop Ltd. located in Nyírjákó, Hungary, where it was treated by composting. As a result of this pre-treatment of the poultry manure, its nutrient parameters improve and nitrogen is present in a form that is better utilized for plants. Furthermore, this product is suitable for further utilization and also can be the base material for a brand-new product. For this reason the effects of compost/water ratio, incubation time, low oxygen level, and extraction time on the parameters of the resulted product were studied in the frame of developing new soil-life enhancing microbial product, so-called compost tea.
Chemical parameters of the compost used as base material strongly determined the properties of the resulted compost tea, especially the ratio of the various nitrogen forms, their concentration and the salt content. It was found that adding water at a higher rate that means 1/40 and 1/50 mixing ratios results in more cost-effective production. In the experiment the compost tea were held under oxygen-poor conditions, therefore pH of the extractions decreased, which influenced the quality and quantity of their nutrient content.
