Inland and foreign Erwinia amylovora isolates by carbohydrate utilization
29-33Views:142Fire blight, a plant disease caused by the bacterium Erwinia amylovora, produces serious losses in apple and pear orchards all over the world. Since the appearance of fire blight in Hungary (Hevesi, 1996) Erwinia amylovora isolates were collected in different years, from different hosts and areas in order to establish gene bank for future epidemiological studies. We had isolates from foreign countries as well. The aim of our research was to compare all of the Hungarian and foreign isolates by carbohydrate utilization.
In our experiments effect of carbohydrates on E. amylovora multiplication was determined using API 50 CH strip (bioMérieux, France). By the API 50 CH strip method we checked a number of unstudied carbohydrates. The results of the tests shows colour changes. Based on utilization of 49 carbohydrates of API 50 CH kit by E. amylovora isolates, two groups of carbohydrates can be defined: “Utilized” - and “Not utilized” carbohydrates. All isolates utilized 20 different carbohydrates after 164 hour incubation. Conversely, isolates also could be divided into four groups (1, 2, 3, 4) by arbutin and raffinose utilization. In group 1.-isolates utilize arbutin; 2.- utilize raffinose; 3.- utilize both arbutin and raffinose; 4. -utilize neither arbutin nor raffinose. Presumably carbohydrate content of nectar could play an important role on invasion of the (E. amylovora) bacterium via flower.
It could be concluded that the carbohydrate utilization – completed with genetic analysis – can be used for characterization of Erwinia amylovora isolates. -
The Fattening and Slaughtering Examination at the Hungarian Racka Sheep
37-42Views:211Fattening and slaughtering characteristics of both types (black and white) of the Hungarian racka sheep were investigated at the site of the National Institute for Agricultural Quality Control in Atkár. The Hungarian merino, the dairy cigaja and the British milksheep were applied as control groups. 10 animals were examined from both sexes in each genotype. The examination was carried out according to the principles set in the Codex for sheep breeding. During the test acute diarrhoea occurred in each control group. However, only few lambs were excluded from the test from the British milksheep and the Hungarian merino groups because of significant changes in their condition. Consequently, the results obtained were not significantly influenced. No symptoms of any illness could be recognised in the Hungarian racka sheep, though. The significance of the difference between the average of the examined groups were checked by T-tests.
Daily gain was significantly less compared to the control groups, except for the white racka males. The lambs starving for 24 hours before slaughtering weighed 25.3-30.0 kgs on average. The black and white racka females weighed the least while the Hungarian merino females weighed the most. The quantity of abdominal and kidney fat was significant in the case of our native females. The quantity of abdominal fat was significantly bigger compared to each control group. On comparing the investigated groups it turned out that the white racka males had the biggest skin weight while the white Hungarian racka females had the smallest, that is the two extremes were recognised in the same colour (white) of one bred. On qualifying the slaughtered animals females had better results than males in each bred; the racka sheep were better than the dairy cigaja but were worse than the Hungarian merino and British sheep groups.
The huge differences in the carcasses of the groups were seen clearly when chopping. Examining the leg, it was noticed that the native groups proved to be smaller than the control groups, except for the black racka males. As for the short loin, the Hungarian racka and the Hungarian merino sheep proved to be the best, whereas the dairy cigaja and the British milk males had much smaller ones. The results gained when investigating the rack were similar to those of the short loin. As far as the rib and shoulder are concerned, the black racka females and the dairy cigaja females had the worst results. After chopping, the right side was also boned in every group. The small proportion of bone in the case of racka sheep was easily noticeable that resulted in the fact that the white racka females had the highest quantity of short loin compared to the other groups. When examining the back quarter, the bone proportions of the 4 racka groups proved to be significantly less than those of the control groups but it was not true for the meat proportions. As for the front quarter meat, the racka males were prominent in the great shoulder meat and in the small proportion of bone. The quantity of the first quarter bone was the least in the case of racka sheep and was the biggest for dairy cigajas. This difference proved to be significant in most cases.
In short, the Hungarian racka groups had the best results in meat quantity, which was especially significant compared to the dairy cigaja sheep. However, the quantity of external fat increased. -
The Effect of Mg Foliage Fertilization on Beetroot (Beta vulgaris ssp. esculenta var. rubra) Morphology, Quality Traits and their Relations

The applied foliage fertilization affected inner colour intensity and uniformity in roots positively.
The 2% Mg-sulphate solution affected pigment quantity favourably depending on varieties.
Close correlation (r=0,71) was found between the yellow (BX) and red (BC) pigments which indicates a similar biosynthetic pathway of the two compounds.
The Mg-sulphate foliage fertilization improves quality considerably but the individual reaction of varieties needs further studies. -
Lack of polymorphism of the agouti signaling protein (ASIP) gene among four different brown hare (Lepus europaeus Pallas 1778) populations
81-85Views:311The brown hare (Lepus europaeus Pallas 1778) is a common palearctic and a popular game species therefore it has been an obvious subject for population genetic studies since the second part of the 20th century. Among the several mitochondrial DNA studies some have been carried out concerning nuclear genes as well. The agouti signaling protein gene (ASIP) is involved in regulating the synthesis of eumelanin and pheomelanin in melanocytes of mammals. Though many studies focused on it in relation with several mammalian species, minimal information is available on this topic concerning the brown hare.
Here we present a short communication concerning the agouti signaling protein (ASIP) gene in four different country’s L. europaeus populations, namely Lithuania, Hungary, Serbia and Georgia. N=45 tissue samples have been investigated from overall 17 sampling sites of the different countries. There has not been found any polymorphism among the sequences. In an alignment with other Leporid species’ partial ASIP sequences downloaded from ENA we have found that based on a 178 base pairs long DNA sequence the haplotype of our samples contains three other Lepus species as well. This is concordant with the findings of a previous study focusing predominantly on the European rabbit (Orycto lagus cuniculus Linnaeus 1758) and the several mutations of its ASIP gene.

The methodology of the Hooded Crow’s (Corvus cornix L.) colour ringing and the previous results

It is breeding in Debrecen since 1959, and nowdays it’s presence can be detected at all points of the city, it is an permanent breeding species of the bird fauna. Our knowledge about the nature of urban races, including motion patterns, area fidelity is sufficiently incomplete. The aim of our research is to answer these questions by using our colour-ring program. In this study we present in details the methodology of Hooded Crow’s colour-ring program in urban area, and we also report our previous

A simple method for preparing elemental selenium nano- coating inside a silicone surface
35-43Views:503Selenium nanoparticles (SeNPs) with a bright red colour have aroused worldwide attention due to their unique properties in selenium supplementation because of their low toxicity and favourable bioavailability. A simple method was developed for making a red selenium nanolayer on the inner surface of Polyvinyl chloride (PVC) and silicone tube. The selenium nanoparticles were produced by the reaction of sodium selenite and ascorbic acid. Red amorphous selenium nanoparticles have been successfully synthesized by the reaction of 500 mg dm-3 Se (sodium selenite) solution with 10 g dm-3 ascorbic acid solution at room temperature, and morphology was confirmed by X-ray diffraction analysis (XRD). The coating density was compared on PVC and silicone surfaces by using Scanning Electron Microscopy (SEM) and Energy-dispersive X-ray (EDS) analysis. The nanolayer with about 16 µm thickness on the silicone surface significantly evenly distributed compared to the PVC surface. The selenium coated silicone tube could be a good source of selenium for a continuous, low-level selenium supplementation of farm animals via drinking water.

N-Phenyl-Phthalamic Acid and Fertilization Effects on Flowering, Fruit Set and Fruit Quality of Apple (Malus domestica Borkh.)
24-28Views:275On seven apple cultivars (Gala Must, Gloster, Granny Smith, Idared, Jonagold, Jonathan Csány 1, Mutsu), the authors studied the influence of N-phenyl-phthalamic (PPA) acid and fertilization on flowering, fruit set and fruit quality in the years 2003-2004. The research results showed that PPA application extended the flowering time of the most cultivars. Fruit set of apple cultivars increased in many cases when regulator was applied. The additional nutrient supply could increase the fruit set too. The increase of fruit set increased the number of fruit per tree. Quality parameters determined by fertilization and fruit number per tree. For example, when N-phenyl-phthalamic acid was applied without fertilization the fruit number per tree increased considerably however, it caused a considerable frittering away. The reason for this is that fruit set was high and fruit received not enough nutrition for growing up. The skin colour of fruits was decreased in several cases by the treatments. In one respect the enlarged fruit number was shaded the others and the fertilization increased the vegetative shading leaf area.

Effect of M9, MM106 and Seedling Rootstocks on Sunburn-Sensitivity of 33 Apple Cultivars, and Sunburn Effects on Fruit Quality
236-247Views:218The purpose of this study was to investigate the sunburn-sensitivity of 33 apple cultivars grafted onto 3 rootstocks (M9, MM106 and seedling) in a commercial orchard at Nagykutas (in the western part of Hungary). The authors also searched for any relationship between fruit quality parameters and the frequency of sunburn on the fruit surface.
During the observations, the cultivars had rootstock-specific properties in respect to sunburn-susceptibility. Accordingly, the injury decreased in the order M9, MM106 and seedling rootstocks. The differences in the sensitivity were founded on the foliage-morphological characteristics of trees, caused by the growing vigour of the rootstocks. Accordingly, the highest value of sunburn injury was observed on M9 rootstock, because this rootstock has a dwarfing effect on the grafted main cultivars. Thus, the vegetative area of these trees grew very slowly and the foliage was not compact enough to protect the fruits from the strong rays. The largeness and density of the foliage increased in the order M9, MM106 and seedling rootstocks. Relationships were also demonstrated between the diameter of the upper part of crown, the size of leaves, the number of fruits per tree and the injury from sunburn.
The damage values showed, that the gravity of symptoms did not decrease below a well-defined level of dimension on fruit. Determined potential area of injury was necessary for the symptoms to become visible.
The authors categorized the cultivars in the respect of values of sunburn frequency: I. „Not sensitive”, II. „Moderately sensitive” and III. „Very sensitive” categories were constituted. Generally, the Gala cultivars showed low damage (or were free of symptoms), in contrast, Golden mutants suffered relative strongly. The most sensitive cultivar was Jonica on all three of rootstocks.
We searched for any relationship between the fruit quality parameters and the frequency of sunburn. A significant correlation was found in the cases of fruit weight and colour-coverage. The latter can be related to that fact that highly colour covered fruits are found on the peripherial part of crown, so these are exposed to stronger sun rays -
Mitochondrial DNA-based diversity study of Hungarian brown hares (Lepus europaeus Pallas 1778)
23-29Views:302The brown hare being an important game species which is widespread across the European continent has been in focus of many population genetic studies. However only a few comprising researches can be found on the diversity of Central-European populations.
The aim of our large scale long term ongoing study is to fill this gap of information on the species by describing the genetic history and structure of the brown hare populations of the area using both mitochondrial DNA markers and genomic skin and hair colour regulating genes.
This article gives forth a part of our results concerning the mitochondrial DNA diversity of Hungarian brown hares based on amplification of a 512 bp long D-loop sequence. N=39 tissue or hair samples have been collected from 15 sampling sites on the Hungarian Great Plain. We have described a high level of haplotype diversity (Hd=0.879±0.044) based on a 410 bp alignment of our sequences. We have found 17 haplotypes within our sample set with the nucleotid diversity of π=0.01167±0.0022. Our ongoing research shows high genetic diversity for the brown hare in the studied region and a second alignment with 156 sequences downloaded from GenBank indicates a geographic pattern of haplotypes among the studied populations though these results need confirmation by our further analyses.

The Effect of Year and Irrigation on the Yield Quantity and Quality of the Potato
12-16Views:324In Hungary, the growing area of potato area reduced dramatically in the last few decades, additionally we are lagging behind the Western European countries as regards yields and the competitiveness of production is further decreased by the great alternation in yields from year to year, the unpredictable market conditions, bad consumption habits and many times unfortunately the lack of quality products.
The ecological and climatic conditions of Hungary are not everywhere suitable for potato, in the area of Debrecen the amount of rainfall was lower, and the monthly average temperature was higher than the requirement of potato in its growing season in 2002 and 2003.
The experiment was carried out at the experimental site of the University of Debrecen, Farm and Regional Research Institute, at Látókép. In our experiment we examined the yield and some quality parameters of 8 and 9 medium-early varieties in large parcels in 2002 and 2003 respectively. Out of the examined varieties 3 are of Dutch, and 6 are of Hungarian breeding.
The experiment was set up on 49.5 m2 parcels on calcareous chernozem soil after winter wheat as a forecrop in both years. The 9 varieties were examined in 4 repetitions in randomized blocks, out of which two repetitions were irrigated, and two were non-irrigated.
We examined the yields of the varieties, the distribution of tubers according to size and their percentages, and the changes in specific parameters of quality and inner content due to irrigation. We studied the dry matter content, the starch content, the under-water mass, the amount of reducing sugars, the colour index of frying and the element contents of tubers.
Summing up, it can be stated that among the agrotechnical year effect, variety and irrigation factors have considerable impact on potato yield quality and quantity. On the basis of our results, it can be stated that in potato production variety should be chosen in accordance with the aim of production and technology should be adapted to that specific variety. -

Investigations on Mud on Heavy Metal Contaminated Flood-Plain of Tisza
96-101Views:671At the beginning of the year 2000 subsequently to a mine accident high heavy metal content mud entered the catchment area of the Tisza and was transported through the whole Hungarian section of the river. The majority of the heavy metals had been bounded to the floating sediment that was deposited on the flood-plain soil during flood forming a new, 5-10 cm thick layer. In the mud samples collected after the flood there was a clearly visible dark grey layer with significantly higher heavy metal content that was formed by the pollution wave and it was sorruonded by a light layer. The upper layer of flood-plain soils are formed from this mud layer during the soil development process, so the amount of Lakanen-Erviö soluble heavy metals that correlate with bioavailable heavy metal content was examined as well. In this case only the lead content was significantly higher in the dark layer.
New mud samples were collected after the 2001 flood. Separate layers could not have been identified, their colour was similar to those of the previous year’s light layers’. Comparing to this light layer the total Zn and Lakanen-Erivö soluble metal content was significantly lower in the mud samples of the year 2001. While the proportions of total and Lakanen-Erviö soluble metal concentrations were equal in both of the layers regarding the elements, these ratios have significantly changed next year regarding Pb and Zn: the amount of Lakanen-Erviö soluble metals considerably decreased.
As a result of sequential extraction the heavy metal content was rather low in the water soluble and exchangeable and NaOH-soluble fractions, so heavy metals found in the mud could be released in greater amount only in case of a heavy acidification.
