  • Coincidences between molecular genetic and studbook data of gidrán mare families on the basis of mtDnA

    The traditional Hungarian horse breed, Gidran has been close to the edge of extinction several times. Despite the multiple bottleneck effect, the breed has retained a part of its genetic variability, and performed prominently in carriage driving and show-jumping competitions. Maintaining of the Gidran breed is important in the point of view of world heritage; because besides Hungary, smaller Gidran populations exist only in Bulgaria and Romania. Taking advantage of the special inheritance features of mtDNA, our study focused on two mtDNA regions of Gidran mares. Altogether, 251 hair samples from various Hungarian studs were examined. The analysis was successfully made in case of 251 samples of the cytochrome b and in case of 246 samples of D-loop regions. Because of the distinct mutation rates of the two mtDNA markers, the number of the haplotypes and the way of grouping samples into haplotypes was different. Our key finding was that most haplotypes may be compatible with mare families of the stud book; however incidental mistakes in stud book have occurred only in a few cases. Our results indicate the importance of the preservation and breeding those mare families, which are molecular genetically more diverse than the others, and are in the edge of extinction.

  • Brief overview of the polymorphism analysis of genes affecting pig prolificacy (LEP, PRLP, ESR BF, EGF, FSH-β, H2A.Z)

    Researches are being performed around the world to increase swine prolificacy by using marker-assisted selection (MAS). The present study processes researches of polymorphism examinations on 7 genes. The result of the experiments showed that the leptin gene (LEP) prolactin receptor gene (PRLP), estrogen receptor gene (ESR), properdin B (BF) epidermal growth factor (EGF), follicle-stimulating beta gene (FSH-ß) and Z member of the H2A histon family gene (H2A.Z) and their alleles have a positive effect on reproductive characteristics of different swine breeds. In addition to this, leptin gene (LEP) influences the build, meat production and growth of body fat. Further studies are concerned with the polymorphism of an increasing number of genes, which enables a faster genetic development of swine breeding.

  • Upgrading breeding value estimation in beef cattle

    This paper gives a summary of the possibility for applying genomic information for breeding value estimation in beef cattle breeding. This process is called genomic prediction and is now widely used in dairy cattle globally as well as in some beef and sheep populations. The advantage of genomic prediction is a more accurate estimate of the genetic merit of an individual at a young age thereby facilitating greater annual genetic gain, predominantly through shorter generation intervals. Genomic predictions are more advantageous for sex-linked (e.g., milk yield), low heritability (e.g., fertility) and difficult-to-measure (e.g., feed intake) traits. The larger the reference population, on average, the more accurate the genomic predictions; additionally, the closer genetically the reference population is to the candidate population, the greater the accuracy of genomic predictions. Research is continuing on strategies to generate accurate genomic predictions using a reference population consisting of multiple breeds (and crossbred). Retrospective analysis of real-life data where genomic predictions have been operation for several years clearly shows a benefit of this technology.

  • Horn colour varieties of the Hungarian Grey cattle

    The Hungarian Grey Cattle breed is a ‘success-story’ of the national genetic conservation work. Traits of the breed have been subjects of several research projects, although many relationships remained unclear. Our present research results were meant to call attention to a less emphasized trait of the breed: the different horn colour varieties. Research work was carried out in the Hungarian Grey stock of the Hortobágy Non-profit Company for Nature Conservation and Gene Preservation. Ratio of the three main horn colour varieties (white, ‘cardy’, green) were determined in the observed female, male and steer stocks. Our results showed no significant differences (P<0,05) in the distribution of the horn colour varieties of the female and bull stocks, and of the female and steer stocks. We found that on the basis of the ratio of white colour, four sub-categories can be distinguished within the ‘cardy’ colour variety. Results of the statistical analysis (P<0,05) confirmed that the colour of the horn and the ratio of the black part on the horn tip are two different traits. 

  • An Analysis of Rotational Line Mating Using Computer Simulation

    In a simulation examination, we analyzed the effect of the family size and the rate of pairing on the survival of rare genes, to keep the level of variation of the genepool and to avoid the loss of alleles.
    The population size was 360 animals. In the simulation, we calculated on the basis of a discrete population. We placed the 360 animals into different clusters, with 3 types of frequencies of alleles and 3 types of groups. We assumed 2, 3 or 4 alleles in 8 loci. We generated 15 generations using the same mating and selection system used in practise. The simulation was written with Scilab 2.7.2 software, and evaluated with SPSS software.
    There were significant changes in the effect of family size on the genetic variation in the following cases: when the base population had the same gene frequencies in all loci, and when the gene frequencies were between 0.125-0.75. In these cases, we found that the smaller families (10 animals/cluster) were better than the larger families (30 or 90 animals/cluster). The first generation where there accured a loss of alleles was averagely earliest in larger families (90 animal/cluster). This average was 3.37 generations. When we are searched the effects of the different rates of pairing we found those cases most favourable when the ratio of males and females was 1:2 or 1:4 as compared to 1:9. The first generation where there was a loss of alleles was averagely earliest at the ratio of pairing male and females of 1:9 (the mean was 3.05 generations) when the frequency of the rarest allele was 0.0069.
    The recently introduced rotating-random mating system is an eligible method for small populations for the preservation of genes.

  • Review on the fatty acid profile and free fatty acid of common carp (Cyprinus carpio)

    Carp or ponty in Hungarian, is considered commercial freshwater fish, which is an adaptable species in both wild and cultured conditions. Carp has high nutritional value content, favorable taste, it is rich in protein, and low in saturated fat. The nutritional content in fish is composed of many chemical constituents and influenced by many factors. One of the components that its content may be different due to internal and external factors is fatty acids, which may vary depending on endogenous and exogenous factors. The endogenous or internal factors include the genetic, size, sexual maturity, and life cycle phase. While microclimate, water quality, quality of food or diet habit, and the amount of available food or starvation are considered as exogenous or environmental factors. Freshwater fish has the ability to convert essential fatty acid into long chain polyunsaturated fatty acid like AA, EPA, and DHA. Most results showed that palmitic acid and oleic acid were the dominant SFA and MUFA in carp both for wild and farmed carp in all seasons. The PUFA for wild carp was mainly dominated by DHA, while on farmed carp by LA. It confirmed that high LA content in farmed carp was related to the diet habit. The amount of lipid and FA were changed in line with the season. Even the statistical analysis showed no significant difference, but some studies showed a contrasting result. Moreover, most obtained results acknowledged that FA tends to decrease during the spawning period. The amount and composition of FA were affected by the total lipid content. The lipid must be broken down into simpler compounds such as FA or FFA for the metabolism of fish. The result of metabolism then transported into the utilising tissue and used as energy.

  • Agrobotanical Analysis of Bean (Phaseolus vulgaris L.) Equilibrium Populations Suitable for Regional Land Cultivation

    The local bean landraces maintained at small-scale farming level should have importance as complementary source for completion of plant genetic resources with potential role in local land cultivation. Also, they should be used as primary material in breeding activities targeted on improving the foodstuffs quality. In the research were used especially landraces collected in Nyírség, Körös-mellék and Tápióvidék region. Our main objective was to reveal the relationship between the adaptation ability of landraces and the distance from their original collecting place. For that reason the location of the experimental plots was choosen by taking into account that they must be checked in each region, on it’s original region and similarly in other two different regions. On the base of the recorded agro-morphological traits we can get answers on the shaping of adaptation ability, and in the same time by a general chemical characterisation we should know more about the variation of qualitative parameters with a potential economical role. In the designing of experiment a main importance had the conditions where the research is taking place. All activities related to the maintenance of experiment were done in frame of a low input agricultural system to emphasize as much as possible the natural endowments of regeneration places.

  • Inland and foreign Erwinia amylovora isolates by carbohydrate utilization

    Fire blight, a plant disease caused by the bacterium Erwinia amylovora, produces serious losses in apple and pear orchards all over the world. Since the appearance of fire blight in Hungary (Hevesi, 1996) Erwinia amylovora isolates were collected in different years, from different hosts and areas in order to establish gene bank for future epidemiological studies. We had isolates from foreign countries as well. The aim of our research was to compare all of the Hungarian and foreign isolates by carbohydrate utilization. 
    In our experiments effect of carbohydrates on E. amylovora multiplication was determined using API 50 CH strip (bioMérieux, France). By the API 50 CH strip method we checked a number of unstudied carbohydrates. The results of the tests shows colour changes. Based on utilization of 49 carbohydrates of API 50 CH kit by E. amylovora isolates, two groups of carbohydrates can be defined: “Utilized” - and “Not utilized”  carbohydrates. All isolates utilized 20 different carbohydrates after 164 hour incubation. Conversely, isolates also could be divided into four groups (1, 2, 3, 4) by arbutin and raffinose utilization. In group 1.-isolates utilize arbutin; 2.- utilize raffinose; 3.- utilize both arbutin and raffinose; 4. -utilize neither arbutin nor raffinose. Presumably carbohydrate content of nectar could play an important role on invasion of the (E. amylovora) bacterium via flower.
    It could be concluded that the carbohydrate utilization – completed with genetic analysis – can be used for characterization of Erwinia amylovora isolates.

  • Timing it right: The Measurement and Prediction of Flowering

    Although the model described here was developed from research in controlled environments, there is now considerable evidence that in can be applied to a very wide range of natural environments in several species. Multi-locational trials augmented by successional sowing and, if considered necessary, supplementary illumination in the field to increase daylength, can be used to estimate the values of the model coefficients: (1) to characterize germplasm collections and so predict flowering behaviour elsewhere; (2) for interpreting and understanding crop adaptation; and (3) for genetic analysis of photoperiod sensitivity. We do not yet know whether the model has any contribution to make to the understanding of the biochemical mechanisms of photoperiod and temperature responses, but at the very least, it should provide the basis for indicating the most appropriate environmental conditions, genotypes and physiological stage of the plants most suitable for such investigations.
